报导了Yb :FAP和Yb :C3S2 _FAP晶体在不同温度下的吸收光谱和荧光光谱实验结果 ,研究发现由于电子_声子近共振耦合作用 ,Yb :FAP和Yb :C3S2 _FAP晶体均存在有明显的振动谱 ,Yb :FAP晶体的零声子线在低温下还劈裂为相差 10cm- 1 的两条线 .采用激光选择激发技术研究了Yb3 离子在FAP和C3S2 _FAP晶体中的格位特征 ,结果表明Yb3 离子在这两种基质中都只占据Ca(Ⅱ )格位 ,但由于CaF2 的挥发 ,导致了Ca(Ⅱ )格位的局部畸变 .  相似文献

Fibroblast activation protein (FAP) is expressed in the microenvironment of most human epithelial tumors. 68Ga-labeled FAP inhibitors based on the cyanopyrrolidine structure (FAPI) are currently used for the detection of the tumor microenvironment by PET imaging. This research aimed to design, synthesize and preclinically evaluate a new FAP inhibitor radiopharmaceutical based on the 99mTc-((R)-1-((6-hydrazinylnicotinoyl)-D-alanyl) pyrrolidin-2-yl) boronic acid (99mTc-iFAP) structure for SPECT imaging. Molecular docking for affinity calculations was performed using the AutoDock software. The chemical synthesis was based on a series of coupling reactions of 6-hidrazinylnicotinic acid (HYNIC) and D-alanine to a boronic acid derivative. The iFAP was prepared as a lyophilized formulation based on EDDA/SnCl2 for labeling with 99mTc. The radiochemical purity (R.P.) was verified via ITLC-SG and reversed-phase radio-HPLC. The stability in human serum was evaluated by size-exclusion HPLC. In vitro cell uptake was assessed using N30 stromal endometrial cells (FAP positive) and human fibroblasts (FAP negative). Biodistribution and tumor uptake were determined in Hep-G2 tumor-bearing nude mice, from which images were acquired using a micro-SPECT/CT. The iFAP ligand (Ki = 0.536 nm, AutoDock affinity), characterized by UV-Vis, FT-IR, 1H–NMR and UPLC-mass spectroscopies, was synthesized with a chemical purity of 92%. The 99mTc-iFAP was obtained with a R.P. >98%. In vitro and in vivo studies indicated high radiotracer stability in human serum (>95% at 24 h), specific recognition for FAP, high tumor uptake (7.05 ± 1.13% ID/g at 30 min) and fast kidney elimination. The results found in this research justify additional dosimetric and clinical studies to establish the sensitivity and specificity of the 99mTc-iFAP.  相似文献

A continuous-wave laser oscillation was demonstrated with a Yb-doped fluorapatite (FAP) crystal pumped with a 905 nm Ti:sapphire laser. The output characteristics for different output couplers were investigated. A maximum power of 67mW at a 1043nm wavelength was obtained with 10% output coupler, pumped by a Ti:sapphire laser with 910mW power. This corresponds to a slope efficiency of 26.5%.  相似文献

The spectroscopic performance of Yb-doped calcium fluorapatite (Yb:FAP) was assessed by measuring polarization absorption spectra, polarization emission spectra and fluorescence lifetime. The emission cross section has been evaluated using the absorption cross section and principle of reciprocity. The π polarization absorption and emission cross sections of Yb3+ in FAP are found to be substantially larger than those of other Yb-doped media. Favorable spectroscopic parameters show that Yb:FAP crystal is more desirable for many diode-pumped solid state laser application with low- and medium-power in the near future.  相似文献

The Yb3+ laser emitting at a 1.03μm wavelength has been evoking strong interest recently due to its advantages of long fluorescence lifetime, broad absorption band and the fact that it never shows concentration quenching. On the other hand, as a laser of three-level system it has, in general, a relatively high threshold power, which makes it important to seek some suitable host crystals to reduce this. Here, we present a comparison of the lasing performances of Yb3+-doped YAG [Y3Al5O12], FAP [Ca5(PO4)3F] and KYW [KY(WO4)2] crystals, including threshold power and slope efficiency, with those of the Nd:YAG laser based on the threshold formula of three-and four-level systems deduced by the authors. The results show that the Yb3+ laser can output a power larger than the Nd:YAG laser does in the case of comparably higher pumping power, if the length of the lasing rod and the concentration of the active ions satisfy some conditions. The theoretical results are also close to the experimental results reported.  相似文献

Ionic liquids (ILs) containing the tris(pentafluoroethyl)trifluorophosphate anion [FAP] have attracted increased attention due to their unique properties including ultrahigh hydrophobicity, hydrolytic stability, and wide electrochemical window. In this study, the solvation parameter model is used via gas chromatography to characterize the solvation interactions of seven ILs containing amino, ester, and hydroxyl functional groups appended to the cation and paired with [FAP], as well as three ILs containing the bis[(trifluoromethyl)sulfonyl]imide anion [NTf2]. The role of the functional groups, nature of the counter anion, and cation type on the system constants were evaluated. ILs containing [FAP] possessed lower hydrogen bond basicity than NTf2-based ILs having the same cationic component; in the case of hydroxyl-functionalized cations, the presence of [FAP] led to an enhancement of the hydrogen bond acidity, relative to the NTf2-analogs. The system constants support the argument that [FAP] weakly coordinates the cation and any appended functional groups, promoting properties of the cation which might be masked by stronger interactions with other anion systems. The chromatographic performance of the IL stationary phases was evaluated by examining the retention behavior and separation selectivity for chosen analytes. The results from this work can be used as a guide for choosing FAP-based ILs capable of exhibiting desired solvation properties while retaining important physical properties including high thermal stability and high hydrophobicity. Recently, the first squaramide-(SA) containing FAP inhibitor-derived radiotracers were introduced. DATA5m.SA.FAPi and DOTA.SA.FAPi with their non-radioactive complexes showed high affinity and selectivity for FAP. After a successful preclinical study with [68Ga]Ga-DOTA.SA.FAPi, the first patient studies were realized for both compounds. Here, we present a new squaramide-containing compound targeting FAP, based on the AAZTA5 chelator 1,4-bis-(carboxylmethyl)-6-[bis-(carboxymethyl)-amino-6-pentanoic-acid]-perhydro-1,4-diazepine. For this molecule (AAZTA5.SA.FAPi), complexation with radionuclides such as gallium-68, scandium-44, and lutetium-177 was investigated, and the in vitro properties of the complexes were characterized and compared with those of DOTA.SA.FAPi. AAZTA5.SA.FAPi and its derivatives labelled with non-radioactive isotopes demonstrated similar excellent inhibitory potencies compared to the previously published SA.FAPi ligands, i.e., sub-nanomolar IC50 values for FAP and high selectivity indices over the serine proteases PREP and DPPs. Labeling with all three radiometals was easier and faster with AAZTA5.SA.FAPi compared to the corresponding DOTA analogue at ambient temperature. Especially, scandium-44 labeling with the AAZTA derivative resulted in higher specific activities. Both DOTA.SA.FAPi and AAZTA5.SA.FAPi showed sufficiently high stability in different media. Therefore, these FAP inhibitor agents could be promising for theranostic approaches targeting FAP.  相似文献

Transthyretin (TTR) is a cerebrospinal fluid and plasma prevalent protein implicated in heritable and sporadic amyloidosis. Numerous mutations and a wide range of phenotypes have been associated with TTR-mediated amyloidosis. Among these, V30 M is the most predominant point mutation, inculpated with familial amyloid polyneuropathy (FAP), a life-threatening autosomal dominant genetic disorder characterized by the deposition of amyloid fibrils in crucial areas. Hence, efficacious therapeutics against this detrimental disorder is warranted. Lately, several peptide-based analeptics, especially the ones that are aggregation-prone and the ones derived from aggregation hotspots of amyloidogenic proteins are being increasingly proffered against the amyloid fibrils. In the present study, as an effective precursor to in vitro investigations, we examined and assessed the therapeutic potentials of aggregation-prone peptides (APPs) derived from TTR, against V30 M TTR amyloid fibrils, computationally. Out of five experimentally corroborated APPs availed for this study, molecular dynamics simulation analysis endorses APP TAVVTN to be an effective beta-sheet breaker against V30 M TTR amyloid fibrils. Furthermore, consistent findings from various molecular trajectory analyses, residual frustration analysis and simulated thermal denaturation have indicated that APP TAVVTN could effectually relater the structural dynamics of V30 M TTR amyloid fibrils, to conformationally digress it away from its amyloidogenic propensities. Hence, based on consistent unvarying findings from numerous adept computational pipelines, APP TAVVTN could be an efficacious analeptic to therapeutically intervene and mitigate the amyloidogenic propensities of V30 M TTR amyloid fibrils, thereby ameliorating the pathological ramifications due to FAP.  相似文献
