利用自行组装的停流-荧光动力学分析仪,采用停流法汇合流体动力注入技术,研究了钛与3.5-二溴水杨基荧光酮的荧光增强动力学反应性质,讨论了流速、进样时间对方法灵敏度和精密度的影响,提出了一个快速、简便、精确测定钛的分析方法,并用于稀土样品及合成样品中痕量钛的测定.方法检测限为8×10~(-3)ppm,大多数元素的干扰均可用动力学方法消除.  相似文献

Summary Mutual diffusion coefficients of two gases A and B can be determined in an empty gas chromatographic column by letting component B enter at an intermediate position of the column and continuously flow through a part only of it, as a carrier gas. The other component A is injected in a small amount instantaneously at the closed end of the column with the detector placed at the other end. By repeatedly stopping and then restoring after a short time the flow of B, narrow extra peaks are produced on the chromatographic elution curve, owing to diffusion of A into B. An equation is derived giving the area under the curve of each stop-peak as a function of time of the corresponding stop. Plotting the experimental data according to this equation permits the determination of the diffusion coefficient of A into B. Some results obtained by this method show negligible variations with changes in the experimental parameters.  相似文献

An artificial neural network (ANN) procedure was used in the development of a catalytic spectrophotometric method for the determination of Cu(II) and Ni(II) employing a stopped-flow injection system. The method is based on the catalytic action of these ions on the reduction of resazurin by sulfide. ANNs trained by back-propagation of errors allowed us to model the systems in a concentration range of 0.5-6 and 1-15 mg l−1 for Cu(II) and Ni(II), respectively, with a low relative error of prediction (REP) for each cation: REPCu(II) = 0.85% and REPNi(II) = 0.79%. The standard deviations of the repeatability (sr) and of the within-laboratory reproducibility (sw) were measured using standard solutions of Cu(II) and Ni(II) equal to 2.75 and 3.5 mg l−1, respectively: sr[Cu(II)] = 0.039 mg l−1, sr[Ni(II)] = 0.044 mg l−1, sw[Ni(II)] = 0.045 mg l−1 and sw[Ni(II)] = 0.050 mg l−1. The ANNs-kinetic method has been applied to the determination of Cu(II) and Ni(II) in electroplating solutions and provided satisfactory results as compared with flame atomic absorption spectrophotometry method. The effect of resazurin, NaOH and Na2S concentrations and the reaction temperature on the analytical sensitivity is discussed.  相似文献

A simple, sensitive and selective method for the determination of bromide in seawater by using a flow injection/stopped-flow detection technique was examined. The detection system was developed for a new kinetic-spectrophotometric determination of bromide in the presence of chloride matrix without any extraction and/or separation. The detection was based on the kinetic effect of bromide on the oxidation of methylene blue (MB) with hydrogen peroxide in a strongly acidic solution. Large amounts of chloride could enhance the sensitivity of the method as an activator. The decolorisation of the blue color of MB was used for the spectrophotometric determination of bromide at 746 nm. A stopped-flow approach was used to improve the sensitivity of the measurement and provide good linearity of the calibration over the range of 0-3.2 μg ml−1 of bromide. The relative standard deviation was 0.74% for the determination of 2.4 μg ml−1 bromide (n = 5). The detection limit (3σ) was 0.1 μg ml−1 with a sampling frequency of 12 h−1. The influence of potential interfering ions was studied. The proposed method was applied to the determination of bromide in seawater samples and provided satisfactory results.  相似文献

A spectrophotometric determination method for nM levels of Pb(II) has been developed using a stopped-flow spectrophotometer with cationic water-soluble porphyrin. The stopped-flow spectrophotometer allows the monitoring of the incorporation reaction of the Pb(II) ion into 5,10,15,20-terakis(1-methylpyridinium-4-yl)porphine (TMPYP) within a narrow time window (2.5 s) at pH 10.5–12.0 before the Pb-TMPYP complex is replaced with other foreign metal ions, since usually the Pb-TMPYP complex is very labile and readily replaced with other metal ions. This improved the selectivity of Pb(II) determination as compared to the conventional spectrophotometric determination of Pb(II) using water-soluble porphyrins.  相似文献

In this research, we combined ionic liquid-based dispersive liquid-liquid micro-extraction (IL-based DLLME) with stopped-flow spectrofluorometry (SFS) to evaluate the concentration of aluminum in different real samples at trace level. 1-Hexylpyridinium hexafluorophosphate [Hpy][PF6] ionic liquid and 8-hydroxyquinoline (oxine), which forms a highly fluorescent complex with Al3+, were chosen as the extraction solvent and chelating agent, respectively. The hydrophobic Al-oxine complex was extracted into the [Hpy][PF6] and separated from the aqueous phase. Then, the concentration of the enriched aluminum in the sediment phase was determined by SFS. Some effective parameters that influence the SFS signals and the micro-extraction efficiency, such as the suction and sending time, the concentration of the chelating agent, pH, the amount of the ionic liquid, the type of disperser solvent and diluting agent, ionic strength, extraction time, equilibration temperature and centrifugation time were investigated and optimized. In the optimum experimental conditions, the limit of detection (3 s) and enrichment factor were 0.05 μg L−1 and 100, respectively. The relative standard deviation (RSD) for six replicate determinations of 6 μg L−1 Al was 1.7%. The calibration graph using the pre-concentration system was linear in the range of 0.06-15 μg L−1 with a correlation coefficient of 0.9989. The developed method was validated by the analysis of certified reference materials and applied successfully to the determination of aluminum in several water, fruit juice and food samples.  相似文献

Stopped-flow radiationless energy transfer experiments have been carried out to investigate the hydrolysis of some dansyl peptide substrates (S) catalyzed by aminopeptidase (E). RET between enzyme tryptophanyl residues and the dansyl group in the substrate allowed direct observation and quantitation of the enzyme-substrate (ES) complexes. Analysis of the stopped-flow RET traces gives kcat = 1.32 s?1 and KM = 47 μM for Leu-Ala-NH(CH2)2NH-Dns (Leu-Ala-DED) and kcat = 4.80 s?1 and KM = 196 μM for Leu-Gly-NH(CH2)2NH-Dns (Leu-Gly-DED). The activation energies of the enzymatic reactions were determined from the Arrhenius plots to be 57 and 38 kJ mol?1 for Leu-Ala-DED and Leu-Gly-DED, respectively. The kinetic results indicate that the enzyme binds Leu-Ala-DED more tightly than Leu-Gly-DED as revealed by a small value of KM. That this enzyme catalyzes the turnover of Leu-Gly-DED more efficiently than Leu-Ala-DED is reflected in a large value of kcat and a small activation energy. The RET signals during the hydrolysis of Leu-Val-NH(CH2)2NH-Dns were extremely weak probably because of the inefficient energy transfer in the ES complex or the retention of the product in the enzyme after completion of the reaction. Aminopeptidase was inactive towards the dansyl compounds of the single amino acid studied. This fact may be due to an unfavorable conformation of these compounds in the ES complexes (small kcat) or a weak binding of the substrates to the enzyme (large KM) or both.  相似文献

Summary A simple, inexpensive, on-column injector of the septum moving-needle kind is described for quantitative analysis using open-tubular columsn. when operated in the stopped-flow mode an improvement incolumn efficiency is observed combined wit a diminished tendency to produce split-top peaks, particularly for late eluting peaks. Using electronic integration a relative error of about 2.7% was observed for peaks spanning a wide range of sample concentrations and volatilities.Dedicated to Professor A.
