The first stereoselective total synthesis of natural product, dendrodolide A (1) is described from readily available (R)-propylene oxide and 3-buten-1-ol as starting materials. The synthesis was achieved in 10 steps with an overall yield of 19.1%. The key steps involved in the synthesis are Jacobsen hydrolytic kinetic resolution, epoxide ring opening with 2-allyl-1, 3-dithiane, Yamaguchi esterification, and ring-closing metathesis (RCM).  相似文献

Even though the macrolides of the iejimalide family are of marine origin, whereas those of the archazolid series derive from terrestrial myxobacteria, a comparison of their constitution, stereochemistry, and biological activity suggests that these natural products are close structural and functional relatives. Guided by this perception, compound 5 was prepared, which hybridizes the macrolactone core of iejimalide B (2) with the tail of archazolid A (3). The cytotoxicity profile of this chimera, as determined with a panel of 12 human cancer cell lines, corresponds to that of the parent compound 2, although its potency is lower. This outcome may be interpreted on the basis of molecular dynamics calculations, which suggest that the low energy conformations of 2 and 5 are similar but the energetic barriers between the relevant conformers are distinctly higher for the hybrid structure. The synthesis of 5 hinged on a regioselective functionalization of 2,4-dibromothiazole 6, a highly selective CBS-reduction of ketone 8, a Suzuki cross coupling of vinyl boronate 17 with the elaborate alkenyl iodide 16, and a productive closure of the macrocycle by RCM, which requires the selective activation of two out of eight double bonds present in the cyclization precursor 20 by the second-generation Grubbs catalyst 21.  相似文献

A stereoselective synthesis of 14‐membered macrolide hamigeromycin E ( 6 ) has been studied by employing ortho‐lithiated formylation, Barbier allylation, JuliaKocienski olefination, Mitsunobu esterification, and ring‐closing metathesis (RCM) reactions. The final RCM reaction did not provide the target molecule. This study has prompted us to synthesize a stereoisomer of zeaenol and accomplish the total synthesis with the above protocols.  相似文献

Two wastewater treatment plants (WWTPs) of northern Colorado were monitored for anhydroerythromycin and tylosin. An analytical method has been developed and validated for the trace determination and confirmation of these compounds in the raw influent and final effluent water matrices. This method was used to evaluate the occurrence and fate of these compounds in WWTPs. The method uses solid-phase extraction and liquid chromatography–tandem mass spectrometry with positive electrospray ionization. Detection and quantification was performed using selected reaction monitoring, and a method detection limit of between 0.01 and 0.06 μg/L was obtained. Unequivocal confirmation analysis of analyte identity according to the criteria (based on the use of identification points) of the 2002/657/EC European Commission Decision was possible with satisfactory results. Average recoveries for the two compounds ranged from 89.2±9.7% for raw influent to 93.7±6.9% for effluent wastewaters. The within-run precision of the assay was found to be always less than 14.1% for the two analytes. The overall precision was always less than 13.7%. The relative uncertainty of the present assay was also evaluated and the combined relative uncertainty ranged from 6.4 to 15.5% over three days of the validation study. These compounds were partially removed in the WWTPs with a removal efficiency of >50%. The measured concentrations in raw influents and effluents ranged from 0.09–0.35 and 0.04–0.12 μg/L for anhydroerythromycin to 0.06–0.18 and ND–0.06 μg/L for tylosin, respectively. The results indicate that WWTP effluents are relevant point sources for residues of these compounds in the aquatic environment. These occurrence results were compared with those in WWTP wastewaters of other countries.  相似文献

Total synthesis of neomethymycin and novamethymycin has been achieved. These two macrolides contains 12-membered macrolactones as aglycones and belong to the methymycin family of antibiotics, which appears in the pikromycin biosynthetic pathway. The segments in the 12-membered macrolactone that are responsible for causing structural difference in neomethymycin and novamethymycin were synthesized by starting with methyl d-(+)-lactate and d-glucose, for neomethynolide, and for novamethynolide, respectively. The key steps in synthesis of neomethynolide and novamethynolide, which are aglycones for neomethymycin and novamethymycin, respectively, were asymmetric aldol reactions, Yamaguchi esterification, and ring-closing metathesis using Grubbs’ second generation catalyst. Finally, the coupling of agylcones with the corresponding trichloroacetimidates, followed by deprotection, completed the total synthesis of these two macrolide antibiotics.  相似文献

Summary.  Microtubule stabilizing natural products, as exemplified by paclitaxel (taxol?), are being considered as novel drugs against malignant therapy resistent solid tumors. Among these compounds, epothilone B and some of its derivatives have emerged as particularly promising candidates for industrial development. The total and partial syntheses of these compounds are described in detail, and some of the most important recent results on their biological activity are discussed. Received December 3, 1999. 建立了水产品中大环内酯类抗生素[红霉素(ERM)、罗红霉素(ROM)、替米卡星(TIL)、泰乐菌素(TYL)、北里霉素(KIT)、交沙霉素(JOS)、竹桃霉素(OLM)、螺旋霉素Ⅰ(SPM-Ⅰ)]和林可胺类(林可霉素(LIN)和氯林可霉素(CLD))的高效液相色谱-电喷雾串联质谱(LC-ESI-MS/MS)检测方法.样品经提取、反相液相色谱分离净化后进行质谱分析,在选择反应监测模式(SRM)下进行特征母-子离子对信号采集.根据保留时间和母离子及两个特征子离子信息定性分析,以基峰离子进行定量.大环内酯类残留的检出限(S/N=3)为0.1~0.2μg/kg,定量限为1.0μg/kg,在1.0~200 ng/mL时峰强度与质量浓度的线性关系良好(R~2 >0.99).在虾、鳗鱼和带鱼3种基质中1.0、2.0、10.0μg/kg 3个添加水平下,除个别药物外,药物的平均回收率范围为64%~114%,RSD<12%.该法适用于各种水产品中大环内酯类残留的分析.  相似文献

Tricrilactones A–H ( 1 – 8 ), a new family of oligomeric 10-membered macrolides featuring collectively five unique ring skeletons, were isolated from a hitherto unexplored fungus, Trichocladium crispatum. Compounds 1 and 7 contain two unconventional bridged (aza)tricyclic core skeletons, 2 , 3 , 5 , and 6 share an undescribed tetracyclic 9/5/6/6 ring system, 4 bears an uncommon 9/5/6/10/3-fused pentacyclic architecture, and 8 is a dimer bridged by an unexpected C−C linkage. Their structures, including absolute configurations, were elucidated by spectroscopic analysis, quantum chemical calculations, and X-ray diffraction analysis. Importantly, the absolute configuration of the highly flexible side chain of 1 was resolved by the asymmetric synthesis of its four stereoisomers. The intermediate-trapping and isotope labeling experiments facilitated the proposal of the biosynthetic pathway for these macrolides. In addition, their antiosteoporosis effects were evaluated in vivo (zebrafish).  相似文献
