Novel hexadecyl 3- { 4-[2-hydroxy-3(isopropylamino)propoxy]phenyl }propionate (HPP)was synthesized and its effect on delivery of liposomes into cultured cardiomyocytes was examined. The structure of HPP was characterized by IH NMR, 1R and MS. The amount of cardiomyocytes uptake of HPP-liposome was 3.9-fold higher than plain-liposome, and the increase was 6.2-fold when hypoxia happens. It indicated that HPP was a potential ligand for liposome targeting to ischemic myocardium.  相似文献

In the current research, through employing the dual ligand method, a novel coordination polymer has been produced in success via the reaction between H2glu and In(NO3)3·6H2O in the existence of the nitrogen-donor chelating 2,2′-bpy ligand in CH3CN In(III) and DMF mixed solvent, and its chemical formula is In(glu)(Hglu)(2,2′-bpy) (1, 2,2′-bpy is 2,2′-bipyridine and H2glu is glutaric acid). Its application values on the acute stroke were assessed and the corresponding mechanism was investigated simultaneously. Firstly, the levels of inflammatory response in the astrocytes were evaluated by ELISA by measuring the content of inflammatory cytokines released into cerebrospinal fluid. Additionally, the miRNA199a relative expression levels in astrocytes were measured via exploiting real time RT-PCR. Molecular docking simulation demonstrated that synthesized In ion complex exhibited excellent biological activities, multiple binding interactions were formed by the carboxyl groups on the In ion complex.  相似文献

Maca (Lepidium meyenii) has emerged as a popular functional plant food because of its medicinal properties and nutritional value. Macamides, as the exclusively active ingredients found in maca, are a unique series of non-polar, long-chain fatty acid N-benzylamides with multiple bioactivities such as antifatigue characteristics and improving reproductive health. In this study, a new kind of macamide, N-benzyl eicosapentaenamide (NB-EPA), was identified from maca. We further explore its potential neuroprotective role in hypoxic–ischemic brain injury. Our findings indicated that treatment with biosynthesized NB-EPA significantly alleviates the size of cerebral infarction and improves neurobehavioral disorders after hypoxic–ischemic brain damage in neonatal mice. NB-EPA inhibited the apoptosis of neuronal cells after ischemic challenge. NB-EPA improved neuronal cell survival and proliferation through the activation of phosphorylated AKT signaling. Of note, the protective property of NB-EPA against ischemic neuronal damage was dependent on suppression of the p53–PUMA pathway. Taken together, these findings suggest that NB-EPA may represent a new neuroprotectant for newborns with hypoxic–ischemic encephalopathy.  相似文献

11C, 18F and 123I fatty acids are used for myocardial imaging, and 99mTc‐labeled fatty acids are more desirable substitutes than other radiolabeled fatty acids. In the work reported, [99mTc]‐CpTT‐10‐oxo‐FPA ( 1c ), [99mTc]‐CpTT‐12‐oxo‐FPA ( 2c ), [99mTc]‐CpTT‐14‐oxo‐FPA ( 3c ) and [99mTc]‐CpTT‐16‐oxo‐FPA ( 4c ) were prepared with 60.76–70.92% of radiochemical yield and purity of more than 95%. These radiotracers ( 1c , 2c , 3c , 4c ) were chemically stable when incubated in Sprague Dawley rat serum for 3 h at 37 °C. Tissue distribution studies in female mice indicated that 2c had high initial heart uptake (8.84%ID g?1 at 1 min post‐injection) and 4c had long retention in the heart (1.45%ID g?1 at 30 min post‐injection). Metabolite analysis showed 4c could be metabolized to 5c via β‐oxidation with loss of two ? CH2? in the myocardium, the radiometabolite being excreted via urine. However, low heart uptake suggested that 4c cannot be used as a diagnostic imaging agent. Copyright © 2016 John Wiley & Sons, Ltd.  相似文献

The protective effect of ischemic preconditioning on brain tissue during total ischemia is shown by using spectral-luminescence methods for estimating oxidative damage to blood components after ischemia. Fluorescence of blood serum and molecular probes added to it is investigated after producing brain ischemia with preconditioning. Carotid artery stenosis (CAS) is an atherosclerotic disease characterized by a narrowing of the artery lumen and a high risk of ischemic stroke. Risk factors of atherosclerosis, including smoking, hypertension, hyperglycemia, hyperlipidemia, aging, and disrupted circadian rhythm, may potentiate atherosclerosis in the carotid artery and further reduce the arterial lumen. Ischemic stroke due to severe CAS and cerebral ischemic/reperfusion (I/R) injury after the revascularization of CAS also adversely affect clinical outcomes. Melatonin is a pluripotent agent with potent anti-inflammatory, anti-oxidative, and neuroprotective properties. Although there is a shortage of direct clinical evidence demonstrating the benefits of melatonin in CAS patients, previous studies have shown that melatonin may be beneficial for patients with CAS in terms of reducing endothelial damage, stabilizing arterial plaque, mitigating the harm from CAS-related ischemic stroke and cerebral I/R injury, and alleviating the adverse effects of the related risk factors. Additional pre-clinical and clinical are required to confirm this speculation.  相似文献

探讨数字减影血管成像(DSA)、计算机断层扫描血管成像(CTA)联合磁共振(MR)影像评估急性缺血性卒中(AIS)患者脑支循环及预后性关系。选取60例大脑中动脉M1段急性闭塞所致AIS患者为研究对象,根据DSA、CTA与MR影像对其脑侧支循环评估,比较患者基线资料、结局指标等,并分析预后性。结果发现:基于DSA、CTA与MR影像对AIS患者脑侧支循环评估结果一致性良好;3种影像模式下脑侧支循环良好组与不良组结局资料差异显著(P<0.05);多因素分析显示,FVH-ASPECTS评分、rLMC评分、ASITN/SIR分级量表均为AIS患者神经功能预后的独立影响因素。总之,DSA、CTA、MR影像对AIS患者脑侧支循环评估具有一致性,且FVH-ASPECTS评分、rLMC评分、ASITN/SIR分级量表均为AIS患者神经功能预后的独立影响因素。  相似文献

为探讨新生鼠缺氧缺血性脑病脑组织中 5种元素 (Fe2 + 、Ca2 + 、Zn2 + 、Cu2 + 、Mg2 + )的变化及意义 ,将新生Wistar鼠 2 4只随机分为正常对照组和缺氧缺血性脑病组 ,乙醚吸入麻醉后用1 0双线结扎右侧颈总动脉 ,术后恢复 4h ,吸入含 8%O2 和 92 %N2 混合气体 2h后将 2组大鼠处死 ,取脑组织 ,用原子吸收分光光度仪测定其微量元素的变化。结果表明 ,与正常对照组相比较 ,缺氧缺血性脑病脑组织中Zn2 + 、Mn2 + 、Cu2 + 含量明显减少 (P <0 0 1 ) ;Ca2 + 增加 (P <0 0 1 )。提示缺氧缺血性脑病能引起大鼠脑组织微量元素的改变  相似文献

Total aralosides of Aralia elata (Miq) Seem (TASAES) from Chinese traditional herb Longya Aralia chinensis L was found to improve cardiac function. The present study was to determine the protective effects of TASAES on diabetic cardiomyopathy, and the possible mechanisms. Therefore, a single dose of streptozotocin was used to induce diabetes in Wister rats. Diabetic rats were immediately treated with low, medium and high doses of TASAES at 4.9, 9.8 mg/kg and 19.6 mg/kg body weight by gavage, respectively, for eight weeks. Cardiac function was evaluated by in situ hemodynamic measurements, and patch clamp for the L-type Ca2+ channel current (ICa2+-L) and transient outward K+ channel current (Ito). Histopathological changes were observed under light and electron microscope. The expression of pro-fibrotic factor, connective tissue growth factor (CTGF) was monitored using immunohistochemistry staining. Compared with diabetic group, medium and high doses, but not low dose, of TASAES showed a significant protection against diabetes-induced cardiac dysfunction, shown by increased absolute value of left ventricular systolic pressure (LVSP) and maximum rates of pressure development (±dp/dtmax), and enhanced amplitude of ICa2+-L (P < 0.05). Histological staining indicated a significant inhibition of diabetes-caused pathological changes and up-regulation of CTGF expression (P < 0.05). The results suggest that TASAES prevents diabetes-induced cardiac dysfunction and pathological damage through up-regulating ICa2+-L in cardiac cells and decreasing CTGF expression.  相似文献
