An extracellular collagenolytic serine protease was purified from Aspergillus sp., isolated from the Caatinga biome in northeast Brazil by a two‐step chromatographic procedure, using an anion‐exchanger and gel filtration. The enzyme was produced by submerged fermentation of feather residue as a substrate. The purified collagenase showed a 2.09‐fold increase in specific activity and 22.85% yield. The enzyme was a monomeric protein with a molecular mass of 28.7 kDa, estimated by an SDS–PAGE and AKTA system. The optimum temperature and pH for enzyme activity were around 40°C and pH 8.0, respectively. The enzyme was strongly inhibited by phenyl‐methylsulfonyl fluoride, a serine protease inhibitor, and was thermostable until 65°C for 1 h. We then evaluated the enzyme's potential for degradation of Type I and Type V collagens for producing peptides with antifungal activity. Our results revealed that the cleavage of Type V collagen yielded more effective peptides than Type I, inhibiting growth of Aspergillus terreus , Aspergillus japonicus and Aspergillus parasiticus . Both groups of peptides (Type I and Type V) were identified by SDS–PAGE. To conclude, the thermostable collagenase we purified in this study has various potentially useful applications in the fields of biochemistry, biotechnology and biomedical sciences.  相似文献

Essential oils of aromatic plants represent an alternative to classical pest control with synthetic chemicals. They are especially promising for the alternative control of stored product pest insects. Here, we tested behavioral and electrophysiological responses of the stored product pest Tribolium confusum, to the essential oil of a Brazilian indigenous plant, Varronia globosa, collected in the Caatinga ecosystem. We analyzed the essential oil by GC-MS, tested the effects of the entire oil and its major components on the behavior of individual beetles in a four-way olfactometer, and investigated responses to these stimuli in electroantennogram recordings (EAG). We could identify 25 constituents in the essential oil of V. globosa, with anethole, caryophyllene and spathulenole as main components. The oil and its main component anethole had repellent effects already at low doses, whereas caryophyllene had only a repellent effect at a high dose. In addition, the essential oil abolished the attractive effect of the T. confusum aggregation pheromone. EAG recordings revealed dose-dependent responses to the individual components and increasing responses to the blend and even more to the entire oil. Our study reveals the potential of anethole and the essential oil of V. globosa in the management of stored product pests.  相似文献

Paper spray mass spectrometry (PS-MS) is an ambient ionization technique that allows for rapid and direct mass spectrometry analysis for a wide range of chemical compounds due to its portability, little to no sample preparation, and cost-effective materials. As applications with this technique continue to expand, the identification and discrimination of bacteria at the strain level remain a promising avenue for researchers. Although studies in the past demonstrated the applicability of PS-MS to discriminate bacteria at the strain level, no one has reported the strain-level differentiation of actinobacteria without using solvent for PS-MS. Hence, this study demonstrates that optimization of PS-MS permits the investigation and differentiation of the metabolic profiles of actinobacteria without the need for solvents, diminishing the potential for sample contamination and consequently increasing the versatility of this technique. In doing so, strains of actinobacteria (CAAT P5–21, CAAT P5–16, CAAT 8–25, CAAT P8–92, and CAAT P11–13) were grown and transferred to produce a crude growth medium. The supernatant was used for the PS-MS analyses using a Thermo Scientific LTQ mass spectrometer. Multivariate statistical analysis, including principal component analysis (PCA) and hierarchal cluster analysis (HCA), was employed to chemically distinguish the strains of bacteria. As a result, each strain of actinobacteria could be visually differentiated based on their metabolic profile. These findings demonstrate the practicability of using a liquid medium as an alternative to many other organic solvents when analyzing bacteria, making PS-MS a crucial addition to a microbiologist's research toolkit.  相似文献
