白藜芦醇的电化学行为及其与DNA的相互作用

采用电化学方法研究了白藜芦醇在pH=2~13的缓冲溶液中的电化学行为、抗氧化能力及其与DNA的相互作用. 研究结果表明， 在2.011的溶液中, 白藜芦醇产生的P4和P5波分别是其二价和三价阴离子的还原波. 在最佳实验条件下, 微分脉冲极谱图上IP3在8.0×10-8~2.0×10-6 mol/L范围内与白藜芦醇的浓度呈线性关系, 检出限为4×10-8 mol/L. 将该法用于中药虎杖中白藜芦醇含量的测定, 结果与高效液相色谱法一致.     

Oxazoline chemistry. Part 12: A metal-mediated synthesis of DMU-212; X-ray diffraction studies of an important anti-cancer agent

An improved synthesis of the anti-cancer agent DMU-212 (trans-3,4,5,4′-tetramethoxystilbene) is described. The methodology involves the use of a Pd-oxazoline catalyst as a mediator of a regio-selective (Heck) C-C bond formation reaction. A simple isolation step is then used to obtain the title material. The compound has been further characterised in the solid-state by X-ray diffraction methods.     

Determination by LC of Polyphenols in Italian Red Wine

A reversed-phase liquid chromatographic method, optimised for the separation of trans-, and cis-resveratrol, catechin, epicatechin, quercetin and rutin, is reported. Separation was achieved using a C18 column and a gradient elution with acetonitrile and 5% formic acid aqueous solution. The analyses required an equilibration period of 10 min and a run time of 25 min for completion. Identification was based on retention characteristics and by relative UV spectra, obtained by photodiode array detector and were compared with commercial standards. Analyses were performed without any sample pre-treatment. Detection was carried out by UV–Vis detector at three different wavelengths. The detection limit ranged from 0.16 μgm L⁻¹ (cis-resveratrol) to 1.5 μgm L⁻¹ (+)-catechin. Investigation was extended to quantitative determination of phenol compounds in Italian red wine and to investigate the stability of the six antioxidants.     

FeCl3·6H2O oxidation of protected resveratrol for the synthesis of tetraarylfuran-type oligostilbenes

Several 8-8-coupled tetraarylfuran-type oligostilbenes were prepared through FeCl₃·6H₂O-catalyzed regioselective oxidative couplings of protected resveratrol precursors. The global deprotection of the coupling dimers in a one-pot reaction yielded (±)-pallidol instead of the targets (±)-restrytisol B and (±)-tricuspidatol A.     

纳米白藜芦醇脂质体的物理化学稳定性研究

纳米载药脂质体的物理化学稳定性,是其实际应用中的关键问题。文章采用薄膜旋转蒸发法-超声法制备了白藜芦醇纳米脂质体（RES-Lip）,采用透射电子显微镜观察其微观形貌,并考察RES-Lip的物理化学稳定性。通过电导法测定了RES-Lip的相变温度（Tm）,及其在凝聚过程的凝聚速率常数（Kco）和表观活化能（Eco）;采用动态透析-紫外分光光度法,研究了温度和pH对RES-Lip降解的影响。结果表明,RES-Lip为球形囊泡结构,粒径小于100 nm;RES-Lip的相变温度为64℃;凝聚速率常数Kco随体系温度升高而升高;表观活化能Eco为86.056 kJ/mol;RES-Lip的降解符合一级动力学模型,降解的表观活化能Ea为59.3157 kJ/mol,降解过程是吸热、自发、熵增过程。本实验制备得到的RES-Lip在4℃、pH=7.40的条件下储存,稳定性较好。     

Inhibition of resveratrol glucosides (REs) on advanced glycation endproducts (AGEs) formation: inhibitory mechanism and structure-activity relationship

Abstract

The study on inhibitory effects of resveratrol glucosides (REs) on advanced glycation endproducts (AGEs) formation is still unmet. Herein, for the first time, the antiglycation activities of five REs in the fetal bovine serum proteins (FBS)/fructose system were evaluated, and its structure-activity relationship and antiglycation mechanism were further explored. These REs showed remarkable inhibition toward AGEs formation. Among them, Piceatannol-3'-O-glucoside (PG) exhibited highest antiglycation activity as reflected in approximately 80% inhibition of fluorescent AGEs at the concentration of 1.0?mM. The structure-activity relationship analysis indicated that glucoside attached to the B ring of resveratrol displays a superior antiglycation activity. Moreover, the results of antiglycation mechanism showed that the antiglycation activity of REs was proportional to their antioxidant capacity and methylglyoxal (MGO) trapping capacity. Therefore, the REs are promising candidates worthy of further exploration for preventing AGEs accumulation in vivo, thereby treating AGEs-associated diseases.     

密闭系统中微波辅助萃取机制探讨

通过与普通加热萃取虎杖中白藜芦醇的产率进行对比 ,对密闭系统中微波辅助萃取的机理进行了探讨。结果表明 ,微波辅助萃取与普通加热萃取二者的活化能相差不大 ,分别为30.06和30.32kJ·mol-1。但微波辅助萃取的表观速率常数约是普通加热萃取的20倍。并且还利用电子扫描显微镜对样品微结构进行了观察 ,研究表明 ,细胞内极性成分如水吸收了微波能后压力迅速增加导致细胞结构的破坏是MAE快速高效的关键 ,由于细胞的破坏 ,萃取剂和萃取目标化合物更容易通过细胞壁 ,加速了扩散速度 ,进而加速了萃取速度。     

High Performance Liquid Chromatography—Tandem Mass Spectrometric Determination of Resveratrol and Its Metabolites in Rat Tissues

In this study, we established a rapid, sensitive, and specific liquid chromatography method coupled with electrospray ionization–tandem mass spectrometry to evaluate the disposition of resveratrol and its metabolites in rat tissues following oral administration. Here, we validate this method and show that it is useful for quantifying tissue concentrations of resveratrol and its metabolites with acceptable specificity, linearity, lower limits of quantification, precision, accuracy, and recovery. The present study demonstrates that the major distribution of the trans-resveratrol and its metabolites in investigated tissues, which suggests that there is a specified range of target organs for cancer chemoprevention by polyphenols.     

He-Ne激光诱变选育高产白藜芦醇细胞系

以葡萄皮脱分化组织为最初材料,用根癌农杆菌702菌株感染,通过纸电泳证明胭脂碱合成醇基因的存在,并在无激素培养基上培养5～6代以上,将此组织(MS)作为本次实验的研究对象.用He-Ne激光辐照,对MS进行诱变处理,通过继代选育,得到优良细胞系T₁₂,其特点是:生长迅速,生长周期比对照组(CK)缩短10～15天;生长曲线的稳定期明显延长,白藜芦醇平均产量比MS组提高40%.通过稳定性实验证明T₁₂组遗传性状是可靠的.
