Pd-catalyzed arylation/ring-closing metathesis approach to azabicycles

Palladium-catalyzed arylation followed by Grignard addition to imines and ring-closing metathesis, using Grubbs’ catalysts, provides a route to six-, seven-, and eight-membered azabicycles.     

Preparation of a series of novel fluorophores, N-substituted 6-amino and 6,6″-diamino-2,2′:6′,2″-terpyridine by palladium-catalyzed amination

A new series of N-substituted 6-amino- and 6,6″-diamino-2,2′:6′,2″-terpyridine (6-amino- and 6,6″-diamino-tpy) was conveniently synthesized in one-step by Pd-catalyzed amination of bromo-substituted tpys with various amines. For highly coordinating tpy substrates, use of appropriate chelating phosphine ligand was critical to achieve moderate to satisfactory yield. The prepared N-substituted 6-amino- and 6,6″-diamino-tpys exhibited moderate to intense fluorescence in dichloromethane with fine-tuned fluorescence maxima ranging from 385 to 455 nm.     

Suzuki-Miyaura coupling reaction of aryl chlorides using di(2,6-dimethylmorpholino)phenylphosphine as ligand

Suzuki-Miyaura coupling was achieved on a variety of aryl chlorides by using di(2,6-dimethylmorpholino)phenylphosphine (L1) as a bulky electron-rich monoaryl phosphine ligand. We report the couplings of various chlorobenzenes and heteroaryl chlorides.     

Asymmetric construction of quaternary carbon stereocenter by Pd-hemilabile ligand-catalyzed allylic substitution

The catalyst comprised [PdCl(η³-C₃H₅)]₂ and a simple chiral hemilabile ferrocene ligand, 1′,2-bis(diphenylphosphinoethyl)ferrocenyl alcohol, provides synthetically acceptable results for an enantioselective Pd-catalyzed allylic alkylation of cyclohexanone derivatives bearing an electron-withdrawing group at the α-position to form the quaternary carbon with up to 90% enantioselectivity under mild reaction conditions.     

Synthesis of Bicyclic Compounds Derived from 1,2,4-Benzotriazines

The synthesis of bicyclic compounds derived from benzo[1,2,4]triazines is described.     

One-Pot Palladium-Catalyzed Ligand- and Metal-Oxidant-Free Aerobic Oxidative Isocyanide Insertion Leading to 2-Amino-substituted-4(3H)-quinazolinones

An efficient, ligand- and metal-oxidant-free, one-pot, cascade aerobic oxidative, palladium-catalyzed, multicomponent reaction has been developed through isocyanide insertion of less active secondary amide and aromatic amine, which leads to 2-amino-substituted-4(3H)-quinazolinones. This approach proves to be one of the simplest methods for the synthesis of this class of valuable bioactive heterocyclic scaffolds.     

3,4-Thienylene-ethynylene Oligomers

Several well-defined 3,4-thienylene-ethynylene oligomers with different chain lengths have been synthesized and characterized. X-Ray crystallographic analyses proved their unique, totally helical conformations.     

Synthetic Utilization of α-Phosphonovinyl Anions

The α-phosphonovinyl anions, generated in situ from treatment of β-hetero-substituted vinyl-phosphonates 1a-c with LDA (or LTMP), were trapped with various electrophiles such as chlorotriorganosilanes, chlorotrimethylgermane, chlorotriorganotins, dimethyl disulfide, and halogen to afford the corresponding β-hetero-substituted α-functionalized vinylphosphonates 2–14 in good to excellent yields. The Friedel-Crafts reaction of α-(silyl) or α-(germyl)phosphonoketene dithioacetals 2, 9 or 4 with acid chlorides gave α-acylated phosphonoketene dithioacetals 15–19 in 53–91 % yields. The palladium-catalyzed cross-coupling reaction of β-ethoxy-α-(tributylstannyl)vinylphosphonate 13 with a variety of organic halides (R = acyl, allyl, aryl etc.) provided β-ethoxy-α-substituted vinylphosphonates 20–25 in good to moderate yields. The palladium-mediated cross-coupling reaction of α-(iodo)-vinyl-phosphonates 7, 14 with terminal acetylenes afforded α-alkynylated vinylphosphonates 26–29 in 69–83 % yields.     

Regioselective Syntheses of 1-Aryl-substituted-5H-[1,3]thiazolo[3,2-a]quinazoline-5-ones During Sonogashira Coupling

Abstract

Reaction of 2-mercaptopropargylquinazoline-4-one with various aryliodides catalyzed by Pd–Cu leads to the regioselective formation of 1-arylsubstituted-5H-[1,3]thiazolo-[3,2-a]quinazoline-5-ones.
