The Michael–Mannich cascade cyclization of cyano olefins, ethyl 4,4,4-trifluoro-3-oxobutanoate, aromatic aldehydes and ammonium acetate provides convenient stereoselective formation of ethyl 5,5-dicyano-4,6-diaryl-2-hydroxy-2-(trifluoromethyl)piperidine-3-carboxylates with fourstereogenic centers and dialkyl 4,6-diaryl-5-cyano-2-hydroxy-2-(trifluoromethyl)piperidine-3,5-dicarboxylates with five stereogenic centers. Electrolysis of 3-substituted 2,2-dicyanocyclopropane-1,1-dicarboxylic acid esters in alcohols in an undivided cell in the presence of NaBr or NaOAc afforded 6-substituted (1R,5R, 6R)*-4,4-dialkoxy-5-cyano-2-oxo-3-azabicyclo[3.1.0]hexane-1-carboxylic acid esters in 80–95% yields. The solid-state reactions of crystalline ferrocenyl- and ruthenocenylaldehydes with optically active primary β-hydroxyamines were studied at ∼20 °C. The yield of the products increases substantially in the presence of K2CO3. The tautomeric equilibrium between imines and diastereomeric oxazolidines with the predominant formation of one of them is established in solutions of the products in CDCl3. A new macrocyclic NADH model 1 has been designed and synthesised. The new model consists of the same subunits as previously reported models. However, the present model is designed as such that the pyridine nitrogen of the nicotinamide units are not incorporated in the macrocyclic framework. Thus, properties such as solubility can easily be varied by alkylation with an appropriate agent. The macrocyclic framework was prepared in 7 steps. Methylation of the pyridine nitrogens followed by reduction gave the desired model. This model compound was found to reduce methyl benzoylformate stereoselectively in good yield with 48% enantiomeric excess.  相似文献

The regio- and stereoselectivity of the reactions of carbanions, generated from alkanals, carboxylates of the type XYCHCOOEt (X = EtOOC, CN, Ac; Y = H, Br), or derivatives of 3-methyl-4-phosphono-2-butenoic acid using PTC techniques, with aldehydes of various types (alkanals, ,-enals, cross-conjugated enedials, benzaldehydes,etc.) are reviewed. The factors affecting the outcome of these reactions are discussed. The carbanion analogs, triphenylphosphorus ylides, are shown to attack selectively at one of the aldehyde groups of aromatic dialdehydes. The reaction of diethyl 3-ethoxycarbonyl-2-methyl-2-propenylphosphonate (1) with 3-methylbutanal (2) in the system solid MOH (M=K, Na, Li)-benzene without phase transfer catalysts affords ethyl 3,7-dimethyl-2,4-octadienoate with the ratios of 2E,4E stereoisomer (3a) to 2Z,4E-stereoisomer (3b) of 4456, 4654, and 6040 for KOH, NaOH, and LiOH, respectively. The similar tendency is observed when DMSO is substituted for benzene, although the proportion of3a in this case is 65% with any MOH used. The reaction of1 with2 in the presence of a stoichiometric amount of a quaternary ammonium hydroxide QOH (Q=NMe4, BnNMe3, (n-Bu)4N) in benzene gives higher3a3b ratios than those obtained with LiOH in the former reaction system. The increase in the content of 2E,4E-isomer3a is possibly due to the growth of the population of solvent-separated ion pairs or, generally, to the increase in the distance between the charge centers in the ionic intermediates on passing from KOH to LiOH and from [Me4N]OH to [(n-Bu)4N]OH. The position of the equilibrium betweenE- and Z-isomers of phosphonate1 under the reaction conditions (E-1Z-13565) is almost unaffected by the nature of the cation of the base.Part 5, see ref. 1.Translated fromIzvestiya Akademii Nauk.  The enzyme catalyzed conversion of R/S-(±)-2-octanol with hexanoic acid to R/S-(±)-2-octyl hexanoate has been studied in different microenvironments and in the presence of the competing substrate ethanol. The reactions were performed in various gels made from aqueous gelatin solutions and liposome dispersions or isotropic liquid solutions, with or without oil and ethanol. The lipase Candida sp. (SP 525) was dissolved in the dispersions or solutions stabilized by the naturally occurring zwitterionic surfactant soybean lecithin. The sectioned porous gel was immersed in hexane containing 0.33 mol dm-3 of racemic 2-octanol and hexanoic acid. Since ethanol acts both as a substrate and as a part of the gel it is of fundamental interest to know the phase behaviour of the used systems. Partial phase diagrams for the systems ethanol–water–soybean lecithin and ethanol/water (7:3)–oil–soybean lecithin were determined at 298.2 K. The oil was either castor oil or hexadecane. The conversion of R-2-octyl hexanoate was about 0.45 when no or small amounts of ethanol was present, but decreased considerably with high amounts of ethanol present and ethyl hexanoate became the main product. Hydrolysis of R-2-octyl hexanoate was favoured in the latter systems and hexanoic acid formed was immediately esterified to ethyl hexanoate. The conversion of R-2-octyl hexanoate and ethyl hexanoate depends only on the ethanol content present in the systems and is thus independent of the environment of the enzyme. However, the chiral esters synthesized from racemic 2-octanol and hexanoic acid showed high optical purities regardless of the ethanol content. Received: 1 July 1996 Accepted: 30 August 1996  相似文献

The mechanism of formation of alkene stereoisomers in the catalytic olefination reaction of carbonyl compounds was studied. 4-Chlorobenzaldehyde hydrazone 1 stereoselectively reacts with a number of F-, Cl-, Br-, and I-containing polyhaloalkanes in the presence of catalytic amounts of CuCl to give -substituted styrenes 2 with the more thermodynamically stable alkene isomer being the major product. A model for the formation of the stereoisomers of alkenes 2 in the olefination reaction is proposed. Stereoselectivity of the reaction is determined by elimination of copper(ii) halides from the lowest-lying conformers of organocopper intermediates II. According to quantum-chemical calculations, the elimination should involve the staggered conformations with antiperiplanar arrangement of C—Hal and C—Cu bonds and proceed by the E2 anti-elimination mechanism. The results of quantum-chemical calculations are in good agreement with the experimental E/Z alkene isomer ratios.  相似文献
