Five compounds, terephthalic acid mono-[2-(4-carboxy-phenoxycarbonyl)-vinyl] ester (1), (E)-3-(5′-hydroperoxy-2,2′-dihydroxy[1,1′-biphenyl]-4-yl)-2-propenoic acid (2), 3,4,5-trihydroxybenzoic acid (3), succinic acid (or butanedioic acid) (4), and 2,3,4,5,6-pentahydroxybenzoic acid (5), were isolated from Phyllanthus urinaria. 组装了一种新型的微波辅助旋转回流装置,该装置的基本原理与微波辅助溶剂萃取的原理相同,但采用了旋转的技术以加速提取.应用该装置以正交实验筛选优化叶下珠中有机酸的提取工艺,并利用毛细管电泳技术分离测定了提取液中有机酸的含量以评价提取效果.最佳提取工艺:乙醚为溶剂,微波功率800 W,提取时间4 min,溶剂用量300 mL.在该提取条件下,平行5次提取叶下珠中丁二酸、原儿茶酸、没食子酸、咖啡酸、阿魏酸的平均含量分别为42.2、103.5、436.2、123.8、67.4 μg/g,相对标准偏差为0.87% ~3.7%,加标回收率为94% ~104%.将该法与常规的微波辅助提取法及回流提取法进行比较,结果表明,微波辅助旋转回流提取法提取效率明显优于其它2种方法.  相似文献

The efficient production of plant-derived medicinal compounds (PDMCs) from in vitro plants requires improvements in knowledge about control of plant or organ development and factors affecting the biosynthesis pathway of specific PDMCs under in vitro conditions, leading to a realistic large-scale tool for in vitro secondary metabolite production. Thus, this study aimed to develop an in vitro technique, through the induction and proliferation of calli, for production of plant fresh weight, and to compare the PDMC profile obtained from the plants versus in vitro calli of Phyllanthus amarus. It was successfully possible to obtain and proliferate two types of calli, one with a beige color and a friable appearance, obtained in the dark using Murashige and Skoog (MS) medium plus 2,4-dichlorophenoxyacetic acid (2,4-D), and a second type with a green color, rigid consistency, and nonfriable appearance obtained under light conditions and MS medium plus 6-benzyladenine (6-BA). In vitro micropropagated plants that gave rise to calli were also acclimatized in a greenhouse and cultivated until obtaining the mass for PDMC analysis and used as a control. While the micropropagated-derived plants concentrated the lignans niranthin, nirtetralin, and phyllanthin, the Phyllanthus amarus calli proliferated in vitro concentrated a completely different biochemical profile and synthesis of compounds, such as betulone, squalene, stigmasterol, and β-sitosterol, in addition to others not identified by GC-MS database. These results demonstrate the possibility of applying the calli in vitro from Phyllanthus amarus for production of important PDMCs unlike those obtained in cultures of differentiated tissues from field plants.  相似文献

Disease progression in Tuberculosis (TB) is dependent on host’s immune system. Phyllanthus niruri, a traditional herb, has long been used to boost immune system in Indonesian society. This study aimed to observe the potential role of P. niruri in inducing immune cells activity in TB patients by in vitro approach. Peripheral blood mononuclear cells (PBMCs) and macrophages were collected from active pulmonary TB patients. After stimulation with graded doses of P. niruri aqueous extract, cell proliferation, phagocytic activity and nitric oxide (NO) release were analysed. P. niruri aqueous extract induced proliferation of PBMCs, increased NO release, and improved macrophages phagocytic activity. These effects were observed in a dose-dependent manner. This may lead to further research for the potential role of P. niruri as immunomodulatory adjuvant therapy for TB patients.  相似文献

A simple and rapid high-performance thin-layer chromatographic method for quantification of gallic acid and ellagic acid in dried fruits of Terminalia chebula, Phyllanthus emblica, and Quercus infectoria has been developed. The chromatographic development was carried out on precoated silica gel 60 F254 plates in a mixture of toluene:ethyl acetate:chloroform:formic acid (4:8:1:3 v/v/v/v). The plate was scanned densitometrically at a wavelength of 280 nm. The retention factor value of gallic acid and ellagic acid was found to be 0.63 ± 0.2 and 0.53 ± 0.1, respectively. The developed method was validated in terms of linearity, precision, accuracy, sensitivity, robustness, specificity and stability as per the international conference of harmonization guidelines. The method showed good linear relationship over a range of 100–600 ng/band (gallic acid) and 100–500 ng/band (ellagic acid) with a regression coefficient (r2) of 0.997 (gallic acid) and 0.996 (ellagic acid). The method showed high accuracy (99.65%–100.85%). The percentage relative standard deviation of intra-day and inter-day precision studies was not more than 2%. The method is highly robust and has displayed high specificity. The developed method is new, simple, and accurate and can be successfully employed in routine analysis of raw materials and formulations containing gallic acid and ellagic acid.  相似文献

Three compounds were isolated from Phyllanthus niruri Linn. One effective method, improved dry column liquid chromatography for isolating these compounds, was applied on a silica gel column. It is a combination of conventional dry column chromatographic and flash chromatographic techniques. Structures of those obtained compounds were elucidated by means of spectral techniques including IR, MS, and 1D NMR and 2D NMR. Compound 1 was elucidated as 2,3,5,6-tetrahydroxybenzyl acetate. Compound 2 , 2,4,5-trihydroxy-3-(4,6,7-trihydroxy-3-oxo-1,3-dihydroisobenzofuran-5-yl)-benzoic acid methyl ester, was given the name phyllangin, and compound 3 was named corilagin. Phyllanthus emblica L. is an economic plant used in Chinese medicine for the treatment of various diseases. The bark of P. emblica is rich in polyphenols and its extractions have shown strong antioxidative and radical scavenging activity. Response surface methodology (RSM) was used to assess the optimal extraction of polyphenols from P. emblica bark. Various extraction parameters including ethanol concentration, extraction time, temperature, solid–liquid ratio, and extraction times were chosen to identify their effects on polyphenols extraction. Among these parameters, extraction times and solvent concentration were found to have significant effect on polyphenols extraction. RSM was applied to obtain the optimal combination of solvent concentration, extraction time, temperature, and extraction time for maximum rate of extraction. The most suitable condition for the extraction of polyphenols was at ethanol concentration 75%, extraction time 25 min, extraction temperature 45°C, and extraction times 3. At these optimal extraction parameters, the maximum extraction of polyphenols obtained experimentally was found to be very close to its predicted value. The extraction rate of polyphenols was 19.78% at the optimum conditions. The mathematical model developed was found to fit with the experimental data of polyphenols extraction.  相似文献

A combination of flash chromatography, solid phase extraction, high-performance liquid chromatography, and in vitro bioassays was used to isolate phytocomponents endowed with anticholinesterase activity in extract from Phyllanthus muellarianus. Phytocomponents responsible for the anti-cholinesterase activity of subfractions PMF1 and PMF4 were identified and re-assayed to confirm their activity. Magnoflorine was identified as an active phytocomponent from PMF1 while nitidine was isolated from PMF4. Magnoflorine was shown to be a selective inhibitor of human butyrylcholinesterase—hBChE (IC50 = 131 ± 9 μM and IC50 = 1120 ± 83 μM, for hBuChE and human acetylcholinesterase—hAChE, respectively), while nitidine showed comparable inhibitory potencies against both enzymes (IC50 = 6.68 ± 0.13 μM and IC50 = 5.31 ± 0.50 μM, for hBChE and hAChE, respectively). When compared with the commercial anti-Alzheimer drug galanthamine, nitidine was as potent as galanthamine against hAChE and one order of magnitude more potent against hBuChE. Furthermore, nitidine also showed significant, although weak, antiaggregating activity towards amyloid-β self-aggregation.  相似文献

Abstract

Phyllanthus phillyreifolius (Euphorbiaceae), poorly studied plant species, was fractionated using conventional and high pressure extraction techniques such as supercritical fluid and pressurized liquid extractions. Lipophilic substances were extracted with n-hexane and supercritical CO2 with or without co-solvent ethanol, meanwhile higher polarity fractions were recovered with acetone and 70% ethanol. Antioxidant potential was assessed by various chemical assays, which revealed that 70% ethanol was the most effective solvent for recovery of antioxidants. UPLC-MS phytochemical analysis of hydrophilic extracts confirmed geraniin as the main constituent of P. phillyreifolius. Other quantitatively important compounds were phyllanthusiin D and elaeocarpusin. Three isomers of tocopherol (α, β and γ) were quantified by HPLC in lipofhilic extracts. Generally, the results from this study revealed high antioxidant potential of P. phillyreifolius; consequently the plant may be considered as a promising source of antioxidants for functional foods, nutraceuticals and pharmaceutical formulations.  相似文献

Fifty-eight phenolic compounds isolated from Phyllanthus emblica were screened and compared for their in vitro and in vivo antioxidant properties, as well as hyaluronidase (HAase) inhibitory activities. Among them, 20 compounds showed to be promising antioxidants due to the stronger scavenging activity in both DPPH radical and Danio rerio reactive oxygen species assays, while nine compounds were potential HAase inhibitors with 100-fold stronger activities than that of the positive control, DSCG.
