Abstract

Sample handling is still a weak point in chromatography and in analytical chemisty in general. One consideration is the automation potential of new procedures. Solid-liquid extraction techniques in combination with pre-column technology are particularly promising in this regards. The construction and geometry of pre-columns both for conventional and narrow-bore HPLC are of major importance, since band broadening should be kept at a minimum for an optimal functioning of the analytical system. The various operations that can be carried out with such a pre-column are trace-enrichment, clean-up of the sample which depends on the type of adsorbents used in the precolumn, i.e., polar or apolar materials, ion exchangers or metal covered surfaces, etc., protection of the analytical column, field sampling and storage of samples and as a substrate for on-column chemical derivatizations. These various operations are demonstrated with practical examples from the fields of environmental and biological analysis. The selectivity can be further enhanced by coupling precolumn technology with selective detection modes such as diode array UV, electrochemical or fluorescence detection. This enables the construction of optimal and integrated analysis sytems which are fully automated and microprocessor controlled. They can also be made compatible with miniaturized LC-technology.  相似文献

美国《科学教师培养标准(2020年版)》针对《新一代科学教育标准》的推行而修订,立足于科学教育最新研究成果,尤其突出学科内容知识的重要性,补充对科学本质和科学文化的要求,强化对工程和技术、交叉概念、学科观念的理解,突出学习进阶规律在科学教师培养中的应用。此外,在学科教学法、学习环境、安全、对学生学习的影响等维度做出相应的改动。该标准启示我国:借鉴国际改革趋势出台中国特色的科学教师培养标准,但科学教师培养也要本土化;学科内容知识对科学教师专业发展非常重要;科学教师的培养应立足于学习进阶的规律;职前科学教师培养需要注重学习环境建设。  相似文献

采用定量的方法对112位化学师范生和83位中学化学教师进行问卷调查,了解职前、在职教师对教学能力要素重要性认识的情况,问卷涉及本体要素、工具要素、条件要素、实践要素、情意要素和发展要素等6大维度。基于Rasch测量模型,利用ConQuest,Winstep,SPSS22.0软件对数据进行统计分析。研究发现:(1)问卷具有良好的信效度;(2)师范生、在职教师均把情意要素作为教学能力的首要成分,其次依次是本体要素、发展要素、实践要素,工具要素与条件要素排在其他要素之后;(3)工具要素重要性赞同度师范生显著高于在职教师,其余维度差异不显著,不同性别各维度重要性赞同度也无显著性差异;(4)由ConQuest多维分析所建立的项目被试图可以直观展示不同维度上各教学能力的重要程度。  相似文献

运用复杂网络研究并联式项目群的结构脆弱性。将项目群中的任务用节点表示、任务之间的依赖关系用边表示,依据任务的工期确定点权、任务之间的依赖强度确定边权,则并联式项目群被抽象为一个有向加权网络。在分析并联式项目群网络拓扑特征的基础上,构建并联式项目群脆弱性评估模型,并对一个并联式项目群进行脆弱性评估。仿真结果表明该方法可以对并联式项目群脆弱性进行定量化评估、能够有效识别结构中的脆弱环节。本研究为保障项目群顺利实施、抵御风险提供了新的思路。  相似文献

Varying the temperature has been proven to be beneficial for improving the screening efficiency of protein crystallisation, and thus a crystallisation screening strategy based on this phenomenon can be developed. Such a temperature varying strategy can be applied in practical crystallisation screening, however, there are no guidelines for determining what temperature programme should be utilised. It is therefore necessary to investigate how the temperature programme affects the crystallisation process, so as to help people design a suitable temperature programme. For this purpose, we investigated the effect of temperature programmes on the protein crystallisation (lysozyme, proteinase K, and concanavalin A) that are characterised by different solubility behaviours with respect to temperature. Judging from the reproducibility studies of protein crystallisation with different temperature programmes, we recommend using linear temperature programmes for a moderate time period (24 to 48 h) and a large temperature range according to the properties of the proteins. The purpose of this study was to investigate pre-service teachers’ views about the history of mathematics course in which GeoGebra was used. The qualitative research design was used in this study. The participants of the study consisted of 23 pre-service mathematics teachers studying at a state university in Turkey. An open-ended questionnaire was used as a data collection tool. Qualitative data obtained from the pre-service teachers were analyzed by means of content analysis. As a result, it was determined that GeoGebra software was an effective tool in the learning and teaching of the history of mathematics.  相似文献

This paper describes students’ solutions to a commonly taught and not commonly taught inequality. The findings showed students’ difficulties. Participants implicitly and explicitly exhibited two intuitive beliefs: inequalities must result in inequalities and solving inequalities and equations are the same process. Following the analysis of students’ written solutions, individual interviews were conducted that gave a better insight into their reasoning and provided some ideas for teaching. The concluding section of the paper offers relevant educational implications.  相似文献

The objective of this case study was to investigate the ability of 10th graders and pre-service teachers to solve logical–mathematical thinking challenges. The challenges do not require mathematical knowledge beyond that of primary school but rather an informed use of the problem representation. The percentage of correct answers given by the 10th graders was higher than that of the pre-service teachers. Unlike the 10th graders, some of whom used various strategies for representing the problem, most of the pre-service teachers’ answers were based on a technical algorithm, without using control processes. The obvious conclusion drawn from the findings supports and recommends expanding and enhancing the development of logical–mathematical thinking, both in specific lessons and as an integral part of other lessons in pre-service frameworks.  相似文献
