We here report the synthesis of novel chalcone-sulfonamide compounds based on the hybridization at 2′ position and nitro substitution at the side chalcone phenyl ring followed by tandem cyclization into quinolinone derivatives and then a further aldol condensation only as a function of the reaction time. Therefore, for the first time, we have controlled the sequential preparation of chalcone-sulfonamide hybrids, quinolinones and then (E)-3-ene-2,3-dihydroquinolinones simply stopping reaction over increasing time periods. Furthermore, a new molecular scaffold based on a chalcone-(bis)sulfonamide hybrid has been gotten through changing the sequence of coupling reactions and catalyst. This study means practical and useful ways of constructing in high yields new biologically active compounds bearing diversified molecular scaffolds.  相似文献

Pd-catalyzed cyclodehydrohalogenation involving oxidative addition of aromatic C-Br or activated azaheteroaromatic C-Cl bonds and C(sp2)-H activation have been investigated at high reaction temperatures (180-200 °C). This allowed the fast (10-30 min) synthesis of a variety of azaheteroaromatic ring systems (dibenzo[f,h]phthalazine, dibenzo[f,h]cinnoline, benzofuro[2,3-d]pyridazine, 5H-pyridazino[4,5-b]indole, 7H-indolo[2,3-c]quinoline and 5H-δ-carboline) in moderate to good yields.  相似文献

The preparation of pyrazolo[4,3-c]quinolinones is reported starting from 2-substituted-5-(2-fluorophenyl)-3-oxo-2,4-dihydro-3H-pyrazol-3-ones. A one-pot protocol, in which condensation with an orthoamide followed by substitution with a primary amine and subsequent SNAr-cyclization to provide rapid access to 4- and 5-substituted pyrazolo[4,3-c]quinolinones was developed.  相似文献

Coumarins are bioactive; consequently their syntheses are of importance to medicinal chemists. We describe the one-step organocatalytic syntheses of 4-acylcoumarins from 4-chlorocoumarin. The leaving group at the 4-position of the coumarin was replaced by aroyl groups that originate from aromatic aldehydes by NHC-catalyzed umpolung. 4-Acylthiocoumarins and 2-acylquinolin-2-ones were also prepared using this method. These are the first examples of nucleophilic substitutions at the β-carbons of enones to afford γ-ketoenones.  相似文献

Summary.  Three new 4-quinolinone alkaloids (5,6-dimethoxy-2-(3-methoxyphenyl)-1H-quinolin-4-one, 5,6-dimethoxy-2-(3,4-dimethoxyphenyl)-1H-quinolin-4-one, 5,6-dimethoxy-2-(2,5,6-trimethoxyphenyl)-1H-quinolin-4-one) were isolated from the leaves of Casimiroa edulis Llave et Lex (Rutaceae) cultivated in Egypt. Their structures were determined by UV/Vis, IR, 1H and 13C NMR, and EI mass spectroscopy. The alkaloids were also detected in the kernels of the seeds. Received May 28, 2001. 设计合成了一种以喹啉酮为荧光团,具有新型结构的荧光探针(E)-N-(4-甲基-2-氧代-1,2-二氢喹啉-7-基)-3-(3-苯基-[1,2,4]三唑[3,4-b][1,3,4]噻二唑-6-基)丙烯酰胺(MNT).研究结果表明,MNT可通过不饱和酰胺键异构化后与Hg2+配位,从而产生显著的荧光猝灭.1,2,4-三唑[3,4-b]-1,3,4-噻二唑缺电子的特征有助于提高猝灭效果的同时,能提供与Hg2+配位的杂原子.MNT探针对Hg2+具有高选择性、较高的量子产率和较强的抗干扰性,检测限为6.35×10-8 mol/L,响应时间25 s.进一步研究发现,MNT在pH=4~13范围内均能特异性检测Hg2+.基于核磁滴定实验结果推测了该探针荧光检测Hg2+的机理,并由Job’s曲线确定了MNT与Hg2+之间的配位比为2:1.MNT在实际水样中的应用结果表明其可作为检测Hg2+的荧光探针.  相似文献

A new application of the Ugi reaction in the synthesis of heterocyclic compounds is described. Substituted quinolin-2-(1H)-ones are formed in one-pot sequential Ugi four-component condensation and intramolecular Knoevenagel cyclization between o-acylanilines, aldehydes, malonic or tosylacetic acids and cyclohexyl isocyanide.  相似文献

N-(1-Ethoxy-2,2,2-trifluoroethyl)anilines 2a-2f, prepared from trifluoroacetaldehyde ethyl hemiacetal and aniline, readily reacted with diethyl malonate in the presence of sodium hydride, giving substituted products 5a-5f in high yields. Compounds 5a-5f subjected to hydrolysis and decarboxylation under specified conditions yielded the 4,4,4-trifluorobutyric acids 6a-6e or 7. Direct ring-closure of 6a-6e with polyphosphoric acid gave 2-trifluoromethyl-2,3-dihydro-1H-quinolin-4-ones 9a-9e.  相似文献

An efficient method for visible-light-induced perfluoroalkylaion of quinolin-4-ol has been reported. In the presence of t-BuONa and perfluoroalkyl iodide, quinolin-4-ol underwent CH perfluoroalkylation under the irradiation of green light. Mechanistic studies demonstrated that visible-light promoted intermolecular charge transfer within the transient electron donor-acceptor complex in the absence of any photocatalysts.  相似文献

The rotational barriers in axially chiral quinolin-2-one and quinazolin-2-one possessing N-(ortho-tert-butyl)phenyl group were found to significantly increase in comparison with those of corresponding dihydroquinolin-2-one and dihydroquinazolin-2-one. Analysis of transition state structure during N-Ar bond rotation based on DFT calculation indicates that the increase in the rotational barrier is due to considerable distortion of the nitrogen-containing heterocyclic part.  相似文献
