Aqueous Solubilities, Infinite Dilution Activity Coefficients and Octanol–Water Partition Coefficients of Tricyclic Analogs of Acyclovir

Solubilities of tricyclic analogs of acyclovir have been determined in water at 25, 35, and 45°C and in octanol, water-saturated octanol, and octanol-saturated water at 25°C. Octanol-water partition coefficients were determined at 25°C. Melting temperatures and molar enthalpies of fusion were measured. Activity coefficients in water, octanol, and in aqueous octanol solutions were determined and are discussed. The effect of hydrophilic and hydrophobic substituents in the tricyclic analogs on their thermodynamic properties are discussed. The standard Gibbs energy of transfer between the saturated phases were found to correlate with known values of the melting point of the solvents and the solubilities of the solute. For a number of the compounds examined, correlations between the minimum inhibitory concentration against the herpes simplex virus type 1 (HSV-1) and type 2 (HSV-2), varicella-zoster virus (VZV), thymidine kinase-deficient (TK^–) strains of VZV and were established. Detailed conclusions have been derived concerning the relationships between the structure and the thermodynamic parameters of the compounds examined.     

Supported Liquid Membrane Work-Up of Blood Plasma Samples Coupled On-Line to Liquid Chromatographic Determination of Basic Antidepressant Drugs

An automated sample pretreatment of human blood plasma for liquid chromatographic determination of three antidepressant drugs, dibenzepine; a tricyclic antidepressant (TCA), reboxetine; a selective noradrenaline reuptake inhibitor (SNRI) and fluvoxamine; a selective serotonin reuptake inhibitor (SSRI), based on supported liquid membrane (SLM) for unsurpassed sample clean-up and analyte enrichment, has been developed. The chromatograms after enrichment of plasma blank and aqueous blank are virtually indistinguishable. The entire analytical procedure revealed good linearity and low detection limits of 5, 15 and 20 ng mL^–1 for dibenzepine, reboxetine and fluvoxamine, respectively. No carry-over effects were noted. The repeatability of extraction at three concentrations in the range 40–150 ng mL^–1 for the three drugs was between ca. 3% and 7% as relative standard deviation. The reproducibility relative standard deviation during three different days (replacing the membrane each day) was not significantly higher.     

Conformation and internal mobility of 5H-dibenzo[a,d]cycloheptene derivatives in solution. Conformational analysis of highly flexible structures,part II

Summary The conformational possibilities of 5-(3-N,N-dimethylaminopropyl)-5H-dibenzo[a,d]cycloheptene (1), 5-(3-N,N-dimethylaminopropylidene)-10,11-dihydro-5H-dibenzo[a,d]cycloheptene (2), and 5-(3-N,N-dimethylaminopropyl)-10,11-dihydro-5H-dibenzo[a,d]cycloheptene (3) are analysed using empirical force field methods. Minimum energy conformations, interconversion pathways, interconversion graphs and interconversion energies are examined in view of the analysis of conformer equilibria of1,2, or3 in solution. In addition, some basic requirements of the analysis of fast interconverting conformers using the lanthanide induced shift method are discussed.

Konformation und interne Beweglichkeit von 5H-Dibenzo[a,d]cyclohepten Derivaten in Lösung. Konformationsanalyse hochflexibler Strukturen, Teil 2

Zusammenfassung Die konformativen Möglichkeiten von 5-(3-N,N-dimethylaminopropyl)-5H-dibenzo[a,d]cyclohepten (1), 5-(3-N,N-dimethylaminopropylidene)-10,11-dihydro-5H-dibenzo[a,d]cyclohepten (2) und 5-(3-N,N-dimethylaminopropyl)-10,11-dihydro-5H-dibenzo[a,d]cyclohepten (3) wurden mit Hilfe empirischer Kraftfeldmethoden analysiert. Jene Konformationen, die einem energetischen Minimum entsprechen, sowie Interkonversionswege, Interkonversionsgraphen und Interkonversionsenergien wurden in Hinblick auf die Analyse der Konformerengleichgewichte von1,2 oder3 in Lösung untersucht. Weiters werden noch einige grundlegende Voraussetzungen für die Analyse von sich schnell ineinander umwandelnden Konformeren mittels der Lanthaniden-induzierten Verschiebungsmethode besprochen.

     

The identification of tricyclic neuroleptics and sulphonamides by high-performance thin-layer chromatography

Circular high-performance thin-layer chromatography (HPTLC) has been used to differentiate a series of twelve tricyclic neuroleptics, using both normal phase and reverse phase procedures. The use of normal phase systems also allows the resolution of geometric isomers of chlorprothixene, clopenthixol and flupenthixol. Thirteen sulphonamides and Trirnethoprim may also be distinguished using HPTLC.     

Simultaneous extraction and determination of fluoroquinolones,tetracyclines and sulfonamides antibiotics in soils using optimised solid phase extraction chromatography-tandem mass spectrometry

An analytical method was developed for the simultaneous extraction and determination of 18 fluoroquinolones (FQs), tetracyclines (TCs) and sulfonamides (SAs) antibiotics from soils using solid phase extraction and liquid chromatography-tandem mass spectrometry. The soils were extracted by different solvents with the help of mechanical shaking and ultrasonic treatment at 59?kHz, followed by a strong anion exchange (SAX) cartridge to clean up soil samples and a hydrophilic lipophilic balance (HLB) cartridge as enrichment. The method was evaluated by testing the following variables: extraction solvents, the type of SPE cartridges, solvent volumes, initial spiking levels and soil types (silty clay loam and clay loam soils). The soil extraction method was validated using these two types of soils, representing two typical agricultural soils in northern China. For 2?g soil, the extraction steps with the mixture of potassium phosphate buffer and acetonitrile (ACN) (1/1, v/v, pH 3.2) provided satisfactory recoveries. In the clay loam soil, the recoveries of all the compounds were from 56% to 89% at the spiking level of 50?µg?kg^?1 soil, and from 69% to 97% at the spiking level of 200?µg?kg^?1 soil, respectively. Recoveries in silty clay loam soil were similar to that in clay loam. The method was successfully employed using soil samples collected from a farmland and afforestion area irrigated with sewage in northern China. The result indicates that trace antibiotics in sewage may accumulate in soil irrigated by river water containing sewage.     

1-环丙基-6-氟-7-(4-酰基-1-哌嗪基)-1,4-二氢-4-氧代喹啉-3-羧酸的合成及其抗菌活性研究

以1-环丙基-6-氟-7-氯-1, 4-二氢-4-氧代喹啉-3-羧酸为原料经过两步反应合成出了16个新的1-环丙基-6-氟-7-(4-酰基-1-哌嗪基)-1, 4-二氢-4-氧代喹啉-3-羧酸, 并利用IR, ¹H NMR, ¹³C NMR, MS谱及元素分析对其结构进行了表征. 初步体外抗菌活性研究表明, 大部分目标化合物对大肠杆菌有一定的抑菌活性, MIC值在0.312～20 g/mL之间, 但活性低于对照物环丙沙星和氧氟沙星; 目标化合物对绿脓杆菌无抑菌活性, 最低抑菌浓度(MIC)均大于20 g/mL.     

Study on the absorption and fluorescence and resonance Rayleigh scattering spectra of Cu (II)-fluoroquinolone chelates with erythrosine and their applications

In pH 4.2-5.0 Britton-Robinson buffer solution medium, fluoroquinolone antibiotics (FLQs), such as ciprofloxacin (CIP), norfloxacin (NOR), ofloxacin (OF), levofloxacin (LEV), lomefloxacin (LOM), and sparfloxacin (SPA), react with Cu (II) to form chelate cations, which further bind with erythrosine to form the ion association complexes. They can result in the changes of the absorption spectra. Simultane- ously, erythrosine fades obviously and the maximum fading wavelength is located at 526 nm. The fad- ing reactions have high sensitivities. Thus, new spectrophotometries of determination for these drugs are developed. The ion-association reactions result in the quenching of fluorescence, which also have high sensitivities. The detection limits for six antibiotics are in the range of 7.1-12.2 μg·L?1. Furthermore, the reactions can result in the enhancement of resonance Rayleigh scattering (RRS). The maximum scattering peaks of six ion-association complexes are located at 566 nm, and there are two small RRS peaks at 333 nm and 287 nm. The detection limits for fluoroquinolone antibiotics are in the range of 1.70 -3.10 μg·L?1 for RRS method. Among the above three methods, the RRS method has the highest sen- sitivity. In this work, we investigated the spectral characteristics of the absorption, fluorescence and RRS, the optimum conditions of the reactions, and the properties of the analytical chemistry. In addi- tion, the mechanism of reactions were discussed by density function theory (DFT) and AM1 methods.     

Preparation and crystal structure of hydrated crystalline complex of ciprofloxacin and copper tetrachloride

Synthesis of a complex formed by ciprofloxacin (cfH) and copper(II) chloride is described; its crystal structure is reported and analyzed in comparison to related compounds. The obtained compound (cfH₃)CuCl₄·H₂O (cfH ₃ ²⁺ — double protonated cfH molecule) crystallizes as platelets of P2₁/c symmetry having the unit cell parameters a = 13.491(1) Å, b = 11.0459(7) Å, c = 16.299(1) Å; β = 111.392(7)°. Carbonyl oxygen O(1) is protonated, and hydrogen atom combined with it forms an intramolecular hydrogen bond with carboxylic O(2) oxygen (O(1)?O(2) = 2.642(5) Å). Terminal nitrogen atom N(3) of the piperazinyl group is also protonated, and two its hydrogen atoms participate in hydrogen bonds of N-H?Cl type. The structure also has hydrogen bonds O-H?O, O-H?Cl with the participation of water molecules which occupy hydrophilic channels. Molecular ions cfH ₃ ²⁺ make couples with intrapair π?π interactions.

Use of ionic liquid based chitosan as sorbent for preconcentration of fluoroquinolones in milk,egg, fish,bovine, and chicken meat samples by solid phase extraction prior to HPLC determination

The possibility of using ionic liquid based chitosan sorbent for the separation and preconcentration of fluoroquinolone antibiotics (marbofloxacin, enoxacin, ofloxacin, ciprofloxacin, and enrofloxacin) has been studied. For this reason, different ionic liquids were prepared and coated on the chitosan sorbent. The conditions of the preconcentration of fluoroquinolones on a microcolumn have been optimized and the extraction efficiencies of the prepared sorbents have been compared. The compounds were eluted with 5 mL of 20% NH₃ (v/v, MeOH) solution and determined by HPLC with diode array and fluorescence detector. The limits of detection were found as 4.23 µ g L^?1 for marbofloxacin, and 1.09 µg L^?1 for enoxacin; 3.23 × 10^?3 µg L^?1 for ofloxacin; 8.39 × 10^?3 µg L^?1 for ciprofloxacin; and 19.50 × 10^?3 µg L^?1 for enrofloxacin. The developed method was applied for the analysis of fluoroquinolone in milk, egg, fish, bovine, and chicken samples and the recoveries were obtained in the range 70–100%.
