To search for more potent antitumor agent,a series of novel nitric oxide-donating colchicine(Col) derivatives(6a-f,8a and b) were synthesized by coupling nitrates with N-methyl colchiceinamide.Their cytotoxicity against four human cancer cell lines in vitro were evaluated by MTT assay.It was found that many of the derivatives displayed significant activity,particularly,compounds 6c,8a and 8b showed more potent cytotoxic activities than Col.  相似文献

The separation and UV absorbance detection of four toxic alkaloids, colchicine, thiocolchicine, colchicoside, and thiocolchicoside, on a microchip-based capillary electrophoresis device are reported. To increase the sensitivity of UV absorbance detection, optical cells with extended path lengths were integrated into the separation channel during the microfabrication process. The absorbance values realized on the microchip using these optical cells were proportional to the increase in average depths according to the Beer-Lambert Law, resulting in sensitivity enhancements by as much as five times. Linearity of response was observed from 5.0 to 500 mg L−1 of colchicine, with detection limits ranging from 2 to 6 mg L−1 depending upon the specific alkaloid and the dimension of the optical cell. The extraction of colchicine from spiked milk samples was performed and an average recovery rate of 83% with a relative standard deviation of 3.8% was determined using the optimized conditions on the microchip.  相似文献

The colchicine complexes with Li+, Na+ and K+ cations have been synthesized and studied by ESI MS, 1H and 13C NMR, FT-IR and PM5 semiempirical methods. It has been shown that colchicine forms stable complexes of 1:1 stoichiometry with monovalent metal cations. For Li+ cations also formation of the 2:1 stoichiometry complexes has been detected. The most stable structures of the complexes are those in which the acetamide groups are involved in the coordination process. The structures of the colchicine complexes with Li+, Na+ and K+ cations are visualized and discussed in details.  相似文献

We investigated the interaction between colchicine and bovine serum albumin (BSA) by fluorescence and UV–Vis absorption spectroscopy. In the mechanism discussion, it was proved that the fluorescence quenching of BSA by colchicine is a result of the formation of colchicine–BSA complex; van der Waals interactions and hydrogen bonds play a major role in stabilizing the complex. The modified Stern–Volmer quenching constant Ka and corresponding thermodynamic parameters ΔH, ΔG, ΔS at different temperatures were calculated. The distance r between donor (BSA) and acceptor (colchicine) was obtained according to fluorescence resonance energy transfer (FRET).  相似文献

Phenylalanine and tyrosine play a central role in Gloriosa superba L. calluses for the production of colchicine. The lack of biosynthetic precursors and signal inducing enzyme activity are responsible for the lower production of colchicine in vitro. B 5 medium nutrient grown calluses have a low content of colchicine, indicating that an optimal precursors level is required to increase PAL and TAL activity for colchicine accumulation. The complex characteristics of p-sulfonated calix[n]arene and colchicine were examined using various techniques. Cyclic voltammetry indicated that the structural matching and electrostatic interactions were the dominant stabilizing factors for the host–guest complexes. The method showed a long linear voltammetric range for p-sulfonated calix[4]arene from 1?×?10?8 to 1?×?10?6?mol?L?1 with a detection limit of 3?×?10?9?mol?L?1. Ultraviolet absorption spectroscopy confirmed that a 1:1 ratio complex was formed. Molecular mechanics showed that the benzene ring of colchicine entered the p-sulfonated calix[4]arene cavity. The solubility of colchicine increased with the p-sulfonated calix[4]arene concentration 50-fold from 0.13 to 6.4?mol?L?1. The simulation of cell membrane permeability indicated that colchicine was released from the colchicine-p-sulfonated calix[4]arene complex and entered the hydrophobic micelles. These results show that p-sulfonated calix[4]arene is suitable as a drug carrier for colchicine. This work has expanded applications of drug loading, transport, and targeted release for the treatment of gout.  相似文献

Numerous studies postulated the possible modes of anthelmintic activity by targeting alternate or extended regions of colchicine binding domain of helminth β-tubulin. We present three interaction zones (zones vide −1 to −3) in the colchicine binding domain of Haemonchus contortus (a helminth) β-tubulin homology model and developed zone-wise structure-based pharmacophore models coupled with molecular docking technique to unveil the binding hypotheses. The resulted ten structure-based hypotheses were then refined to essential three point pharmacophore features that captured recurring and crucial non-covalent receptor contacts and proposed three characteristics necessary for optimal zone-2 binding: a conserved pair of H bond acceptor (HBA to form H bond with Asn226 residue) and an aliphatic moiety of molecule separated by 3.75 ± 0.44 Å. Further, an aliphatic or a heterocyclic group distant (11.75 ± 1.14 Å) to the conserved aliphatic site formed the third feature component in the zone-2 specific anthelmintic pharmacophore model. Alternatively, an additional HBA can be substituted as a third component to establish H bonding with Asn204. We discern that selective zone-2 anthelmintics can be designed effectively by closely adapting the pharmacophore feature patterns and its geometrical constraints.  相似文献

本文研究了抗癌药物秋水仙碱在玻碳电极上的电化学行为。在HAc NaAc缓冲溶液中,秋水仙碱在玻碳电极上产生一不可逆的氧化峰,峰电位为1.22V(vs.SCE)。用差示脉冲伏安法测定,峰电流与秋水仙碱浓度在8.00×10-7~1.00×10-5mol/L范围内有良好的线性关系,检出限达1.00×10-7mol/L。将之应用于模拟血清样品的测定,结果令人满意。同时,对电极反应机理进行了初步探讨。  相似文献
