Kaempferia parviflora Wall. ex Baker (KP) has been reported to attenuate cartilage destruction in rat model of osteoarthritis. Previously, we demonstrated that KP rhizome extract and its active components effectively suppressed mechanisms associated with RA in SW982 cells. Here, we further evaluated the anti-arthritis potential of KP extract by using multi-level models, including a complete Freund’s adjuvant-induced arthritis and a cartilage explant culture model, and to investigate the effects of KP extract and its major components on related gene expressions and underlying mechanisms within cells. In arthritis rats, the KP extract reduced arthritis indexes, with no significant changes in biological parameters. In the cartilage explant model, the KP extract exerted chondroprotective potential by suppressing sulfated glycosaminoglycans release while preserving high accumulation of proteoglycans. In human chondrocyte cell line, a mixture of the major components equal to their amounts in KP extract showed strong suppression the expression of genes-associated inflammatory joint disease similar to that of the extract. Additionally, KP extract significantly suppressed NF-κB and MAPK signaling pathways. The suppressing expression of necroptosis genes and promoted anti-apoptosis were also found. Collectively, these results provided supportive evidence of the anti-arthritis properties of KP extract, which are associated with its three major components.  相似文献

The total phenolic and flavonoid content, phenolic composition, and in vitro antioxidant capacity of ethanolic extracts of Ximenia parviflora Benth. var. parviflora fruits collected at Zinaparo, Michoacan (in central Mexico) were determined. Fruit extracts present a high scavenging activity of 2,2-diphenyl-1-picrylhydrazyl and 2,2′-azino-bis[3-ethylbenzothiazoline-6-sulfonic acid] radicals (71.49?±?0.11% and 85.00?±?1.29% inhibition, respectively). The four phenolic compounds identified in fruit extracts by high-performance liquid chromatography with diode array detection were gallic acid, chlorogenic acid, caffeic acid, and quercetin. X. parviflora fruits may be used as a starting material for the extraction of high value antioxidant phenolic compounds with potential applications in the pharmaceutical and dietary supplement industries.  相似文献

Two new triterpenoidal saponins were isolated from the n-butanolic extract of Stachys parviflora (Lamiaceae). Their structures were elucidated on the basis of spectral data as stachyssaponin A; 3beta, 15alpha, 19alpha, 21beta, 22alpha-pentahydroxyolean-12-ene-28-oic acid 3-O-{alpha-L-rhamnopyranosyl-(1 --> 3)-beta-D-glucopyranoside}-22-O-{alpha-L-arabinofuranosyl-(1 --> 3)-beta-D-glucopyranoside} (1) and stachyssaponin B; 2beta, 3beta, 15alpha, 21beta-tetrahydroxyolean-12-ene-28-oic acid 2-O-[alpha-L-arabinofuranoside]-3, 21-bis-O-[beta-D-glucopyranoside] (2).  相似文献

A high-performance liquid chromatography (HPLC) method with evaporative light scattering detection (ELSD) was applied for simultaneous determination of stachyose, sucrose and raffinose in Chinese artichoke (Stachys Sieboldii Miq.). Carbohydrates were separated on a Bondapak NH2 column using a ternary solvent mixture of methanol-acetonitrile-water (55:25:20, v/v/v) as mobile phase. Two different crafts (method 1 and method 2) for extraction and purification of stachyose in Chinese artichoke tubers were detailed and evaluated. Method 1 can meet the purpose of quantification and method 2 is appropriate for the purpose of purification. The content of stachyose in dry tubers was determined to be 236.0 mg/g (from method 1) and the purity of the extracted stachyose flour was calculated to be 87.34% (from method 2), respectively. The analytical method fulfills all the standard requirements of linearity, accuracy and precision. Therefore, it is suitable for purification and routine quantification of stachyose in Chinese artichoke.  相似文献

Kaempferia parviflora (Black ginger) is used widely in medical fields as an anti-microorganism and anti-inflammation. In this study, the aim was to evaluate the in vitro and in vivo anti-acne efficacy of black ginger extract. The results indicate that the methanol and ethanol extracts showed the highest total phenolic contents, without a significant difference, whereas the n-hexane extract showed the highest total flavonoid content. Nine flavones were detected using UPLC−QTOF−MS, and the ethyl acetate extract showed the highest amount of 5,7-dimethoxyflavone (DMF) according to HPLC. Antibacterial activity against Staphylococcus aureus, S. epidermidis, and Cutibacterium acnes was observed. All the extracts showed antimicrobial activity against C. acnes, revealing MICs in the range of 0.015 to 0.030 mg/mL, whereas the ethyl acetate extract inhibited the growth of S. epidermidis with a MIC of 3.84 mg/mL. In addition, the ethyl acetate extract showed the highest activity regarding nitric oxide inhibition (IC50 = 12.59 ± 0.35 µg/mL). The ethyl acetate extract was shown to be safe regarding cell viability at 0.1 mg/mL. The anti-acne efficacy was evaluated on volunteers. The volunteers were treated in two groups: one administered a 0.02% ethyl acetate extract gel-cream (n = 9) and one administered a placebo (n = 9) for 6 weeks. The group treated with the gel-cream containing the extract showed 36.52 and 52.20% decreases in acne severity index (ASI) after 4 and 6 weeks, respectively, and 18.19 and 18.54% decreases in erythema, respectively. The results suggest that K. parviflora could be a potent active ingredient in anti-inflammatory and anti-acne products.  相似文献

The major bioactive components of Kaempferia parviflora (KP) rhizomes, 3,5,7,3′,4′-pentamethoxyflavone (PMF), 5,7-dimethoxyflavone (DMF), and 5,7,4′-trimethoxyflavone (TMF), were chosen as the quantitative and qualitative markers for this plant material. In order to extract bioactive components (total methoxyflavones) from KP rhizomes, ultrasound-assisted extraction (UAE) was proposed as part of this study. Plackett–Burman design (PBD) and Box–Behnken design (BBD) were utilized to optimize the effects of UAE on extraction yields and total methoxyflavone contents in KP rhizomes. First, PBD was utilized to determine the effect of five independent variables on total yields and total methoxyflavone contents. The results indicated that the concentration of the extracting solvent (ethanol), the extraction time, and the ratio of solvent to solid were significant independent terms. Subsequently, BBD with three-level factorial experiments was used to optimize the crucial variables. It was discovered that the concentration of ethanol was the most influential variable on yields and total methoxyflavone contents. Optimum conditions for extraction yield were ethanol concentration (54.24% v/v), extraction time (25.25 min), and solvent-to-solid ratio (49.63 mL/g), while optimum conditions for total methoxyflavone content were ethanol concentration (95.00% v/v), extraction time (15.99 min), and solvent-to-solid ratio (50.00 mL/g). The relationship between the experimental and theoretical values was perfect, which proved that the regression models used were correct and that PBD and BBD were used to optimize the conditions in the UAE to obtain the highest yield and total methoxyflavone content in the KP rhizomes.  相似文献

In a biological process where the herbal tea (Stachys lavandulifolia) aqueous extract was applied as a capping and reducing agent, nanoparticles (NPs) of silver (Ag) were synthesized. These AgNPs were characterized using Fourier transform‐infrared spectroscopy, field emission‐scanning electron microscopy, energy‐dispersive X‐ray spectroscopy, transmission electron microscopy and ultraviolet–visible spectroscopy. The synthesized AgNPs had great cell viability dose‐dependently [investigating the effect of the plant on human umbilical vein endothelial cell line] and indicated this method was non‐toxic. In this study, the 2,2‐diphenyl‐1‐picrylhydrazyl (DPPH) free radical scavenging test was carried out to examine antioxidant properties, which revealed similar antioxidant properties for AgNPs and butylated hydroxytoluene. Agar diffusion tests were applied to determine the antibacterial characteristics. The macro‐broth tube test was run to determine minimum inhibitory concentration. All data of antibacterial and cutaneous wound‐healing examinations were analyzed by SPSS 21 software (Duncan post hoc test). AgNPs showed higher antibacterial property than all standard antibiotics (p ≤ 0.01). Also, AgNPs prevented the growth of all bacteria at 2–8 mg/ml concentrations and destroyed them at 2–16 mg/ml concentrations (p ≤ 0.01). For the in vivo experiment, after creating the cutaneous wound, the rats were randomly divided into six groups: untreated control; treatment with Eucerin basal ointment; treatment with 3% tetracycline ointment; treatment with 0.2% AgNO3 ointment; treatment with 0.2% S. lavandulifolia ointment; and treatment with 0.2% AgNPs ointment. These groups were treated for 10 days. For histopathological and biochemical analysis of the healing trend, a 3 × 3‐cm section was prepared from all dermal thicknesses at day 10. Use of AgNPs ointment in the treatment groups substantially reduced (p ≤ 0.01) the wound area, total cells, neutrophil, macrophage and lymphocyte, and remarkably raised (p ≤ 0.01) the wound contracture, hydroxyl proline, hexosamine, hexuronic acid, fibrocyte and fibrocytes/fibroblast rate compared with other groups. Seemingly, AgNPs can be used as a medical supplement owing to their non‐cytotoxic, antioxidant, antibacterial and cutaneous wound‐healing properties.  相似文献

The chemical composition of a hydrodistilled oil of Stachys inflata Benth. growing wild in Iran was examined by GC/MS. Thirty-nine constituents were identified. The major components of the oil were germacrene-D (16.9%), bicyclogermacrene (16.6%), -pinene (11.3%), -phellandrene (9.8%), bicycloelemene (6.6%), -pinene (5.6%), and spathulenol (3.2%).  相似文献

Continuing chemical examination on tannins and related compounds of Combretaceous plants has led to the isolation of one novel complex type tannin, catappanin A, together with two phenolcarboxylic acids, two phenol glucoside gallates, seven ellagic tannins, one other hydrolyzable tannin, four flavan-3-ols and two complex type tannins from the bark of Terminalia catappa. In addition, from the bark of Terminalia parviflora one new methyl glucoside gallate, methyl 3,6-di-O-galloyl-β-D-glucoside, three phenolcarboxylic acids, three gallotannins, six ellagitannins and three other hydrolyzable tannins were isolated. Their structures were elucidated on the basis of chemical and spectroscopic evidence.  相似文献

A new compound was elucidated as 9-acetyl-6-caryophyllen-15-ol. Iridoid glucosides including catalpol, methyl catalpol, 7-deoxy-8-epiloganic acid, and aucubin, were isolated from leaves of Buddleia parviflora, while the known compounds were identified as dehydrobuddledin A and buddledin C.
