The adsorption of cellulose acetate (CA), cellulose acetate propionate (CAP) and cellulose acetate butyrate (CAB) from solutions
prepared in acetone onto silicon wafers led to ultrathin films, which were characterized by ellipsometry, atomic force microscopy
(AFM) and contact angle measurements. The polysaccharides films were characterized in the air just after their formation and
after annealing at temperatures higher than their glass transition temperature or melt temperature. The films thickness close
to 2 nm and surface roughness did not vary significantly upon annealing. AFM images revealed the presence of small clumps
dispersed on a homogeneous layer, which covered completely the Si wafers. Such topographic details were also observed after
annealing. However, upon annealing the films surfaces changed from hydrophilic to hydrophobic, evidencing molecular re-orientation
at the solid–air interface. The adhesion of bovine serum albumin (BSA) and lipase onto the cellulose esters films was quantified
in order to evaluate the possibility of applying such films as selective support for biomolecules. 相似文献

In the enantioselective esterification of racemic naproxen with 4(2-hydroxyethyl) morpholine by Lipase MY in organic solvents,
a productivity improvement of the desired (S)-naproxen ester from 0.42 to 0.72 mM at the reaction time of 130 h was observed,
when the surfactantbis (2-ethylhexyl) sodium sulfosuccinate (AOT) was added in the reaction mixture. The presence of a small amount of exogenously
added water dramatically activated the enzyme in AOT/cyclohexane-reversed micelles. Desorption of the surfactant molecule
from the enzyme mass and solubilization of the enzyme into reversed micelles were used to elucidate an existing maximum of
the initial rate of (S)-naproxen synthesis with the water content. Moreover, the effects of alcohol and surfactant concentration

The effects of sodium dodecyl sulfate on extracellular lipase produced byCandida lipolytica have been studied. The microorganism was grown in culture medium containing different sodium dodecyl sulfate concentrations
added to the culture at different intervals of growth. The extracellular lipase activity was not detected when the treated
culture supernatants were directly tested in Yeast Mold Agar-Triolein-Rhodamine plates, regardless of surfactant addition
time and concentrations. However, after ammonium sulfate precipitation and dialysis, the extracellular lipase activity could
be recovered. Therefore, the surfactant, under the experimental conditions used here, does not seem to be able to inhibit

Mucor miehei lipase was immobilized on magnetic polysiloxane-polyvinyl alcohol particles by covalent binding with high activity recovered.
The performance of the resulting immobilized biocatalyst was evaluated in the synthesis of flavor esters using heptane as
solvent. The impact on reaction rate was determined for enzyme concentration, molar ratio of the reactants, carbon chain length
of the reactants, and alcohol structure. Ester synthesis was maximized for substrates containing excess acyl donor and lipase
loading of 25 mg/mL. The biocatalyst selectivity for the carbon chain length was found to be different concerning the organic
acids and alcohols. High reaction rates were achieved for organic acids with 8 or 10 carbons, whereas increasing the alcohol
carbon chain length from 4 to 8 carbons gave much lower esterification yields. Optimal reaction rate was determined for the
synthesis of butyl caprylate (12 carbons). Esterification performance was also dependent on the alcohol structure, with maximum
activity occurring for primary alcohol. Secondary and tertiary alcohols decreased the reaction rates by more than 40%. 相似文献

An extracellular lipase was purified from the fermentation broth of Bacillus coagulans ZJU318 by CM-Sepharose chromatography, followed by Sephacryl S-200 chromatography. The lipase was purified 14.7-fold with
18% recovery and a specific activity of 141.1 U/mg. The molecular weight of the homogeneous enzyme was (32 kDa), determined
by sodium dodecyl sulfate (SDS) polyacrylamide gel electrophoresis. The enzyme activity was maximum at pH 9.0 and was stable
over a pH range of 7.0–10.0, and the optimum temperature for the enzyme reaction was 45°C. Little activity loss (6.2%) was
observed after 1 h of incubation at 40°C. However, the stability of the lipase decreased sharply at 50 and 60°C. The enzyme
activity was strongly inhibited by Ag+ and Cu2+, whereas EDTA caused no inhibition. SDS, Brij 30, and Tween-80 inhibited lipase, whereas Triton X-100 did not significantly

Lipase fromCandida rugosa was stabilized against thermal inactivation in the presence of polyvinylalcohols (PVA) of different molecular weights. The
apparent rate constant of the lipase inactivation, kd, at 49°C is 0.049/min and 0.022/min in the absence and in the presence of PVA (mol wt 22,000), respectively. The improvement
of the lipase thermostability by adding PVA was confirmed by differential scanning calorimetry. The presence of PVA had also
an effect on the hydrolytic activity of the enzyme. Furthermore, lipase was modified by covalent linkage to PVA by means of
an original procedure. With respect to the native enzyme, the modified lipase has a slightly lower specific activity, but
it is more stable against heat denaturation (k
