An easy, fast and reliable analytical method is proposed to determine the concentration of volatile phenols (ethyl- and vinylphenols) in fino sherry wines. The technique employed is a single stage sample treatment by solid-phase extraction (SPE) following a simple, fast procedure that enables 12 samples to be extracted simultaneously and requires a small volume sample and little time. Subsequently, the extracts are analyzed by gas chromatography (GC) with flame ionization detection. The method proposed has been applied to the study of fino sherry wines affected by microbial contamination with yeasts of the Brettanomyces genus, and the relationship of these yeasts with the concentrations of volatile phenols present in this wine.  相似文献

β-glucans are known as biological response modifiers. However, different sources can result in structural differences and as a result differences in their biological activity. The hot water extraction method allows to obtain, high molecular weight β-glucans without altering their structure by using strong chemicals, such as alkalis or acids. Analysis of β-glucans by FT-IR and NMR spectroscopy in solid state is superior to analysis in solution as it allows researchers to study the preserved structure of the extracted polysaccharides. FT-IR spectroscopy was used in this study to make side-by-side comparison analysis of hot water extracted β-glucans from different yeast sources. NMR spectroscopy was used to confirm findings made by FT-IR spectroscopy. Extracted β-glucans exhibit characteristic structure of β-1,3/1,6-linked glucans with noticeable levels of proteins, possibly in a form of oligopeptides, chitin and other impurities. β-glucans obtained from C. guilliermondii, P. pastoris and S. pastorianus exhibited higher protein content. Differences in mannan, chitin and α-glucan content were also observed; however, the species-specific structure of obtained β-glucans could not be confirmed without additional studies. Structural analysis of high molecular weight β-glucans in solid state by FT-IR spectroscopy is difficult or limited due to band intensity changes and overlapping originating from different molecules.  相似文献

研究了脉冲宽度对高压脉冲下细胞膜和细胞器膜跨膜电压的影响及细胞膜能量的变化,以及13 s,200 ns和2 ns脉冲对酿酒酵母(CICC31180)杀灭作用时脉冲数的影响。结果表明,随着脉冲数增加,13 s,200 ns和2 ns脉冲对啤酒酵母的灭杀效果均增强。在13 s脉宽,14 kV电压和50个脉冲数下,酵母达到最小存活率-4.3个对数;在200 ns脉宽,40 kV电压和500个脉冲数下,酵母达到存活率-2.6个对数;在2 ns脉宽,200 kV电压和500个脉冲数下,酵母达到存活率-2.8个对数。三种脉冲对酵母菌杀灭的存活率对数与脉冲数的关系表现不是线性的,脉冲数增加到一定程度,存活率下降变缓。在同等灭杀效果-2.0个对数下,2 ns脉冲比13 s脉冲在同轴电极中损耗的能量更少。  相似文献

蛋白质-蛋白质、蛋白质-DNA相互作用网络决定了细胞中各种关键功能的执行.基于芽殖酵母(budding yeast saccharomyces cerevisiae)的蛋白质-蛋白质相互作用网络数据和相关的实验文献,我们建立了调控细胞周期和生命周期(cell cycle and life cycle)的蛋白质网络,并利用离散模型研究了该网络的动力学性质,研究表明:细胞周期网络的动力学性质具有很强的稳定性,约94%的蛋白质初态将演化到对应于生物学G1基态的稳定态,使其成为惟一的全局吸引点;同时,绝大多数的初态的演化路径都通过由G1激发态到G1基态的细胞周期演化路径,使细胞周期路径成为全局性的“吸引”路径。  相似文献

以鱼蛋白为原料,在酶解与曲酶发酵相结合的基础上,于后熟阶段加入硒酵母作为有机硒的载体和增香工具,生产品质优良的富硒营养鱼酱油。试验结果表明:采用后熟温度35℃,酵母液量10%,酱醅盐度8%和12%,后熟发酵周期12d的工艺条件,可生产出优质的富硒鱼酱油。产品的氨基氮、有机酸等营养成分均高于国家标准,有机硒的含量达到国内专用硒保健品的水平。  相似文献

Yarrowia lipolytica, an oleagineous species of yeast, is a carrier of various important nutrients. The biomass of this yeast is an extensive source of protein, exogenous amino acids, bioavailable essenctial trace minerals, and lipid compounds as mainly unsaturated fatty acids. The biomass also contains B vitamins, including vitamin B12, and many other bioactive components. Therefore, Y. lipolytica biomass can be used in food supplements for humans as safe and nutritional additives for maintaining the homeostasis of the organism, including for vegans and vegetarians, athletes, people after recovery, and people at risk of B vitamin deficiencies.  相似文献

A method based on ICP collision-cell MS detection in capillary HPLC was developed to gain an insight into the purity and identity of selenium-containing proteins separated by 1-D and 2-D electrophoresis. The bands and spots obtained after the separation of water-soluble proteins in selenized yeast were digested with trypsin prior to chromatography. Selenium could be detected down to the subpicogram level. The method, assisted by information obtained by MALDI TOF MS on the 5000 Da cut-off fraction, permitted the purity of bands and spots to be estimated and the efficiency of tryptic digestion and the quantity of selenium present in individual peptides to be evaluated. Owing to the high sensitivity and the lack of matrix suppression effects, the method provided chromatograms with signal-to-noise ratios of 10–1000 in conditions where the common ES Q–TOF MS detection failed.   相似文献

l-Arabinose is one of the sugars found in hemicellulose, a major component of plant cell walls. The ability to convert l-arabinose to ethanol would improve the economics of biomass to ethanol fermentations. One of the limitations for l-arabinose fermentation in the current engineered Saccharomyces cerevisiae strains is poor transport of the sugar. To better understand l-arabinose transport and use in yeasts and to identify a source for efficient l-arabinose transporters, 165 non-Saccharomyces yeast strains were studied. These yeast strains were arranged into six groups based on the minimum time required to utilize 20 g/L of l-arabinose. Initial transport rates of l-arabinose were determined for several species and a more comprehensive transport study was done in four selected species. Detailed transport kinetics in Arxula adeninivorans suggested both low and high affinity components while Debaryomyces hansenii var. fabryii, Kluyveromyces marxianus and Pichia guilliermondii possessed a single component, high affinity active transport systems.  相似文献

Pilot plant trials were conducted in a corn wet mill with a 7000-L membrane recycle bioreactor (MRB) that integrated ceramic microfiltration membranes in a semi-closed loop configuration with a stirred-tank reactor. Residence times of 7.5–10 h with ethanol outputs of 10–11.5% (v/v) were obtained when the cell concentration was 60–100 g/L drywt of yeast, equivalent to about 109−1010 cells/mL. The performance of the membrane was dependent on the startup mode and pressure management techniques. A steady flux of 70 L/(m2·h) could be maintained for several days before cleaning was necessary. The benefits of the MRB include better productivity; a clear productstream containing no particulates or yeast cells, which should improve subsequent stripping and distillation operations; and substantially reduced stillage handling. The capital cost of the MRB is $21–$34/(m3·yr) ($0.08–$0.13/[gal·yr]) of ethanol capacity. Operating cost, including depreciation, energy, membrane replacement, maintenance, labor, and cleaning, is $4.5–9/m3 ($0.017–$0.034/gal) of ethanol.  相似文献
