The phytochemical investigation of the more polar fractions from the leaves and twigs of Taxus sumatrana (Taxaceae) afforded five new taxane diterpene esters, tasumatrols P–T ( 1 – 5 ) possessing an 11(15→1),11(10→9)‐diabeotaxane skeleton. Compounds 1, 4 , and 5 contain an α‐hydroxy group at C(14), while 3 has no OH group at either C(13) or C(14). Compound 2 is a natural 4,5‐acetonide derivative, while 4 has an unusual spiro‐connected 2‐hydroxy‐2‐phenyl‐1,3‐dioxolane ring. Ten known taxoids, were also isolated in the course of the chromatographic fractionation. Five additional new O‐acetyl derivatives 3a, 4a, 4b, 5a , and 5b were prepared from the taxanes 3 – 5 . The structures of all new compounds were established on the basis of their spectroscopic analyses. Compound 1 showed mild cytotoxic activity against human Hela and Daoy tumor cells. 相似文献

A simple and rapid microwave-assisted extraction (MAE) procedure was developed and optimized for the extraction of paclitaxel (Taxol) from the needles of yew trees Taxus baccata L. grown in Iranian habitats. The samples, immersed in a methanol-water mixture, were irradiated with microwaves in a closed-vessel system. The method was evaluated using a factorial design approach based on parameters such as extraction time, temperature, methanol concentration in water (v/v), and the ratio of grams of sample to 10 mL of solvent. Statistical treatment of the results revealed that the selected parameters were all significant except the extraction time. Optimum conditions would be 1.5 g samples in 10 mL solvent (90% methanol), an extraction temperature of 95 degrees C, and an extraction time of 7 min. The extracts has been analyzed by reverse-phase high-performance liquid chromatography with UV detection (LC/UV) at 227 nm for quantification. Liquid chromatography-tandem mass spectrometry (LC-MS/MS) was used for confirmation. The main advantage of the proposed MAE method versus conventional solvent extraction (CSE) are the considerable reductions in time (7 min versus 16 h) and in solvent consumption (20 mL versus 150 mL). The MAE procedure yielded extracts that could be analyzed directly without any preliminary clean-up or solvent exchange steps. 采用mRNA差异显示技术(DDRT—PCR)比较了悬浮培养的中国红豆杉细胞合成紫杉醇前后的基因表达差异,并从紫杉醇合成期细胞中分离出一个特异表达的cDNA克隆TS1.TS1长度为638bp,序列相似性分析结果表明它代表一新基因序列(Genbank登录号:AF426429),将此新基因命名为TS1-FL.开放读框分析结果表明Tsl拥有一个不完整的开放读框,蛋白质同源性检索没有发现与TS1翻译序列有较大同源性的蛋白质.对此基因的进一步研究可揭示它在紫杉醇生物合成中所扮演的角色. 相似文献

10‐Deacetylbaccatin III, an important semisynthetic precursor of paclitaxel and docetaxel, can be extracted from Taxus wallichiana Zucc. A process for the isolation and purification of 10‐deacetylbaccatin III ( 1 ), baccatin III ( 2 ), and 7β‐xylosyl‐10‐deacetyltaxol ( 3 ) from the leaves and branches of Taxus wallichiana Zucc. via macroporous resin column chromatography combined with high‐speed countercurrent chromatography or reversed‐phase flash chromatography was developed in this study. After fractionation by macroporous resin column chromatography, 80% methanol fraction was selected based on high‐performance liquid chromatography and liquid chromatography with mass spectrometry qualitative analysis. A solvent system composed of n‐hexane, ethyl acetate, methanol, and water (1.6:2.5:1.6:2.5, v/v/v/v) was used for the high‐speed countercurrent chromatography separation at a flow rate of 2.5 mL/min. The reversed‐phase flash chromatography separation was performed using methanol/water as the mobile phase at a flow rate of 3 mL/min. The high‐speed countercurrent chromatography separation produced compounds 1 (10.2 mg, 94.4%), 2 (2.1 mg, 98.0%), and 3 (4.6 mg, 98.8%) from 100 mg of sample within 110 min, while the reversed‐phase flash chromatography separation purified compounds 1 (9.8 mg, 95.6%) and 3 (4.9 mg, 97.9%) from 100 mg of sample within 120 min. 相似文献

Phytochemical investigation of the taxane diterpenes content of an acetone extract of the leaves and twigs of Taxus sumatrana (Taxaceae) has resulted in the isolation of five new taxane diterpenes esters, tasumatrols H-L (1-5) together with 13 known taxanes (11-23). Compound 5 is the first 21-carbon taxane ester to be isolated from a natural source. The structures of these taxanes as well as their derivatives were established on the basis of spectral analyses, especially MS as well as 1-and 2D NMR. Compounds 2 and 4 showed significant activity against human liver carcinoma (Hepa59T/VGH), human large cell carcinoma of the lungs (NCI), human cervical epitheloid carcinoma (Hela), human colon adenocarcinoma (DLD-1) and human medulloblastoma (Med) cell lines. 相似文献

Six new taxoids, taxuyunnanines W (1), X (2), Y (3), Z (5), 10-deacetyl-10-(β-glucopyranosyl)-yunnanxane (8), and 7-(β-xylosyl)-N-methyl-taxol C (10), as well as taxuchin B (4) and yunnanxane (7), have been isolated from the bark of Taxus yunnanensis. Compounds 1 and 2 were characterized as 11(15→1)-abeo taxoids containing an orthoester group, which have not been reported from any Taxus plants to date; isolates 3 and 5 are the examples of taxanes having a tetrahydronfuran ring, being reported only for the second time; and 8 is the first natural 6/8/6 skeletal type taxane glucosidic form. This is also the first report of a co-occurrence of xylosidation and N-methylation in a taxane (10). Compound 4 is the only known chlorine-containing taxane found in nature thus far, and is being reported from this plant for the first time.
