在弱酸性条件下,丽春红G和妥布霉素反应后形成的产物导致体系的共振散射强度急剧增强,并在276、382和597 nm波长处产生3个新的共振散射峰,最大散射波长为597 nm.研究了反应的适宜条件和体系的光谱特征,妥布霉素的浓度在0.05~6.88μg/mL范围内与共振散射强度(ΔIRS)呈良好的线性关系,线性回归方程为△I597 nm=1.52c-2.68(R=0.9986),方法的检出限(3σ)为22.9 ng/mL.方法已用于硫酸妥布霉素注射液和滴眼液中含量的测定,结果满意. 相似文献

研究发现过渡金属超常氧化态配合物(二羟基二过碘酸根合铜(Ⅲ)配离子(DPC)、二羟基二过碘酸根合银(Ⅲ)配离子(DPA)和二羟基二过碘酸根合镍(Ⅳ)配离子(DPN))在碱性条件下可以氧化鲁米诺而产生化学发光,氨基糖苷类抗生素对该化学发光体系有增敏作用。以DPC为例研究了氨基糖苷类抗生素——托普霉素对该体系的增敏作用,建立了测定血清中托普霉素含量的新方法。考察了溶液酸碱度和化学发光试剂对化学发光强度的影响,在最佳实验条件下,托普霉素浓度在6.0×10-8~2.0×10-6g/mL范围内与化学发光强度呈良好的线性关系,方法的检出限(3σ)为1.5×10-8g/mL,对浓度为5.0×10-7g/mL的托普霉素溶液连续测定7次,相对标准偏差为2.7%。该方法用于血清中托普霉素含量的测定,结果令人满意。 相似文献

Elimination of pulmonary Pseudomonas aeruginosa (PA) infections is challenging to accomplish with antibiotic therapies, mainly due to resistance mechanisms. Quorum sensing inhibitors (QSIs) interfering with biofilm formation can thus complement antibiotics. For simultaneous and improved delivery of both active agents to the infection sites, self-assembling nanoparticles of a newly synthesized squalenyl hydrogen sulfate (SqNPs) were prepared. These nanocarriers allowed for remarkably high loading capacities of hydrophilic antibiotic tobramycin (Tob) and a novel lipophilic QSI at 30 % and circa 10 %, respectively. The drug-loaded SqNPs showed improved biofilm penetration and enhanced efficacy in relevant biological barriers (mucin/human tracheal mucus, biofilm), leading to complete eradication of PA biofilms at circa 16-fold lower Tob concentration than Tob alone. This study offers a viable therapy optimization and invigorates the research and development of QSIs for clinical use. 相似文献

Two new related methods, 2BOB and H2OBC, for tracking out the backbone of protonated 13C nuclei are presented. 2BOB extracts an H2BC and an HSQC‐type spectrum from one and the same data set, and the combined information of these two spectra tracks out the molecular backbone. The faster method, H2OBC, typically requiring only a few minutes of instrument time, yields a single spectrum with distinct and different phases imposed on the H2BC and one‐bond peaks thus obviating the need to separate them in the absence of complicating spectral overlap. Colorimetric silver nanoparticle sensor was developed for determination of aminoglycosides in milk. Silver nanoparticles were synthesized by using sodium borohydride as reducing agent and sodium dodecyl sulfate as stabilizer. Yellow color of silver turned into orange and red in proportion to the concentrations of analytes. Quantitative analyses were performed by using decrease in absorbance of silver nanoparticles at 394 nm. Linear ranges were 20–60 ng mL?1, 23–60 ng mL?1, and 60–100 ng mL?1 for gentamicin, tobramycin, and amikacin, respectively. The method was optimized in terms of pH, ionic strength, and time. This simple and validated method was applied to milk samples and pharmaceutical preparations. 相似文献

本文提出了一种新型的检测妥布霉素的电化学适配体传感器,以差分脉冲伏安法(DPV)作为检测技术,亚甲基蓝作为电化学响应信号. 构建了以纳米复合材料金纳米粒子/聚苯胺/二氧化钛纳米管(AuNPs/PANI/TNTs)修饰玻碳电极的电极支架. 通过透射电子显微镜和X-射线光电子能谱对纳米复合材料进行详细的表征. 循环伏安图和电化学阻抗谱显示AuNPs/PANI/TNTs可以很好地增加电极的界面传导性能. DPV结果显示电流密度的响应和妥布霉素浓度之间存在一个很好的线性关系,并且得到一个宽广的检测范围为0.5 μmol·L-1到70 μmol·L-1. 本文提出的适配体基的传感器有很好的重复性和稳定性,作为一个潜在的手段可以应用在生物分析和医疗诊断中. 相似文献

Defective osteogenesis and latent infections continue to be two major issues in the therapy of bone tissue regeneration. In this study, a unique hierarchically micro/nanoscale‐architecture is first proposed and produced on polyetheretherketone (PEEK). Besides, a “simvastatin‐PLLA film‐tobramycin microspheres” delivery system is subsequently fabricated to endow the PEEK implant with osteogenic and antibacterial capabilities. In vitro antibacterial evaluations confirm that the decorated PEEK scaffolds possess excellent resistance against planktonic/adherent bacteria. In vitro cell attachment/proliferation, lactate dehydrogenase (LDH) content, alkaline phosphatase (ALP) activity, calcium mineral deposition experiments, and real‐time PCR analysis all exhibit that the superior proliferation rate and osteo‐differentiation potential of MC3T3‐E1 pre‐osteoblasts are presented on the PEEK samples with dual functional decoration. In the mouse calvarial defect model, the micro‐CT and histological results demonstrate that our scaffolds display a remarkable bone forming capability. Generally, the PEEK scaffolds co‐endowed with simvastatin and tobramycin microspheres possess great potential in clinics. 相似文献
