The widespread occurrence of antibiotics as contaminants in the aquatic environment has increased attention in the last years. The concern over the release of antibiotics into the environment is related primarily to the potential for the development of antimicrobial resistance among microorganisms. This article presents an overview of analytical methodologies for the determination of quinolone (Qs) and fluoroquinolone (FQs), macrolide (MLs), tetracycline (TCs), sulfonamide (SAs) antibiotics and trimethoprim (TMP) in different environmental waters. The analysis of these antibiotics has usually been carried out by high-performance liquid chromatography (HPLC) coupled to mass spectrometry (MS) or tandem mass spectrometry (MS/MS) and to a lesser extent by ultraviolet (UV) or fluorescence detection (FD). A very important step before LC analysis is sample preparation and extraction leading to elimination of interferences and prevention of matrix effect and preconcentration of target analytes.  相似文献

5-Fluorouracil (5FU) is in clinical use as an antitumor agent for the treatment of several types of solid tumors and cancers. However, development of drug resistance within the tumor cells and side effects has been a major limitation for the clinical use of 5FU. Preparation polymer-5FU conjugation is a promising potential antitumor drug and alternative pathway that could be used to treatment of cancer. For this purpose, water soluble poly(maleic anhydride-alt-N-vinyl pyrrolidone)[Poly(MA-alt-NVP)] is synthesized via charge transfer complex (CTC) copolymerization with benzoyl peroxide as an initiator at 80°C under nitrogen atmosphere. CTC mechanism is formed between through MA and NVP by determined using Uv-vis spectroscopy. Molar absorption coefficient (?AD) and equilibrium constant (KAD) of the complex are determined by Scott equation. The results obtained from indicate that copolymerization of MA:NVP system is preceded via alternating mechanism. The compositions of synthesized copolymers are also investigated by elemental analysis and the reactivity ratios of these monomers are calculated by using the elemental analysis data through Kelen-Tüdös, Mayo-Lewis, Fineman-Ross and Inverted Fineman-Ross equations. For preparation of polymer-drug conjugate, chemical modification is employed between the copolymer and 5FU. Polymer-drug conjugate and conjugation mechanism are enlightened by ATR-FTIR, NMR, HR-Raman and XRD methods.  相似文献

Circulating tumor cells (CTCs) present in the bloodstream are strongly linked to the invasive behavior of cancer; therefore, their detection holds great significance for monitoring disease progression. Currently available CTC isolation tools are often based on tumor-specific antigen or cell size approaches. However, these techniques are limited due to the lack of a unique and universal marker for CTCs, and the overlapping size between CTCs and regular blood cells. Dielectrophoresis (DEP), governed by the intrinsic dielectric properties of the particles, is a promising marker-free, accurate, fast, and low-cost technique that enables the isolation of CTCs from blood cells. This study presents a continuous flow, antibody-free DEP-based microfluidic device to concentrate MCF7 breast cancer cells, a well-established CTC model, in the presence of leukocytes extracted from human blood samples. The enrichment strategy was determined according to the DEP responses of the corresponding cells, obtained in our previously reported DEP spectrum study. It was based on the positive-DEP integrated with hydrodynamic focusing under continuous flow. In the proposed device, the parylene microchannel with two inlets and outlets was built on top of rectangular and equally spaced isolated planar electrodes rotated certain degree relative to the main flow (13°). The recovery of MCF7 cells mixed with leukocytes was 74%–98% at a frequency of 1 MHz and a magnitude of 10–12 Vpp. Overall, the results revealed that the presented system successfully concentrates MCF7 cancer cells from leukocytes, ultimately verifying our DEP spectrum study, in which the enrichment frequency and separation strategy of the microfluidic system were determined.  相似文献

Monosaccharide osazones are utilized in the spectrophotometric determination of their parent compounds through charge transfer complexes (CTC) formation with three-electron acceptor reagents. These acceptors indlude iodine (as -π-acceptor) and chloranil (tetrachloro P-benzoquinone) & TCNE (tetracyano ethylene) as π-acceptors. The presence of two phenylhydrazone moieties in the osazone molecule imparts electron-donating properties toward electron-accepting reagents, yielding spectrophotometrically measurable complexes. However, the reaction of osazone intermediates with TCNE reagent gave tricyanovinyl derivative rather than CTC formation. The above complexes together with their corresponding intermediates were subjected to structure elucidation studies to confirm the results and conclusions obtained. These studies include IR, Raman spectra, H1, C13-NMR, MS, ESR (electron spin resonance spectra) and microanalysis.  相似文献

A rapid epifluorescence staining method using the LIVE/DEAD® BacLight Bacterial Viability kit was applied to differentiate both viable and total counts of acetic acid bacteria in vinegar fermentation. The results obtained were compared with those from other measurement techniques: 4,6-diamidine-2phenyl indole (DAPI) and 5-cyano-2,3-ditolyl tetrazolium chloride (CTC) and colony counts. BacLight total counts were comparable to DAPI (differing by <3.5%). BacLight viable counts were similar to CTC counts but considerable higher than colony-forming cells in plates.  相似文献

The structures and stabilization of three crystal surfaces of TCNQ-based charge transfer complexes (CTCs) including PrQ(TCNQ)2, MPM(TCNQ)2, and MEM(TCNQ)2, have been investigated by scanning tunneling microscopy (STM). The three bulk-truncated surfaces are all ac-surface, which are terminated with TCNQ molecular arrays. On the ac-surface of PrQ(TCNQ)2, the TCNQ molecules form a tetramer structure with a wavelike row behavior and a γ angle of about 18° between adjacent molecules. Moreover, the dimer structures are resolved on both ac-surfaces of MPM(TCNQ)2 and MEM(TCNQ)2. In addition, the tetramer structure is the most stable structure, while the dimer structures are unstable and easily subject to the STM tip disturbance, which results in changeable unit cells. The main reasons for the surface stabilization variation among the three ac-surfaces are provided by using the 'π-atom model'.  相似文献
