通过比较法研究了不同钙源在水热条件下合成羟基磷灰石的方法。SEM,XRD和FT-IR的测试结果表明,蛋壳不仅可以成为有效钙源,而且多孔结构还可充当硬模板,在磷酸二氢钠缓慢腐蚀CaCO_3的同时提供PO_4~(3-)诱导生成花状羟基磷灰石。研究表明,磷酸肌酸钠水热缓慢分解释放磷酸根在合成高纯度羟基磷灰石方面有着广阔的应用前景。  相似文献

A nonradioactive 31P nuclear magnetic resonance (NMR) spectroscopy protocol has been developed and used to investigate in vitro autophosphorylation of insulin receptors. Optimum experimental conditions have been explored, and the effects of Mn2+ and phosphocreatine (PCr) on the determination of the phosphorylation reaction have been assayed. The method was used to monitor the time courses of the phosphorylation reaction in solution. The results from this NMR study were in agreement with observations of insulin receptor phosphorylation made by using Western blotting.   相似文献

1H MR spectra of human muscles were recorded before, during, and after fatiguing exercise. In contrast to expectations, it was found that the spectral contributions of creatine/phosphocreatine (Cr/PCr) were subject to change as a function of exercise. In particular, the dipolar-coupled methylene protons of Cr/PCr were found to be reduced in intensity in proportion to the co-registered PCr levels. Recovery after exercise and behavior under ischemic conditions provide further evidence to suggest that the contributions of the CH2 protons of Cr/PCr to 1H MR spectra of human muscle in vivo reflect PCr rather than Cr levels. Variation of experimental parameters showed that this effect is not due to a trivial change in relaxation times. At present it can only be speculated about why the Cr resonances have reduced NMR visibility. If temporary binding to macromolecules should be involved, the free Cr concentration-important for equilibrium calculations of the creatine kinase reaction-might be different from what was previously assumed.  相似文献

The nuclear magnetic resonance (NMR) technique was used as analytical tool to determine the complete metabolic profiling of sea bass extracts: water-soluble metabolites belonging to different classes such as sugars, amino acids, dipeptides and organic acids as well as metabolites soluble in organic solvent such as lipids, sterols and fatty acids were identified. The metabolite profiling together with a suitable statistical analysis were used to discriminate between wild and cultured sea bass samples. Preliminary results show that discrimination between wild and cultured sea bass was obtained not only using fatty acid composition but also cholesterol and phosphatidylethanolamine and some water-soluble metabolites such as choline, trimethylamine oxide, glutamine, fumaric and malic acids.  相似文献

Localized water suppressed proton spectroscopy has opened up a new field of pathophysiological studies of severe brain ischemia. The signals obtained with the pulse sequences used so far are both T1 and T2 weighted. In order to evaluate the extent to which changes in metabolite signals during the course of infarction can be explained by changes in T1 and T2 relaxation times, eight patients with acute stroke were studied. STEAM sequences with varying echo delay times and repetition times were used to measure T1 and T2 of N-acetyl-aspartate (NAA), creatine plus phosphocreatine (Cr+PCr) and choline containing compounds (CHO) in a 27-ml voxel located in the affected area of the brain. Ten healthy volunteers served as controls. We found no difference in T1 or T2 of the metabolites between the patients and the normal controls. The T2 of CHO was longer than that of NAA and Cr+PCr. Our results indicate that spectra obtained in brain infarcts and normal tissue with the same acquisition parameters are directly comparable with respect to relative signal intensities as well as signals scaled with internal and external standards.  相似文献

The forearm muscles of five healthy males were monitored for changes in microvessel hemoglobin saturation (SO2-TRS) by near infrared time-resolved spectroscopy (NIRTRS) and changes in phosphorus metabolites by magnetic resonance spectroscopy (31P-MRS) during 12 min of resting arterial occlusion. Muscle oxygenation and phosphorus metabolites were also measured during grip exercises at varying intensities. Upon the initiation of occlusion, SO2-TRS fell progressively until it reached a plateau in the latter half of the occlusion. Phosphocreatine (PCr) began to decrease around 6 min after the initiation of arterial occlusion. The resting O2 store and O2 consumption were 295 μM and 0.95 μM/sec, respectively-values which reasonably agree with the reported results. A significant correlation was observed between the changes in SO2-TRS and PCr during exercise (r2 = 0.80, p < 0.001). These results indicate that NIRTRS is able to provide reliable information about resting metabolism and oxidative rate during exercise. NIRTRS and MRS are useful to monitor oxygenation and energetics noninvasively in the human muscle.  相似文献

建立了离子对反相高效液相色谱法(IP-RP-HPLC)同时测定家兔血浆中外源性磷酸肌酸(PCr)及其代谢产物肌酸(Cr)的方法,用于研究外源性PCr在家兔体内的药代动力学。以含离子对试剂四丁基硫酸氢铵(TBA)的磷酸盐缓冲液-甲醇为流动相,在Kromasil-C18色谱柱上进行梯度洗脱。采用内标法定量、以基线扣除法计算外源性PCr和Cr的浓度。PCr和Cr的线性范围分别为10~7500 mg/L和10~1500 mg/L;日内和日间精密度均≤6.2%,准确度分别为99.7%~102.2%和96.5%~102.4%;萃取回收率均大于92%。静脉注射PCr后,血浆中PCr的消除为二室模型,消除半衰期为(20.4±2.7) min;表观分布容积为(0.179±0.037) L/kg;清除率为(0.019±0.002) L/(kg\5min);静脉注射PCr后血浆中迅即出现降解产物Cr,其达峰时间为30 min;消除半衰期为(43.7±4.5) min。本方法的专属性强,准确度和精密度高,能特异性地测定家兔血浆中的PCr和Cr。实际应用结果表明,该方法完全符合PCr药代动力学生物分析方法学的要求。  相似文献

A simple and rapid HPLC–MS/MS method was developed and validated for simultaneous measurement of phosphocreatine and its metabolites creatine and creatinine in children's plasma. A 50 μL aliquot of plasma was prepared by protein precipitation with acetonitrile–water (1000 μL, 1:1, v/v) followed by separation on a Hypersil Gold C18 column (35°C) with gradient mobile phase consisting of 2 mm ammonium acetate aqueous solution (pH 10) and methanol at a flow rate of 0.3 mL/min and analyzed by mass spectrometry in both positive (phosphocreatine) and negative (creatine and creatinine) ion multiple reaction monitoring mode. Good linearity (r > 0.99) was obtained for the three analytes. The intra‐day and inter‐day values of CV were <5.46% (?13.09% ≤ RE ≤ 2.57%). The average recoveries of the three analytes were 70.9–97.5%. No obvious impact was found for the quantitation of three analytes in normal, hemolyzed and hyperlipemic plasma. In the end, this method was successfully applied to a pharmacokinetic study of phosphocreatine in children (six cases) with viral myocarditis of children after intravenous infusion of 2 g of the test drug. The pharmacokinetc parameters of phosphocreatine/creatine were as follows: t1/2 0.24/0.83 h, Tmax 0.49/0.55 h, Cmax 47.34/59.29 μg/mL, AUClast 17.07/59.63 h μg/mL, AUCinf 17.16/79.01 h μg/mL and MRT 0.29/0.67 h.  相似文献

 采用反相离子对液相色谱法测定肌肉组织中的磷酸肌酸 (PCr)的含量。样品经高氯酸溶液沉淀蛋白 ,匀浆后离心 ,上清液用KOH溶液中和后用反相液相色谱法分离测定。方法的最低检测限为 2mg/L(S/N≥ 3) ,PCr在 5mg/L~ 10 0mg/L范围内与其峰面积呈良好的线性关系 (r =0 .9992 ) ;PCr的平均回收率为 99.34 % ;测定结果的日内RSD <4.42 % ,日间RSD <8.6 7%。方法准确、灵敏、快速 ,适用于动物和人体肌肉组织中PCr含量的测定。  相似文献
