Preliminary Contribution to the Quantification of HMF in Honey by Electrochemical Biosensor Chips

Several hydroximethylfurfural (HMF) microbiosensors, based on a microelectrode configuration built by photolithographic techniques, have been developed. Two different biosensors based on the aldehyde dehydrogenase (ALDH) enzyme coupling with NADH oxidase (NOD) or diaphorase (Diaph) are described. Experimental variables were optimized using experimental design methodology, by central composite designs, taking the intensity registered for a given HMF solution as response. Given the reproducibility (residual standard deviation (RSD), 7.20%), repeatability (RSD=3%) and limit of detection (LOD)=1.68×10^?4 mol dm^?3 (α=β=0.05 and a replicate) reached by the ALDH‐Diaph microbiosensor under the optimum conditions, the analysis of HMF in honey was successfully accomplished.     

Implementation of an integrated glucose POCT system

In response to a change of the Belgian National Directives whereby hospital laboratories became responsible for all point-of-care testing (POCT) performed within hospital walls a standardized and automated POC glucose-testing system was implemented in our hospital. The system consists of 50 AccuCheck Inform instruments (Roche Diagnostics, Vilvoorde, Belgium), 50 docking stations, a DataCare Server, and connections to the medical laboratory information system (MOLIS, Sysmex, Barchon, Belgium) and to the hospital information system. Implementation involved many parties and extensive preparation and communication. Key issues were bar-coded patient and user identification, training, and responsibilities. One year after the hospital wide implementation of this system the quality of POC glucose testing has significantly increased, thereby improving patient safety. This study describes a stepwise change over involving the medical laboratory and with a focus on hands-on quality.Presented at the ninth conference on Quality in the Spotlight, 18–19 March 2004, Antwerp, Belgium.     

吡罗红为底物的辣根过氧化物酶催化荧光反应测定葡萄糖

吡罗红在辣根过氧化物酶催化下可被过氧化氢氧化而使其荧光猝灭。在pH 7.2中性介质中,稳态催化速率由酶和底物浓度决定,催化体系服从Michaelis-Menten方程,用Lineweaver-Burk作图法求得米氏常数、最大反应速度、催化常数分别为2.4×10~(-5)mol·L-1,2.5×10~(-6)mol·L-1s-1,75.8 s-1。在最佳反应条件下,荧光F0/F的猝灭程度与过氧化氢浓度在0-3.6×10~(-7)mol·L-1范围内成线性关系,检测限为6.3×10~(-9)mol·L-1;当与葡萄糖氧化酶联用时,可定量检测葡萄糖,线性关系为0-8.0×10~(-7)mol·L-1,检测限为3.4×10~(-8)mol·L-1。方法用于分析人血清中葡萄糖含量,分析结果与苯酚-4-氨基安替比林法基本一致。     

Application of enzyme-field effect transistor sensor arrays as detectors in a flow-injection analysis system for simultaneous monitoring of medium components. Part II. Monitoring of cultivation processes

Glucose, maltose, sucrose, lactose, ethanol and urea concentrations were monitored simultaneously during the cultivation of Escherichia coli and Saccharomyces cerevisiae by means of enzyme field effect transistors (EnFETs) applying glucose dehydrogenase (GDH), maltase (MAL)/GDH, invertase (INV)/GDH, β-galactosidase (β-GAL)/galactosedehydrogenase (GALDH), alcoholdehydrogenase (ADH)/aldehydedehydrogenase (ALDH), and urease. These enzymes were (co)immobilized on the pH sensitive gates of an eight-FET array. The FET array was integrated in a commercial FIA system.     

伏安型植物组织生物微电极的研究——茄子组织碳糊微电极的研究

茄子组织生物微电极在儿茶酚测定中表现出了较高的生物催化活性,而对抗坏血酸测定则为稳态响应,这表明该电极能有效地消除抗坏血酸的干扰。该电极的灵敏度高,重现性好,对多巴胺测定的线性范围为5.8×10~(-6)～6.5×10~(-4)mol/L,检出下限为:2.9×10~(-6)mol/L。     

一种新的乙酰胆碱酶电极的研究

本文制备了一种以溶剂聚合膜ＰＨ电极作原电极，经高碘酸钠活化的醋酸纤维膜固定乙酰胆碱酯酶的乙酰胆碱生物电极。研究了酶固定化条件的影响，测得静态和动力学条件下的线性中响应范围分别为２４０－１３００μｇ／ｍＬ和３００－１７００μｇ／ｍＬ将电极用于实际样品的回收率试验中，获得了满意的结果。     

Electrodeposited Nickel/Platinum Alloy as a Biosensor for Acetyl Choline

Nickel and platinum find extensive use in preparation of biosensors. In the present work, Ni/Pt alloy was plated on graphite to make acetylcholine sensor. The microstructure, surface composition and electrochemical performance of the electrode was analyzed by different techniques. The sensing performance was evaluated by cyclic voltammetry. The prepared alloy plate exhibited very good linear relationship between acetyl choline concentration and response current. The sensitivity and reproducibility of the prepared electrode were found better than other nickel electrodes reported.

Evanescent fluorobiosensor for the detection of polyaromatic hydrocarbon based on DNA intercalation

A flow-injection analysis (FIA) system coupled with an evanescent wave (EW) Biosensor employing total internal reflection of fluorescence radiation (TIRF) for the detection of polyaromatic hydrocarbon that intercalates into DNA is reported. A highly fluorescent intercalator, “ethidium bromide,” has been used as the reference compound for detection. The EW Biosensor was developed according to the procedure described earlier (1,2). Data on the analysis of Naphthalene, 3-methy cholanthrene, 7,12-dimethylbenz(a)anthracene, 1,2-benzanthracene, and some standard reference materials supplied by the National Institute of Standards and Technology are reported. The relative ability of the polyaromatic hydrocarbon to displace ethidium bromide, based on the relative binding ratio, is found to be on the order of 7,12-dimethylbenz[a]anthracene > 3-methylcholanthrene > 1,2-benzanthracene > napthalene.
