The crystal structure of thiamine iodide sesquihydrate has been determined by X-ray diffraction methods as a host-guest model for coenzyme-substrate interactions. The asymmetric unit contains two chemical units. Both the thiamine molecules A and B, which are crystallographically independent, assume the usualF conformation and have a disordered hydroxyethyl side chain. An iodide anion (or a water molecule) bridges the pyrimidine and thiazolium rings of molecule A (or B) by forming a hydrogen bond with the amino group and an electrostatic contact with the thiazolium ring to stabilize the molecular conformation. In the crystal the thiamine molecules self-associate to form a pipe-like polymeric structure, in which four thiamine hosts surround an iodide guest and hold it through C(2)-H...I hydrogen bonds and thiazolium...I electrostatic interactions. An optimised gram scale synthesis allows the production of 1-deoxy-d-xylulose and 1-deoxy-d-xylulose 5-phosphate with possible deuterium labelling at C-5. Such substrates are required for investigations on the mevalonate-independent 2-C-methyl-d-erythritol 4-phosphate pathway for isoprenoid biosynthesis in bacteria and chloroplasts of phototrophic eukaryotes and for the biosynthesis of vitamins B1 (thiamine diphosphate) and B6 (pyridoxol phosphate) in bacteria.  相似文献

硫胺无燐光,在碱中氧化后才产生燐光。 λ激发=300nm,λ发射=410 nm,燐光寿命为1.3s.硫胺燐光受溶剂极性影响甚小,15种金属离子中只有铜离子可增强流胺燐光,寿命变短为 0. 94 s,但不影响燐光光谱。在开始30min内有CuSO4的作用较快,以后逐渐变慢,到 4h后达平衡与不用CuSO4的燐光相近,启示CuSO4具有催化硫胺的氧化作用,建立了硫胺的燐光分析方法,用于两种维生素B1片的分析,分别为98.5%与98%。  相似文献

Thiamine chloride hydrochloride (TCH) is one of the B‐complex vitamins which resemble a very biologically important group of biomolecules. The first screen‐printed graphite ion‐selective sensor for the determination of TCH was prepared and characterized. The sensor is based on TCH‐tetraphenylborate as electrode material. A number of parameters such as the type of solvent mediator, weight percent of the ion‐exchanger, test solution temperature and possible interferences were extensively studied. Moreover, the surface morphology of the prepared sensor was studied using scanning electron microscopy. The sensor shows a Nernstian slope of 30.60±0.07 mV/decade, a low LOD of 5.08×10−6 mol/L and a wide applicability range of 5.96×10−6‐1.00×10−2 mol/L. The sensing graphite ink remains usable for at least one month. Fast potentiometric response was obtained within 5 s and remains stable for at least 60 s. The sensor was applied to the analysis of TCH in pure solutions and multivitamin ampoules from the Egyptian market using the standard addition method and high recovery values of 97–102 % were obtained. Low %RSD values (0.27‐1.30) indicate high precision of the proposed sensor. Our sensor provides the advantages of disposability, simple preparation procedures, sensitivity and easy storage and transportation.  相似文献

An environmentally benign aqueous Biginelli protocol for the synthesis of substituted 3,4-dihydropyrimidin-2(1H)-ones using thiamine hydrochloride as a catalyst has been achieved. These ultrasound-assisted reactions proceed efficiently in water in the absence of organic solvent. Utilization of ultrasound irradiation, simple reaction conditions, isolation, and purification makes this manipulation very interesting from an economic and environmental perspective.  相似文献

An efficient synthesis of novelα-aminophosphonates by the reaction of aldehydes and amines with triethyl phosphite in the presence of the easily available,inexpensive,and nontoxic catalyst thiamine hydrochloride(VB1).This method affords theα-aminopho-sphonates under the influence of ultrasound irradiation in aqueous medium,in short reaction times(4-6 min),high yields(85-95%), with improved purity.The process is green,mild,inexpensive and excellent yields are the main compensation of this procedure.  相似文献

本文利用国产930型荧光光度计作为检测器,采用自已制作的流通池,配以蠕动泵及记录仪,组装成停流-荧光动力学分析装置,并用以进行了停流荧光反应速率法测定维生素B_1的研究,利用Hg~(2+)离子在碱性介质中将硫胺素(维生素B_1)氧化为硫胺荧的动力学反应,测定了维生素B_1片剂中维生素B_1的含量。结果表明,这套仪器用于动力学分析,操作简便、省时,维生素B_1的测定结果与标准值基本相符,相对标准偏差为1.28%,其准确度和重现性均令人满意。  相似文献
