Solid-phase synthesis of liquid crystalline isoxazole library

Combinatorial library of isoxazoles was prepared by 1,3-dipolar cycloaddition on solid support. p-Cyano derivatives showed nematic and/or smectic A phases. A bilayer smectic phase for 2h is proposed by the combination of molecular mechanics calculation and X-ray diffraction experiment.     

An environmentally benign synthesis of isoxazolines and isoxazoles mediated by potassium chloride in water

An effective and environmentally benign procedure for the synthesis of isoxazolines and isoxazoles has been developed by a cycloaddition of nitrile oxides with alkenes or alkynes in water. In this approach, potassium chloride is first oxidized into chlorine in water by the environmentally friendly oxidant Oxone®, then aldoximes are oxidized into nitrile oxides by the in situ generated hypochlorous acid, finally a 1,3-dipolar cycloaddition between nitrile oxides and alkenes or alkynes occurs to provide the corresponding isoxazolines and isoxazoles in good yields.     

Synthesis of novel isoxazole-benzoquinone hybrids via 1,3-dipolar cycloaddition reaction as key step

An efficient method for the preparation of novel 2-(5-arylisoxazol-3-yl)cyclohexa-2,5-diene-1,4-dione hybrids via 1,3-dipolar cycloaddition followed by an oxidation reaction using ceric ammonium nitrate (CAN) has been described. Using this method, various aryl as well as alkyl substituted isoxazole-benzoquinone hybrids were synthesized in high yields.     

Intramolecular 1,3-dipolar nitrone and nitrile oxide cycloaddition of 2- and 4-O-allyl and propargyl glucose derivatives: a versatile approach to chiral cyclic ether fused isoxazolidines, isoxazolines and isoxazoles

2-O- and 4-O-Allyl and -propargyl glucose and the corresponding oxime derivatives were prepared from readily available glucose dithioacetals. Intramolecular 1,3-dipolar cycloaddition of the N-benzyl and N-methyl nitrones of the above acyclic 2-O-allyl glucose derivatives led to the diastereoselective formation of chiral isoxazolidines incorporating the tetrahydrofuran ring. The EI mass spectra revealed a characteristic cleavage of the C-alkyl group adjacent to the furan oxygen atom. An enantiopure trisubstituted tetrahydrofuran was obtained by the reductive cleavage of the isoxazolidine ring of one of the cycloadducts. In contrast, the nitrile oxide cycloaddition of the 2-O-allyl derivatives afforded diastereomeric mixtures of the corresponding dihydroisoxazolines, the stereochemistry of which was tentatively assigned on the basis of the principle of optical superposition. The exclusive formation of a tetrahydrofuran ring from pentaallyl nitrone or nitrile oxide demonstrated the preferred formation of a five-membered ring to that of six or seven-membered rings. The nitrile oxide generated from a 3,4,5,6,7-pentaallyloxy-1-nitroheptane derivative obtained from pentaallylglucose underwent diastereoselective cycloaddition to give an isoxazoline fused to a pyran ring. Enantiopure isoxazoles containing tetrahydrofuran and oxepane rings were also prepared in good yields by the nitrile oxide cycloaddition of the 2-O- and 4-O-propargyl derivatives.     

Synthesis,crystal structure,spectroscopic, and biological studies on Cu(II) complexes of N,O donor dimethyl isoxazole Schiff bases

Cu(II) complexes with Schiff bases DMIIMP, DMIIMBD, DMIIMBP, DMIIMCP, DMIIMMP, and DMIIMNP (see Introduction for definitions) are derived from condensation of 3,4-dimethyl 5-amino-isoxazole with salicylaldehyde and substituted salicylaldehydes. The newly synthesized ligands were characterized by IR, UV-Vis, ¹H NMR, ¹³C NMR, mass spectra, and elemental analysis. The Cu(II) complexes were characterized by IR, UV-Vis, ESR, elemental analysis, magnetic moments, thermogram, DTA, and single crystal analysis. The complexes have general formula [M(L)₂]. The Schiff bases are bidentate coordinating through the azomethine nitrogen and phenolic oxygen of salicylaldehydes. Based on the analytical and spectral data, four-coordinate geometry is assigned for all the complexes. ESR and single crystal analysis suggests square planar geometry for all complexes. [Cu(DMIIMP)₂] crystallizes in the orthorhombic system. Antimicrobial studies of Schiff bases and their metal complexes show significant activity with the metal complexes showing more activity than corresponding Schiff bases. Cytotoxicity of the copper complexes on human cervical carcinoma cells (HeLa) was measured using the Methyl Thiazole Tetrazolium assay.     

Concise Synthesis and Antimicrobial Activities of New Substituted 5‐Isoxazolpenicillins

The synthesis of a series of new 5‐isoxazolpenicillins is described, which were obtained by coupling substituted isoxazoles with 6‐APA. Concise large‐scale synthesis of 3,5‐disubstituted isoxazoles by 1,3‐dipolar cycloaddition using copper(I) as catalyst was also investigated. Representative compounds were assayed for antimicrobial activities, showing satisfactory antimicrobial activities against Gram‐negative bacteria.     

新型甘草次酸异噁唑衍生物的合成

以甘草次酸(脱氧甘草次酸)和3-取代苯基-5-氨甲基-异噁唑为原料, 合成了4种甘草次酸异噁唑衍生物, 通过IR, 1H NMR, 13C NMR及FABMS等方法确定了化合物的结构. 同时用L9(34) 正交试验对酰胺化反应的条件进行优化, 确定了酰化反应的最佳条件.     

Synthesis of novel isoxazole-linked steroidal glycoconjugates—an application of a novel steroidal nitrile oxide

Isoxazole-linked steroidal glycoconjugates are prepared by 1,3-dipolar cycloaddition reactions of an in situ generated and hitherto unknown steroidal nitrile oxide with appropriate propargyl ethers of sugars. The methodology provides a novel vector in the form of an easily accessible nitrile oxide having the ability to couple with many biomolecules, thus offering a new pathway to construct biologically significant novel steroidal conjugates.     

Base‐Catalyzed Formation of Isoxazoles from Dialkyl Acetylenedicarboxylates and 2‐Nitroacetophenones

The reaction of dialkyl acetylenedicarboxylates with 1‐aryl‐2‐nitroethanones in the presence of pyridine leads to dialkyl 5‐aryloyl‐isoxazole‐3,4‐dicarboxylates through a novel mechanism, which involves a [2+2] cycloaddition/ring opening/cyclization sequence.
