A series of pure silica MSU and carboxylic-modified MSU materials were prepared. The formation of mesoporous silica materials with terminal carboxylic groups on pore surface was performed by the acid-catalyzed hydrolysis of cyano to carboxylic. Then their potential applications in controlled drug delivery carriers were investigated. Drug famotidine was selected as a model molecule out of the consideration of the terminal amino groups in its molecule. The adsorption experiments show significant adsorption of famotidine on the carboxylic-modified MSU materials. And, the functionalization level of carboxylic groups has been found to be the key factor affecting the adsorption capacities of the modified MSU materials for famotidine. Subsequently, three kinds of release fluids, including simulated gastric medium, simulated intestinal medium, and simulated body fluid, were used to test the famotidine release rate from the carboxylic-modified MSU material. SLIDE software, which models the flexibility of protein and ligand side chains while docking, was used to screen several large databases to identify inhibitors of Brugiamalayi asparaginyl-tRNA synthetase (AsnRS), a target for anti-parasitic drug design. Seven classes of compounds identified by SLIDE were confirmed as micromolar inhibitors of the enzyme. Analogs of one of these classes of inhibitors, the long side-chain variolins, cannot bind to the adenosyl pocket of the closed conformation of AsnRS due to steric clashes, though the short side-chain variolins identified by SLIDE␣apparently bind isosterically with adenosine. We hypothesized that an open conformation of the motif 2 loop also permits the long side-chain variolins to bind in the adenosine pocket and that their selectivity for Brugia relative to human AsnRS can be explained by differences in the sequence and conformation of this loop. Loop flexibility sampling using Rigidity Optimized Conformational Kinetics (ROCK) confirms this possibility, while scoring of the relative affinities of the different ligands by SLIDE correlates well with the compounds’ ranks in inhibition assays. Combining ROCK and SLIDE provides a promising approach for exploiting conformational flexibility in structure-based screening and design of species selective inhibitors. 相似文献
Summary: Mesoporous silica was used as substrate for the grafting of alkyl halides initiators. The control over the surface‐initiated polymerization of styrene and MMA, in terms of molar mass and molar mass distribution, was successfully achieved using an ATRP mechanism. The occurrence of the polymerization inside the mesopores was confirmed by thermogravimetric analysis.

Four common mesostructured aluminosilicates (e.g. AlMCM‐41, AlMCM‐48, AlMSU‐1 and AlSBA‐15, Si/Al = 7.5) with different pore framework structures were prepared via post‐synthetic method utilizing aluminum isopropoxide as the aluminating agent and characterized by 27Al MAS NMR, XRD, N2‐adsorption isotherm and NH3‐TPD. It was found that, on the basis of the above analysis results, AlMCM‐41, AlMSU‐1 and AlSBA‐15 retained more mesoporosity than cubic Al/MCM‐48 after exposure to pure steam at 800 °C, which may be responsible for their much higher activities in bulky 1,3,5‐triisopropylbenzene (TIPB) conversion. 相似文献
