Seven flavonol glycosides were isolated and identified from the aerial parts of Dorycnium hirsutum, together with catechin, D-pinitol, β-sitosterol, and stearic acid. The extracts and the isolated flavonol glycosides were evaluated for their antioxidant activity, using the DPPH test (radical scavenging) and the lipoxygenase assay (lipid peroxidation). Abstract

Phytochemical studies on the leaves of Epimedium koreanum Nakai have resulted in the discovery of two new flavonol glycosides, koreanoside F (1) and koreanoside G (2), along with six known flavonoids. Their structures were elucidated on the basis of HRESIMS, UV, IR, 1?D NMR and 2?D NMR data. Absolute configurations of 1 and 2 was further determined by 13C-NMR spectra with gate decoupling (GD). All of the compounds were evaluated for cytotoxic activities by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl-2H-tetrazoliumbromide (MTT) assay. The results indicated that compounds 3, 5, 6, 7 and 8 inhibited the proliferation of A549 and NCI-292 cells with IC50 values of 5.7–23.5?μM. Real-time monitoring in three kinds of lung cancer cells and a kind of human bronchial epithelial cells treated with compound 6 was also assessed.  相似文献

Obesity is becoming increasingly common all over the world and global strategies are accordingly being developed to prevent it. In order to support the strategies, the effects of green apple (Golden Delicious) and the consumption of its three major flavonols (quercetin-3-glucoside, quercetin-3-D-galactoside, and quercetin-3-rhamnoside) on body weight; the weight of liver, kidney, and spleen; some lipid parameters in serum; and total lipid ratios of liver and kidney and oxidative stress parameters of obese rats were studied. This study was conducted on two experimental groups: one of which was given an apple, and the other was given flavonols, in addition to their high-energy diet; along with a sham and a control rat group, for 4 weeks. According to results, there was no difference in body and organ weights between groups. The liver and kidney weights increased in obese rats, but there was no difference between the total lipid ratios in these organs. The addition of green apple and selected flavonols to the high-energy diet of rats was not sufficient to prevent the increase in body and organ weights, but it supported the reduction in some lipid fractions and in oxidative stress parameters of obese rats. Moreover, this study supported the argument that obesity causes most of the lipid fractions increase in serum and induces oxidative stress.  相似文献

在B3LYP/6-31G水平下优化了3种黄酮醇类(山奈酚,槲皮素,杨梅素)化合物的几何构型. 在振动分析中,均未出现虚频率. 在B3LYP/6-31G的水平下计算了该类化合物的核磁共振碳谱. 研究结果表明:3种分子均有分子内氢键形成,且分子内氢键的键长为0.17~0.18 nm左右 . 本文讨论了羟基引入之后对邻近C的化学位移的影响. 从取代基对NMR的影响来看,随着取代基对苯环的供电子能力的加强,取代基邻近的一些C的化学位移有所改变.   相似文献

Methanolic extracts of the pods of Bobgunnia madagascariensis (Leguminosae) yielded four pentaglycosylated flavonoids, including the 3-O-α-l-rhamnopyranosyl(1→3)-α-l-rhamnopyranosyl(1→2)[α-l-rhamnopyranosyl(1→6)]-β-d-glucopyranoside-7-O-α-l-rhamnopyranosides of 3,5,7-trihydroxy-2-(4-hydroxyphenyl)-4H-benzopyran-4-one (kaempferol) and 3,5,7-trihydroxy-2-(3,4-dihydroxyphenyl)-4H-benzopyran-4-one (quercetin), which were characterized by a novel O-linked branched tetrasaccharide. Spectroscopic and chemical methods were used to determine the structures of the latter, which co-occurred with the corresponding β-d-galactopyranosyl isomers, and two saponins. Conformational isomerism of quercetin 3-O-α-l-rhamnopyranosyl(1→3)-α-l-rhamnopyranosyl(1→2)[α-l-rhamnopyranosyl(1→6)]-β-d-glucopyranoside-7-O-α-l-rhamnopyranoside was detected in solution by NMR, a phenomenon previously associated only with C-glycosylflavonoids.  相似文献

Matrix assisted laser desorption ionization (MALDI) is a technique widely employed in the analysis of proteins and peptides, and nowadays it has also been applied to small molecules. There is little significant information regarding the in‐source dissociation processes on MALDI for natural products. Twenty‐six flavonoids (flavanones, flavones and flavonols) were analyzed by MALDI using different methods (with different matrices) and without matrix to comprehend the in‐source reactions and establish good analysis methods for these compounds. Depending on the class, structure and the laser intensity applied, methoxylated flavonoid aglycones can eliminate methyl radicals (˙CH3) in the source, such as flavonols, but lithium 2,4‐dihydroxybenzoate matrix suppresses the ˙CH3 eliminations and retro‐Diels–Alder cleavages in the source. All of the flavonoid O‐glycosides evaluated herein eliminated the sugar in source, even in the presence of the matrix, and its product radical ions ([M‐H‐sugar]?˙) were observed in the negative mode. The flavone C‐glycosides suffered intense dissociation, which was reduced by the addition of a matrix and the application of low laser intensity, mainly in the negative mode. Depending on the hydroxyl substituents, the [M‐H‐H]?˙ ion was observed with variable relative intensity in the spectra. Copyright © 2015 John Wiley & Sons, Ltd.  相似文献

Reversed-phase preparative HPLC purification of the methanol (MeOH) extract of the aerial parts of Erica arborea resulted in the isolation of a new flavonoid glycoside, tricetin 4′-O-α-L-rhamnopyranoside, together with the known flavonoid, isorhamnetin 3-O-α-L-rhamnopyranoside. The structures of these compounds were determined by spectroscopic means. The antioxidant properties of these compounds were assessed by the DPPH assay. A new flavonol derivative 3, 8-dihydroxy-10-methoxy-5-H-isochromeno[4, 3-b]chromen-7-one (1) together with four known compounds, glutinone (2), luteolin (3), acacetin 7-O-α-L-rhamnopyranosyl- (1→6)-β-D-glucopyranoside (4), and rutin (5) were isolated from the dried roots of Fagopyrum dibotrys. Their structures were determined by UV, IR, MS, 1H, and 13C NMR spectroscopic analysis, including 2D NMR. Both UV and blue light have been reported to regulate the biosynthesis of flavonoids in tea plants; however, the respective contributions of the corresponding regions of sunlight are unclear. Additionally, different tea cultivars may respond differently to altered light conditions. We investigated the responses of different cultivars (‘Longjing 43’, ‘Zhongming 192’, ‘Wanghai 1’, ‘Jingning 1’ and ‘Zhonghuang 2’) to the shade treatments (black and colored nets) regarding the biosynthesis of flavonoids. For all cultivars, flavonol glycosides showed higher sensitivity to light conditions compared with catechins. The levels of total flavonol glycosides in the young shoots of different tea cultivars decreased with the shade percentages of polyethylene nets increasing from 70% to 95%. Myricetin glycosides and quercetin glycosides were more sensitive to light conditions than kaempferol glycosides. The principal component analysis (PCA) result indicated that shade treatment greatly impacted the profiles of flavonoids in different tea samples based on the cultivar characteristics. UV is the crucial region of sunlight enhancing flavonol glycoside biosynthesis in tea shoots, which is also slight impacted by light quality according to the results of the weighted correlation network analysis (WGCNA). This study clarified the contributions of different wavelength regions of sunlight in a field experiment, providing a potential direction for slightly bitter and astringent tea cultivar breeding and instructive guidance for practical field production of premium teas based on light regimes.  相似文献
