A molecularly imprinted polymer solid-phase extraction method is used to extract esculetin from the ash bark of Chinese traditional medicine. Ratio of ethanol and water as washing solution were investigated. Data of accumulative adsorption on molecularly imprinted polymers from the continuous loading experiment suggests that there are two different kinds of recognition sites in molecularly imprinted polymers. By selecting the washing and eluting solution a scheme was designed to separate esculetin and its analogues including esculin, coumarin, 7-methoxylcoumarin and daphnetin. Finally, by applying the revised scheme esculetin was extracted from the ash bark of Chinese traditional medicine that was purchased from two big drugstores, respectively, with both molecularly imprinted polymers and non-molecularly imprinted polymers.  相似文献

The interaction between esculin and serum albumins was investigated to illustrate that the fluorescence resonance energy-transfer (FRET) affects the determination of the binding constants obtained by fluorescence quenching method. The binding constants (Ka) obtained by the double-logarithm curve for esculin–BSA and esculin–HSA were 1.02 × 107 and 2.07 × 104 L/mol, respectively. These results from synchronous fluorescence showed that the Tyr and Trp residues of HSA were affected more deeply than those in BSA. The excitation profile of esculin showed that in the presence of BSA and HSA, the S0 → S1 transition of esculin () appears, which is similar to the of BSA and HSA. The critical distance (R0) between BSA and esculin is higher than that of HSA, which showed that the affinity of esculin and HSA should be higher than that of BSA. After centrifugation, the concentrations of esculin bound to albumins were determined by means of the fluorescence of esculin. It was found that much more esculin was bound to HSA than to BSA. However, the bound models for BSA and HSA are almost the same. The concentration of esculin bound to serum albumin at first decreased with the addition of esculin and then increased. From above results, it can be concluded that the affinity of esculin and HSA should be higher than that of esculin and BSA. This example showed that in the presence of FRET, the binding constants between ligands and proteins based on fluorescence quenching might be deviated.  相似文献

Four molecularly imprinted polymers (MIPs) were prepared in MeOH with esculin (=6,7‐dihydroxycoumarin 6‐(β‐D ‐glucopyranoside)=6‐(β‐D ‐glucopyranosyloxy)‐7‐hydroxy‐2H‐1‐benzopyran‐2‐one) as the imprinted molecule, methacrylic acid (=2‐methylprop‐2‐enoic acid; MAA), acrylamide (=prop‐2‐enamide; AM), 4‐vinylpyridine (=4‐ethenylpyridine; 4‐VP), or 2‐vinylpyridine (=2‐ethenylpyridine; 2‐VP) as the functional monomer, respectively, as well as ethylene glycol dimethacrylate (=2‐methylprop‐2‐enoic acid ethane‐1,2‐diyl ester; EGDMA) as the cross‐linking agent. The interaction between the template and the functional monomers was investigated by fluorescence and UV spectrophotometry, respectively, which revealed the presence of esculin/monomer complexes in the stoichiometric ratio 1 : 2 in the pre‐polymerization mixture. The resultant polymers were studied in equilibrium binding experiments to evaluate the recognition ability and the binding capacity towards esculin. The results showed that MIP1, prepared with MAA as the functional monomer, exhibited advantageous characteristics of high binding capacity, optimal imprinting effect, and good selectivity towards esculin. The Scatchard analysis indicated that there are two types of binding sites in MIP1, and its binding parameters including the apparent maximum numbers of binding sites and the dissociation constants were calculated. Finally, by packing an SPE column (SPE=solid‐phase extraction) with MIP1, the esculin was separated and enriched successfully by this sorbent from samples of Cortex fraxini, and the average recovery was up to 74.7%.  相似文献
