利用激光拉曼光谱技术研究了新型光敏剂血卟啉单甲醚对DNA(脱氧核糖核酸)分子的光动力学损伤。结果表明:DNA分子由典型B型构型变成了一种修饰过的B型。连接碱基对的氢键被部分打断,碱基堆叠也有所与解。碱基本身也遭到了破坏,有脱氨基现象发生。腺嘌呤和胞嘧啶对血咔啉单甲醚的光动力学损伤更敏感。  相似文献

By the use of the advanced ESR technique and through comparing with BHPD, the characteristic of YHPD photosensitization is discussed in this paper in the respect of the primary process of photosensitization. The experiment results showed: (ⅰ) not only ~1O_2, but also free radicals(O_2· OH and YHPD)can be formed by the aid of YHPD; and (ⅱ) as to the ability of producing ~1O_2, YHPDBHPD. Two points are indicated: first, the photosensitized damage of YHPD is interrelated to not only ~1O_2, but also free radicals (O_2. OH and YHPD); second, although the photosensitized damage of YHPD is stronger than that of BHPD, yet the photosensitized damage is negatively correlated to the yield of ~1O_2 but positively correlated to those of O_2 and OH. Based on these two points, it is suggested that activated oxygen free radicals(O_2 and. OH) instead of ~1O_2 play the main role as instantaneously activated material in the photosensitized damage of YHPD.  相似文献

Reduction of dioxygen at a carbon paste electrode (CPE) modified with cobalt hematoporphyrin complex immobilized on silica gel (CoHP–Si) was studied by cyclic voltammetry and rotating disk electrode. In 0.5molL–1 KCl solution, the supported complex showed significant catalytic activity towards four-electron reduction of O2 to H2O at a relatively high pH (5.4). The reduction of dioxygen proceeds at a more negative potential (–0.375V) than the redox reaction CoIII/CoII (0.464V), indicating that the mechanism does not involve the complex as an electron transfer mediator. Catalytic activity of CoHP–Si towards reduction of H2O2 was tested, but no activity was observed.Received October 26, 2002; accepted March 10, 2003 Published online July 16, 2003  相似文献

Abstract

The results of spectral studies of iron(II) protopor-phyrin IX and iron(II) hematoporphyrin IX with several substituted pyridines are reported. The existence in solution of an iron(II) porphyrin complex coordinated to a water molecule and to a substituted pyridine was shown by isolation of the complex from solution. The complex isolated was dimeric inono-4,4′-dipyridyl diaquo iron(II) hematoporphyrin. Addition of ethanol to the aqueous solvent inhibits coordination of iron(II) porphyrins with substituted pyridines. The protoporphyrin ring enhances coordination relative to the hematoporphyrin ring.  相似文献

以氯化血红素(Ⅰ)为原料,经过溴化氢-冰醋酸加成反应、羟基亲核取代反应和无水氯化氢催化酯化反应制得3,8-双-(1-羟基乙基)次卟啉二甲酯(Ⅲ),然后通过琼斯试剂氧化反应制备了3,8-双乙酰基次卟啉二甲酯(Ⅳ)。 考察了血红素与溴化氢-冰醋酸饱和溶液反应过程中温度和时间对3,8-双-(1-羟基乙基)次卟啉二甲酯(Ⅲ)产率的影响;改进了酯化反应的实验条件;选用了廉价易得、选择性较好的羟基选择性氧化剂。 实验结果表明,当反应温度为35 ℃、反应时间为25 h时,血卟啉(Ⅱ)的产率最高,为98.5%;当催化剂为无水氯化氢时产物(Ⅲ)的产率最高,为72.1%;使用琼斯试剂做氧化剂使实验成本大大降低。 通过1H NMR、MS和IR测试技术对产物结构进行了表征。  相似文献

利用1,3-二苯基异苯并呋喃(DPBF)作为单线态氧捕获剂,研究了超声激活血卟啉的机理,通过用紫外-可见分光光度计测量了超声辐照配制好的血卟啉和DPBF溶液后的DPBF相对消耗量,表明了单线态氧的存在量.实验结果显示了DPBF的消耗量随超声照射时间和强度的增加而增加.从而支持了超声激活血卟啉产生单线态氧的说法.并提出了采用各向同性集光球把声致发光和声场分开的方法,进一步研究了是超声空化产生的荧光,还是高温热点激活血卟啉产生单线态氧.与白光对比结果表明,声致发光虽能使照相底版感光,但不足以激活血卟啉产生单线态氧.  相似文献

The photodynamic effect of hypericin on EMT6 mouse mammary carcinoma cells was investigated in vitro under aerobic and hypoxic conditions. Under aerobic conditions, hypericin-induced photocytotoxicity was dose dependent within a 1-50 microM range. Under hypoxic conditions, cells were resistant to hypericin-induced phototoxicity. In the dark, no cytotoxicity was observed at any hypericin concentration tested either aerobically or hypoxically. Cellular accumulation of hypericin, examined by chemical extraction and spectroscopy, occurred under both hypoxic and aerobic conditions. Fluorescence photomicrographs of cells exposed to hypericin corroborate drug uptake in the plasma membrane and subcellular regions. Our results demonstrate that hypericin cytotoxicity to EMT6 mouse mammary carcinoma cells in vitro is both light and oxygen-dependent. These results suggest that EMT6 cell kill caused by photoactivated hypericin is mediated by an oxygen-dependent mechanism, rather than by a type I oxygen-independent mechanism.  相似文献

The hematoporphyrin derivative YHPD, a China-made product, has been clinically used in photodynamic therapy of tumors as a good photosensitizing drug. The NMR study on the structure of its major components is reported here. In terms of high performance liquid chromatography (HPLC) four major components A, B, C and D were isolated. The NMR results showed that the component A is O-acetylhematoporphyrin, B and C are two isomers of vinyldeuteroporphyrin. The spectra of 2-dimensional homonuclear correlation NMR, 2-dimensional NOE (nuclear overhauser enhancement), ~(13)C-NMR and off-resonance as well as FAB (fast atom bombarding) mass spectrum of component D indicate that it is a protoporphyrin dimer linked by carbon-carbon bond. This finding may providea chemical basis for understanding the difference in biological activity between YHPD and other foreign commercial HPD, as well as the composition of clincally used alkali-treated HPD and its effective component.  相似文献
