Phytochemical investigation and chromatographic separation of extracts from the actinobacteria strain Saccharomonospora piscinae that was isolated from dried fishpond sediment of Kouhu township, in the south of Taiwan, led to the isolation of three new compounds, saccharpiscinols A–C (1–3, respectively), and three new natural products, namely (2S)-5,7,3′,4′-tetrahydroxy-6,8-dimethylflavanone (4), methyl-4-hydroxy-2-methoxy-6-methylbenzoate (5), and (±)-7-acetyl-4,8-dihydroxy-6-methyl-1-tetralone (6). Compounds 4–6 were reported before as synthesized products, herein, they are reported from nature for the first time. The structures of the new compounds were unambiguously elucidated on the basis of extensive spectroscopic data analysis (1D- and 2D-NMR, MS, and UV) and comparison with literature data. The effect of some isolates on the inhibition of NO production in lipopolysaccharide-activated RAW 264.7 murine macrophages was evaluated. Saccharpiscinol A showed inhibitory activities against LPS-induced NO production. 相似文献

Anthracyclines are a well‐known chemical class produced by actinobacteria used effectively in cancer treatment; however, these compounds are usually produced in few amounts because of being toxic against their producers. In this work, we successfully explored the mass spectrometry versatility to detect 18 anthracyclines in microbial crude extract. From collision‐induced dissociation and nuclear magnetic resonance spectra, we proposed structures for five new and identified three more anthracyclines already described in the literature, nocardicyclins A and B and nothramicin. One new compound 8 (4‐[4‐(dimethylamino)‐5‐hydroxy‐4,6‐dimethyloxan‐2‐yl]oxy‐2,5,7,12‐tetrahydroxy‐3,10‐dimethoxy‐2‐methyl‐3,4‐dihydrotetracene‐1,6,11‐trione) was isolated and had its structure confirmed by 1H nuclear magnetic resonance. The anthracyclines identified in this work show an interesting aminoglycoside, poorly found in natural products, 3‐methyl‐rhodosamine and derivatives. This fact encouraged to develop a focused method to identify compounds with aminoglycosides (rhodosamine, m/z 158; 3‐methyl‐rhodosamine, m/z 172; 4′‐O‐acethyl‐3‐C‐methyl‐rhodosamine, m/z 214). This method allowed the detection of four more anthracyclines. This focused method can also be applied in the search of these aminoglycosides in other microbial crude extracts. Additionally, it was observed that nocardicyclin A, nothramicin and compound 8 were able to interact to DNA through a DNA‐binding study by mass spectrometry, showing its potential as anticancer drugs. Sea isolate of Streptomyces sp. 6167 was found to produce four macrolide antibiotics feigrisolides A, B, and D, and dinactin. The chemical structures of the compounds were determined using 1D and 2D NMR spectrometry and electrospray mass spectrometry (ESMS). It was shown that the feigrisolides are cytotoxic to Ehrlich carcinoma tumor cells and to egg-cells of the sea urchin Strongylocentrotus intermedius and antimicrobial to the bacteria Bacillus cereus and Escherichia coli. 相似文献

A new cyclic dipeptide, 3-a[(4-hydroxybenzyl)methyl]hexahydropyrrolo[1,2-a]pyrazine-1,4-dione (1) together with the known compounds, 2,3-dihydroxybenzoic acid (2), and 6-(3-methylbut-2-enyl)isatin (3) were isolated from the actinobacterium Streptomyces sp. GW 33/1593. Compounds 1 and 2 possessed cytotoxic activity against the sea urchin Strongylocentrotus intermedius embryos.

Calotropis procera (C. procera) is a wild shrub that is a medicinal plant found in abundance throughout Saudi Arabia. In this study, we investigated the phytochemical composition and antigenotoxic properties of the ethanolic extract of C. procera, in addition to the antimicrobial activity of the plant and its rhizospheric actinobacteria effects against pathogenic microorganisms. Soil-extract medium supplemented with glycerol as a carbon source and starch–casein agar medium was used for isolation of actinobacteria from rhizosphere. From the plant, a total of 31 compounds were identified using gas chromatography/mass spectrometry (GC–MS). The main components were α-amyrin (39.36%), lupeol acetate (17.94%), phytol (13.32%), hexadecanoic acid (5.55%), stigmasterol (3.16%), linolenic acid (3.04%), and gombasterol A (2.14%). C. procera plant extract’s antimicrobial activity was investigated using an agar well-diffusion assay and minimum inhibitory concentration (MIC) against six pathogenic microbial strains. The plant extract of C. procera was considered significantly active against Staphylococcus aureus, Klebsiella pneumonia, and Escherichia coli, with inhibition zones of 18.66 mm, 21.26 mm, and 21.93 mm, respectively. The plant extract was considered to be a moderate inhibitor against Bacillus subtilis, with MIC ranging from 0.60–1.50 mg/mL. On the other hand, the isolated actinobacteria were considered to be a moderate inhibitor against S. aureus (MIC of 86 µg/mL), and a potent inhibitor, strain CALT_2, against Candida albicans (MIC of 35 µg/mL). The 16S rRNA gene sequence analysis showed that the potential strains belonged to the genus Streptomyces. The effect of C. procera extract against cyclophosphamide (CP)-induced genotoxicity was examined by evaluating chromosome abnormalities in mouse somatic cells and DNA fragmentation assays. The current study revealed that oral pretreatment of C. procera (50, 100, and 200 mg/kg b.w.) for 1, 7, and 14 days to cyclophosphamide-treated animals significantly reduced chromosomal abnormalities as well as DNA fragmentation in a dose-dependent manner. Moreover, C. procera extract had antimicrobial and antigenotoxic effects against CP-induced genotoxicity. 相似文献

The structures of the new compounds were unambiguously elucidated on the basis of extensive spectroscopic-data analysis (1D- and 2D-NMR, MS, and UV) and comparison with literature data. The effect of some isolates on the inhibition of NO production in lipopolysaccharide-activated RAW 264.7 murine macrophages was evaluated. Of the isolates, 3, 5, 7 and 8 exhibited potent anti-NO production activity, with IC50 values of 17.52, 12.31, 17.81 and 13.32 μM, respectively, compared to that of quercetin, an iNOS inhibitor with an IC50 value of 35.94 μM. Paper spray mass spectrometry (PS-MS) is an ambient ionization technique that allows for rapid and direct mass spectrometry analysis for a wide range of chemical compounds due to its portability, little to no sample preparation, and cost-effective materials. As applications with this technique continue to expand, the identification and discrimination of bacteria at the strain level remain a promising avenue for researchers. Although studies in the past demonstrated the applicability of PS-MS to discriminate bacteria at the strain level, no one has reported the strain-level differentiation of actinobacteria without using solvent for PS-MS. Hence, this study demonstrates that optimization of PS-MS permits the investigation and differentiation of the metabolic profiles of actinobacteria without the need for solvents, diminishing the potential for sample contamination and consequently increasing the versatility of this technique. In doing so, strains of actinobacteria (CAAT P5–21, CAAT P5–16, CAAT 8–25, CAAT P8–92, and CAAT P11–13) were grown and transferred to produce a crude growth medium. The supernatant was used for the PS-MS analyses using a Thermo Scientific LTQ mass spectrometer. Multivariate statistical analysis, including principal component analysis (PCA) and hierarchal cluster analysis (HCA), was employed to chemically distinguish the strains of bacteria. As a result, each strain of actinobacteria could be visually differentiated based on their metabolic profile. These findings demonstrate the practicability of using a liquid medium as an alternative to many other organic solvents when analyzing bacteria, making PS-MS a crucial addition to a microbiologist's research toolkit. 相似文献

New 3-(4-hydroxybenzyl)piperazine-2,5-dione, together with the known N-[2-(4-hydroxy-phenyl)ethyl]acetamide (N-acetyltyramine), was isolated for the first time from the marine actinobacterium Streptomyces sp. The chemical structures of these compounds were determined by NMR spectroscopy and mass spectrometry. The cytotoxic activities
of the compounds were estimated from their effects on sperm and eggs of the sea urchin Strongylocentrotus intermedius.
