An alternative synthetic strategy for (3R),(5R)-5-hydroxy-de-O-methyllasiodiplodin and its epimer

A concise total synthesis of (3R),(5R)-5-hydroxy-de-O-methyllasiodiplodin and its epimer have been synthesized from commercially available (R)-propylene oxide and Orsellinic acid as starting materials. The key steps involved in the synthesis are Wittig reaction, epoxide ring opening with 1,3-dithiane intermediate and Yamaguchi macrolactonization.

Two further lasiodiplodins, (3R,4R)-4-hydroxy-de-O-methyl-lasiodiplodin and (E)-9-etheno-de-O-methyl-lasiodiplodin, together with three known lasiodiplodins from a cytotoxic extract obtained from a culture of Lasiodiplodia theobromae, an endophyte from the root tissues of Mapania kurzii (Cyperaceae) from the Malaysian rain forest, were characterized on microgram scale.

Abstract

An alternative and concise total synthesis of (3?R, 4S)-4-Hydroxylasiodiplodin has been reported from inexpensive and commercially available 6-heptenal and Orsellinic acid starting materials. The key steps involved in the synthesis are Wittig reaction, Sharpless epoxidation, Yamaguchi esterification and ring-closing metathesis (RCM).
