毛细管电泳用于水产品中五种抗生素的同时测定   总被引:29,自引:0,他引:29  
用毛细管电泳-紫外检测法同时测定水产品中的四环素(TC)、金霉素(CTC)、土霉素(OTC)、多西环素(DC)及氯霉素(CAP)的含量.研究了缓冲体系的酸度、浓度、添加剂、分离电压、进样时间以及温度等条件对分离的影响,得到了电泳的最优条件.在278nm波长处,分离电压为22kV,20mmol/L磷酸氢二钠-10mmol/L柠檬酸(pH值2.8,含0.25mmol/L Na2EDTA和体积分数为4.0%的吐温-80)运行缓冲液下,上述5种组份在25min内得到完全分离.5种抗生素的质量浓度和电泳峰面积在2.5~300.0mg/L和10.0~300.0mg/L范围内呈现良好的线性,TC、CTC、OTC、DC和CAP的相关系数(r)分别为0.9996、0.9992、0.9993、0.9934、0.9987,检测下限为0.5~1.5μg/mL。该方法灵敏度高,重现性好,操作简便,并已成功用于水产品鲫鱼中的5种抗生素残留的检测。  相似文献

A simple reversed flow injection colourimetric procedure for determining iron(III) was proposed. It is based on the reaction between iron(III) with chlortetracycline, resulting in an intense yellow complex with a suitable absorption at 435 nm. A 200 μl chlortetracycline reagent solution was injected into the phosphate buffer stream (flow rate 2.0 ml min−1) which was then merged with iron(III) standard or sample in dilute nitric acid stream (flow rate 1.5 ml min−1). Optimum conditions for determining iron(III) were investigated by univariate method. Under the optimum conditions, a linear calibration graph was obtained over the range 0.5–20.0 μg ml−1. The detection limit (3σ) and the quantification limit (10σ) were 0.10 and 0.82 μg ml−1, respectively. The relatives standard deviation of the proposed method calculated from 12 replicate injections of 2.0 and 10.0 μg ml−1 iron(III) were 0.43 and 0.59%, respectively. The sample throughput was 60 h−1. The proposed method has been satisfactorily applied to the determination of iron(III) in natural waters.  相似文献

建立了动物组织中金霉素残留测定的高效液相色谱柱后衍生法,研究了镁离子和草酸体系对金霉素荧光强度的影响。结果表明,镁离子浓度和草酸浓度为1∶1.2时,金霉素的荧光强度最强。动物组织样品以5%高氯酸提取,正己烷脱脂,C18净化,Hy-persil ODS C18(250×4.6 mm,5μm)分离,流动相为甲醇∶0.05 mol/L草酸=80∶20(V/V),流速为0.7 mL/min,柱后0.05 mol/L乙酸镁衍生,流速为0.1 mL/min,紫外检测器和荧光检测器同时测定,提高了金霉素残留定量灵敏度。紫外检测波长365nm,荧光检测波长eλx=360 nm,eλm=520 nm。  相似文献

Adsorption and controlled release of Chlortetracycline HCl to and from multifunctional polymeric materials (HEMA/MAA) hydrogels were investigated. P(HEMA/MAA) hydrogels were synthesized by gamma radiation-induced copolymerization of 2-hydroxyethylmethacrylate (HEMA) and methacrylic acid (MAA) in aqueous solution. The influence of copolymer composition and pH value of the surrounding medium on the type of water diffusion into the glassy polymer were discussed. Drug, Chlortetracycline HCl containing hydrogels, with different drug concentration to polymer ratios, was loaded by direct adsorption method. The influence of MAA content in the gel on the adsorption capacities of hydrogel was studied. Chlortetracycline HCl adsorption capacity of hydrogels was found to increase from 8 to 138 mg Chlortetracycline HCl per gram dry gel with increasing amount of MAA in the gel system and drug concentration. The effect of pH on the releasing behavior of Chlortetracycline HCl from gel matrix was investigated. In vitro drug release studies in different buffer solutions show that the basic parameters affecting the drug release behavior of hydrogel are the pH of the solution and MAA content of hydrogel.  相似文献

盐酸金霉素分子印迹电化学传感器的研制   总被引:1,自引:0,他引:1  
构建了一种选择性检测盐酸金霉素(CTC)的分子印迹电化学传感器。在NaClO4溶液中,以邻氨基酚(OAP)为功能单体,盐酸金霉素(CTC)为模板,通过循环伏安法在玻碳电极表面上聚合制备了CTC印迹敏感膜(MIPs)。在含0.005 mol/L K3[Fe(CN)6]及0.1 mol/L KCl的磷酸盐缓冲液(PBS)中,应用差分脉冲伏安法(DPV)研究了传感器的响应性能。DPV峰电流差与CTC浓度在2.0×10!8~6.1×10!7mol/L范围内呈线性关系,检出限为1.5×10!8mol/L(3σ)。实验表明,用甲醇/H2SO4混合洗脱溶液可以使传感器再生,对CTC的测定具有良好的重现性,并具有良好的储存稳定性。传感器对于干扰物氯霉素及青霉素没有响应,结构相似的四环素、土霉素有微弱的响应,显示了良好的选择性。在牛奶和鸡肉实际样品中所测得的CTC加标回收率为86.4%~96.9%。与文献报道的CTC检测方法相比,本传感器具有低的检测限,操作简便,整个过程无需衍生化处理,响应快,成本低。  相似文献

The rise of monolithic stationary phases offers to routine and research laboratories several advantages. In spite of their recent discovery, they have rapidly become highly popular separation media for liquid chromatography. Time reduction and economic reasons like e.g. a diminished use of mobile phase are the most important ones. At the same time, it was reported that these columns offer a faster and better separation. The aim of this article was to investigate the transferability of methods originally developed on conventional particle-packed C18 columns (XTerra RP18 and Zorbax RX), onto the more recent monolithic columns. Both types, conventional particle-packed and monolithic columns, were able to separate tetracycline, oxytetracycline and chlortetracycline from their respective impurities with sufficient resolution, but showed remarkably shorter analysis times and lower backpressures, improving the lifetime of the column.  相似文献

Residual emerging contaminants in wastewater sludge remain an obstacle for its wide and safe applications such as landfilling and bio-fertilizer. In this study, the feasibility of individual ultrasonication (UlS) and Fenton oxidation (FO) and combined, Ferro-sonication processes (FO) on the degradation of chlortetracycline (CTC) in wastewater sludge was investigated. UlS parameters such as amplitude and sonication time were optimized by response surface methodology (RSM) for further optimization of FS process. Generation of highly reactive hydroxyl radicals in FO and FS processes were compared to evaluate the degradation efficiency of CTC. Increasing in the ratio of hydrogen peroxide and iron concentration showed increased CTC degradation in FO process; whereas in FS, an increase in iron concentration did not show any significant effect (p > 0.05) on CTC degradation in sludge. The estimated iron concentration in sludge (115 mg/kg) was enough to degrade CTC without the addition of external iron. The only adjustment of sludge pH to 3 was enough to generate in-situ hydroxyl radicals by utilizing iron which is already present in the sludge. This observation was further supported by hydroxyl radical estimation with adjustment of water pH to 3 and with and without the addition of iron. The optimum operating UlS conditions were found to be 60% amplitude for 106 min by using RSM. Compared to standalone UlS and FO at 1:1 ratio, FS showed 15% and 8% increased CTC degradation respectively. In addition, UlS of sludge increased estrogenic activity 1.5 times higher compared to FO. FS treated samples did not show any estrogenic activity.  相似文献

A flow injection (FI) spectrophotometric determination of europium (III) is described, based on the complexation between europium (III), and chlortetracycline (CTC) in a Tris-buffer pH 8.0 medium. The resulting yellow-coloured complex is measured at its absorption maximum of 400 nm after 100 μl of sample or standard solution containing europium (III) are injected into the merged streams of CTC and Tris-buffer solutions. Optimum conditions for determining μg amounts of europium (III) are achieved by univariate method. Various types of reactors are also investigated. It is shown that the use of a single bead string reactor gives rise to the enhancement of peak height. A linear calibration curve over the range of 0.10-0.60 μg ml−1 europium (III) is established with the regression equation (n=6) Y=34.93X+0.01 and the correlation coefficient of 0.9994 is obtained. A detection limit (3σ) of 0.01 μg ml−1 of europium (III) and the relative standard deviation (R.S.D.) of 4.32% for determining 1.0 μg ml−1 of europium (III) (n=7) are obtained. The recommended method has been applied to the quantitation of europium (III) in spiked water and stream sediment samples with average recoveries of 99.9 and 97.5%, respectively.
