In this paper, the laser spot diameter and its intensity distribution are measured with a scanning pinhole, and the keyhole shapes are observed using a specially designed setup in laser deep penetration welding of glass GG17. Based on the above experimental results, the effects of the following factors on the keyhole shapes are studied: the laser spot diameter and its intensity distribution, defocus, welding speed and inverse Bremsstrahlung absorption of the plasma.  相似文献

Keyhole shapes are observed experimentally by two high-speed cameras from two perpendicular directions in high-speed laser welding of glass. From the obtained keyhole pictures, it can be seen that in high-speed deep penetration laser welding, the keyhole is not only seriously bent in the direction opposite to that of welding speed, but also elongated along the direction of the welding speed. Based on the so-obtained keyhole photograph, the keyhole profiles in both the symmetric plane and its perpendicular plane (i.e., the cross-section plane) are determined by the method of polynomial fitting. Then, under the assumption of elliptical cross-section of the keyhole at each keyhole depth, a 3D bending keyhole is reconstructed, the behavior of focused Gaussian laser beam in the keyhole is analyzed by tracing a ray of light using geometrical optics theory. Fresnel absorption and multiple reflections in the keyhole are systematically studied, and the laser intensities absorbed on the keyhole walls are calculated. In determining the distribution of laser intensity on the keyhole wall, the bending of the keyhole plays the dominant role, elongation of the keyhole plays just a minor role. Because of the bending of the keyhole, not all the keyhole wall can be irradiated directly by laser beam. The absorbed laser intensity cannot be uniformly distributed on the keyhole wall even after multiple reflections. The keyhole wall absorbs laser intensity mainly on the small area near the front keyhole wall. Recoil pressure plays a dominant role in forming a keyhole and keeping it open.  相似文献

A polyclonal antibody (Ab) for the progestin levonorgestrel (LNG) was generated, and immunochemical assays for its detection, clean-up and concentration were developed. A highly specific microplate diagnostic assay for the detection of LNG was developed that used the enzyme linked immunosorbent assay (ELISA) method. The LNG ELISA developed was sensitive and reproducible; it exhibited I50 and I20 values of 3.3 ± 1.8 ng mL−1 and 0.6 ± 0.4 ng mL−1, respectively, and the Abs did not cross react with any of the tested steroid hormones. The above Abs were used to develop a sol-gel-based immunoaffinity purification (IAP) method for concentration and clean-up of LNG that is compatible with subsequent immunochemical or instrumental chemical analytical procedures, such as liquid chromatography followed by mass spectrometry (LC-MS/MS). Development of the columns included successful entrapment of Abs within a tetramethoxysilane (TMOS)-based SiO2 polymer network. The Abs could bind the free analyte from solution, and the bound analyte could be easily eluted from the sol-gel matrix at high recoveries. The Ab selectivity towards the antigen was high, in both ELISA and the sol-gel columns, but the entrapped Abs cross-reacted with two other steroid hormones - ethynylestradiol (EE2) and nortestosterone (NT) - which share similar epitopes with LNG, despite the lack of cross reactivity in the ELISA. The validity of the method was investigated by LC-MS/MS and a good analytical correlation was obtained.  相似文献

Keyhole diffusion tensor imaging (keyhole DTI) was previously proposed in cardiac imaging to reconstruct DTI maps from the reduced phase-encoding images. To evaluate the feasibility of keyhole DTI in brain imaging, keyhole and zero-padding DTI algorithms were employed on in vivo mouse brain. The reduced phase-encoding portion, also termed as the sharing rate, was varied from 50% to 90% of the full k-space. Our data showed that zero-padding DTI resulted in decreased fractional anisotropy (FA) and decreased mean apparent diffusion coefficient (mean ADC) in white matter (WM) regions. Keyhole DTI showed a better edge preservation on mean ADC maps but not on FA maps as compared to the zero-padding DTI. When increasing the sharing rate in keyhole approach, an underestimation of FA and an over- or underestimation of mean ADC were measured in WM depending on the selected reference image. The inconsistency of keyhole DTI may add a challenge for the wide use of this modality. However, with a carefully selected directive diffusion-weighted image to serve as the reference image in the keyhole approach, this study demonstrated that one may obtain DTI indices of reduced-encoding images with high consistency to those derived with full k-space DTI.  相似文献

Keyhole limpet hemocyanin (KLH): a biomedical review   总被引:10,自引:0,他引:10  
In this review we present a broad survey of fundamental scientific and medically applied studies on keyhole limpet hemocyanin (KLH). Commencing with the biochemistry of KLH, information on the biosynthesis and biological role of this copper-containing respiratory protein in the marine gastropod Megathura crenulata is provided. The established methods for the purification of the two isoforms of KLH (KLH1 and KLH2) are then covered, followed by detailed accounts of the molecular mass determination, functional unit (FU) structure, carbohydrate content, immunological analysis and recent aspects of the molecular genetics of KLH. The transmission electron microscope (TEM) has contributed significantly to the understanding of KLH structure, primarily from negatively stained images. We give a brief account of TEM studies on the native KLH oligomers, the experimental manipulation of the oligomeric states, together with immunolabelling data and studies on subunit reassociation. The field of cellular immunology has provided much relevant biomedical information on KLH and has led to the expansion of use of KLH in experimental immunology and clinically as an immunotherapeutic agent; this area is presented in some detail. The major clinical use of KLH is specifically for the treatment of bladder carcinoma, with efficacy probably due to a cross-reacting carbohydrate epitope. KLH also has considerable possibilities for the treatment of other carcinomas, in particular the epithelially derived adenocarciomas, when used as a carrier for carcinoma ganglioside and mucin-like epitopes. The widespread use of KLH as a hapten carrier and generalised vaccine component represent other major on-going aspects of KLH research, together with its use for the diagnosis of Schistosomiasis, drug assay and the treatment of drug addiction. Immune competence testing, assessment of stress and the understanding of inflammatory conditions are other areas where KLH is also making a useful contribution to medical research.  相似文献

The process of laser metal welding is widely used in industry. Nevertheless, there is still a lack of complete process understanding and control. For analyzing the process we used two high-speed cameras. Therefore, we could image the plasma plume (which is directly accessible by a camera) and the keyhole (where most of the process instabilities occur) during laser welding isochronously. Applying different image processing steps we were able to find a correlation between those two process characteristics. Additionally we imaged the plasma plume from two directions and were able to calculate a volume with respect to the vaporized material the plasma plume carries. Due to these correlations we are able to conclude the keyhole stability from imaging the plasma plume and vice versa. We used the found correlation between the keyhole behavior and the plasma plume to explain the effect of changing laser power and feed rate on the keyhole geometry. Furthermore, we tried to outline the phenomena which have the biggest effect on the keyhole geometry during changes of feed rate and laser power.  相似文献

Pulsed Nd:YAG laser welding of pure niobium plate to titanium alloy Ti-6Al-4V sheet in butt joint is studied regarding the laser/metal interaction modes. To obtain the optimized process parameters in dissimilar welding of Ti-6Al-4V/Nb, the melting ratio of laser beam energy for each weld counterpart is evaluated experimentally. Different laser welding modes of keyhole and conduction are predicted regarding the absorbed energy from the similar laser pulses on each weld counterpart. Laser keyhole and conduction welding were observed simultaneously through direct visualization of laser interaction with dissimilar metals using High Speed Imaging (HSI) system.  相似文献

The paper concerns laser–matter interaction characterisation. In this work, we use a rapid CCD camera located coaxially to the laser beam and we compare recorded images with those obtained by numerical modelling. Because images are difficult to understand, we compute thermal radiation emitted by a keyhole of fixed geometry and we adjust it trying to approach the camera record. The modelling treats radiative heat transfer within the keyhole and determines the sensor illumination map. By adjusting the geometrical characteristics of the hole, we seek to obtain the image that corresponds as well as possible to the realised experiment. Results are compared with other experimental methods simultaneously performed plume characterisation with an electric probe and spectrometric analysis. They show the existence of two distinct behaviours of the keyhole: a pseudo-steady state associated with regular and pseudo-constant keyhole shapes, low frequencies of electric current in the plume, and generally good welding results, and a highly dynamic mode associated with irregular and rapidly varying keyhole shapes, high frequencies in the plume current and generally poor welding results.  相似文献

MR ARFI measures the displacement induced by the ultrasonic radiation force and provides the location of the focal spot without significant heating effects. Displacements maps obtained with MR ARFI provide an indirect estimation of the acoustic beam intensity at the target. This measure is essential for dose estimation prior to focused ultrasound treatments (FUS) and adaptive focusing procedures of MR-guided transcranial and transribs FUS. In the latter case, the beam correction is achieved by maximizing the displacement at focus. A significant number of serial MR ARFI images are required and thus, a partial k-space updating method, such as keyhole appears as a method of choice. The purpose of this work is to demonstrate via simulations and experiments the efficiency of the keyhole technique combined with a two-dimensional spin-echo MR ARFI pulse sequence. The method was implemented in an ex vivo calf brain taking advantage of the a priori knowledge of the focal spot profile. The coincidence of the phase-encoding axis with the longest axis of the focal spot makes the best use of the technique. Our approach rapidly provides the focal spot localization with accuracy, and with a substantial increase to the signal-to-noise ratio, while reducing ultrasound energy needed during MR-guided adaptive focusing procedures.  相似文献

The formation of keyhole and transverse section of a laser-cut kerf with slight stripe undulations by a 1064 nm ms pulse laser on (0 0 0 1) sapphire was investigated. The morphologies of keyhole and transverse section surfaces were evaluated by SEM, and the composition of transverse section of laser-cut kerf was evaluated by EDS, XRD and XPS. The time scale for onset of vaporization and the keyhole depth with different laser pulse energies were calculated. The result suggests that the depth of keyhole is approximately directly proportional to laser pulse energy. On sapphire transverse section surface, the element ratio of Al to O deviates from the stoichiometry of sapphire, perhaps due to the oxygen removal from surface.  相似文献
