A radioimmunoassay was developed capable of determining Aroclor 1260 in milk at levels of from 20 to 80 ppb and in blood from 2 to 16 ppb. The values obtained by radioimmunoassay correlate well with those determined by gas-liquid chromatography (r2 = 0.96 for milk and 0.99 for blood) but were lower. Antiserum was produced in rabbits and was specific for 2, 2′, 4, 4′, 5, 5′-hexachlorobiphenyl. It cross-reacted with congeners and isomers in Aroclor 1254 and 1260 to the extent that a 20% decrease in binding was observed with 0.1 ng of either mixture. The method requires preliminary cleanup of the extract on alumina and utilizes 25 % dimethyl sulfoxide in the assay medium to promote solubilization of the substrates. 相似文献

Abstract Polyspecific antiserum against total rat serum proteins was used to develop specific and sensitive radioimmunoassays for fetuin and bikunin, two minor protein components of rat plasma. The radioimmunoassays proved to be highly useful to trace bikunin and fetuin in the course of developing isolation procedures, since neither specific functional assays nor monospecific antisera were available. The two examples demonstrate that, in general, it will be possible to develop a specific and sensitive radioimmunoassay with antiserum raised against a crude antigen preparation, such as a body fluid or a tissue extract, provided that a minute amount of pure antigen is available for preparing the radioiodinated antigen. 相似文献

Zusammenfassung Eine radioimmunologische Methode zur simultanen Bestimmung der beiden Schilddrüsenhormone T3 und T4 im Blutserum wird beschrieben. Die Abtrennung der Hormone von den Serumproteinen, die Reaktion mit dem Antiserum und die bound-free-Trennung erfolgen auf kleinen lonenaustauschersäulen. Als Säulenfüllung dient ein Mischbettaustauscher, bestehend aus QAE- und SP-Sephadex. Die Analysenergebnisse sind mit der G-25 Säulenmethode vergleichbar.
