Ethanolic extracts of samples of temperate zone propolis, four from the UK and one from Poland, were tested against three Trypanosoma brucei strains and displayed EC50 values < 20 µg/mL. The extracts were fractionated, from which 12 compounds and one two-component mixture were isolated, and characterized by NMR and high-resolution mass spectrometry, as 3-acetoxypinobanksin, tectochrysin, kaempferol, pinocembrin, 4′-methoxykaempferol, galangin, chrysin, apigenin, pinostrobin, cinnamic acid, coumaric acid, cinnamyl ester/coumaric acid benzyl ester (mixture), 4′,7-dimethoxykaempferol, and naringenin 4′,7-dimethyl ether. The isolated compounds were tested against drug-sensitive and drug-resistant strains of T. brucei and Leishmania mexicana, with the highest activities ≤ 15 µM. The most active compounds against T. brucei were naringenin 4′,7 dimethyl ether and 4′methoxy kaempferol with activity of 15–20 µM against the three T. brucei strains. The most active compounds against L. mexicana were 4′,7-dimethoxykaempferol and the coumaric acid ester mixture, with EC50 values of 12.9 ± 3.7 µM and 13.1 ± 1.0 µM. No loss of activity was found with the diamidine- and arsenical-resistant or phenanthridine-resistant T. brucei strains, or the miltefosine-resistant L. mexicana strain; no clear structure activity relationship was observed for the isolated compounds. Temperate propolis yields multiple compounds with anti-kinetoplastid activity.  相似文献

A three-dimensional quantitative structure-activity relationship (3D-QSAR) study using Comparative Molecular Similarity Indices Analysis (CoMSIA) was conducted on a series of 3-azolylmethylindoles as anti-leishmanial agents. Evaluation of 24 compounds synthesized in our laboratory served to establish the model. A random search was performed on the library of compounds, and molecules of the training set were aligned on common elements of template molecule 13, one of the most active compounds. The best predictions were obtained from multifit procedure with a CoMSIA model combining steric, electrostatic, hydrophobic and hydrogen bond acceptor fields (q 2?=?0.594, r 2?=?0.897). The model was validated using an external test set of 7 compounds giving a satisfactory predictive r 2 value of 0.649. Information obtained from CoMSIA contour maps could be used for further design of more promising inhibitors.  相似文献

Determination of berberine, an isoquinoline plant alkaloid, with antibacterial, antiparasitic, antifungal, hypotensive and antitumoral effects, was proposed by introducing square wave voltammetry on boron‐doped diamond electrode. At optimized experimental parameters, in Britton‐Robinson buffer solution pH 5 berberine provides 3 oxidation peaks (+0.63; +1.14 and +1.34 V) and one reduction (+0.15 V) (vs. Ag/AgCl electrode), with good repeatability (relative standard deviation of 2.6 % and 1.9 % for 8 measurements at 0.5 and 10 µM concentration level, respectively). Calibration curve was linear in wade linear range from 0.1 to 50 µM with limit of detection of 0.04 µM. The proposed procedure was successfully applied for the determination of berberine in seed extract from Argemone mexicana with satisfactory recovery (102–102.6 %). The developed method may represent a sensitive alternative to highly toxic mercury electrodes, modified electrodes and chromatographic methods.  相似文献

Tilia americana var. mexicana (T. americana) is a plant widely used in Mexico for its medicinal properties on the central nervous system. In the present study, we designed a protocol to investigate the neuroprotective effects of non-polar and polar extracts of T. americana on damage induced by cerebral ischaemia in mice. Vehicle or extracts were administered immediately after ischaemia. Functional neurological deficit, survival percentage and infarct area were determined in each experimental group. Results showed that groups treated with non-polar or polar extracts of T. americana had increased survival rate, improved neurological deficits and diminished the infarct area in relation to the ischaemic group. In conclusion, this study confirms the neuroprotective activity of T. americana, suggests a possible synergism between non-polar and polar constituents and supports its potential as a useful aid in the clinical management of stroke.  相似文献
