Because of the side effects and drug resistance of cisplatin, a basic clinically approved chemotherapeutic drug, a new attempt is reported to develop a novel antitumor drug based on complexation of iron metal ion with organic moiety that may be effective and safer. A newly synthesized iron(III) diacetylmonoxime‐2‐hydrazinopyridine complex was tested firstly for its cytotoxicity and superoxide dismutase (SOD)‐mimic activity in vitro then for its antitumor activity against Ehrlich ascites carcinoma (EAC) and the related biochemical alterations in vivo in comparison with cisplatin. The complex showed 80.88% SOD‐mimic activity and IC50 of 2.6 μg ml−1. In EAC‐bearing mice, in a dose‐dependent manner, Fe(III) complex treatment exhibited significant hematological profile improvements, tumor volume, viable cell count and hepatic lipid peroxidation level decreases, life span extension, hepatic glutathione and total antioxidant capacity levels enhancements, hepatic SOD and catalase activities augmentations, liver function tests alterations attenuations, and hepatocyte nucleic acids content normalization. Thus, the Fe(III) diacetylmonoxime‐2‐hydrazinopyridine complex is a novel, promising, less toxic antitumor agent. Its killing of tumor cells may be via a reactive oxygen species scavenging mechanism.  相似文献

A mononuclear copper (II) complex of N‐2‐hydroxyhippuric acid (2HHA), [Cu(HA)(H2O)2], has been synthesized and characterized by spectroscopic and X‐ray powder diffraction studies. Crystal structure of [Cu(HA)(H2O)2] reveals a distorted square‐pyramidal geometry around the metal center. The crystal packing in the complex exhibits a three‐dimensional framework formed by intermolecular O? ; H···O and C? H···O hydrogen bonds. Toxicity and antitumor properties of the complex have been studied in vivo. The complex, capable of depleting glutathione (GSH) at nontoxic doses, may be utilized to sensitize drug‐resistant cells where resistance is due to an elevated level of GSH. The anticancer effect of thiacarpine, a synthetic analogue of the known cytotoxic alkaloid polycarpine isolated from the Pacific ascidian Polycarpa aurata, was investigated in vivo in experiments using mouse solid Ehrlich carcinoma tumor as the target. A high-resolution magnetic resonance imaging (MRI) technique using a MR tomograph "PharmaScan" US70/16 (Bruker, Ettlingen, Germany) was used for visualization and quantification of tumor size. Fluorescence microscopy and image analysis were applied to determine Ehrlich carcinoma cell chromatin condensing (apoptosis) and necrosis in Ehrlich carcinoma cells at the action of thiacarpine in in vitro experiments. The scan and size calculations of the tumor and some mouse organs were carried out during the experiments. Thiacarpine in a total dose of 100 mg/kg was found to exhibit the delay in growth of the mouse tumor. The antineoplastic effect of this compound was accompanied by an increase in the lifetime of experimental mice in comparison with the control group of animals. Our data show that the ability of thiacarpine to induce apoptosis in carcinoma cells may contribute to thiacarpine anticancer effects against mice solid Ehrlich carcinoma in vivo detected by MRI.  相似文献

Seven different ferrocene derivatives have been tested in vitro against Ehrlich ascites tumor cells. Neither ferrocene nor the monosubstituted derivative N,N-dimethylaminomethylferrocene showed cytotoxic activity (IC50 > 1000 μM for 3 h treatments). Better results were obtained with 1,2-disubstituted derivatives. The IC50 values ranged from 376.6 μM for 1,2-diformylferrocene to 71.2 μM for racemic 2-(N,N-dimethylaminomethyl)ferrocenecarboxamide. The latter derivative was also encapsulated in native β-cyclodextrin (CD), heptakis-2,3,6-tri-O-methyl-β-CD (TRIMEB) and 2-hydroxypropyl-β-CD (HPβCD) to give 1:1 (host:guest) inclusion compounds. The existence of true inclusion complexes in the solid state was confirmed by a combination of powder X-ray diffraction, thermogravimetric analysis, FTIR and 13C CP MAS NMR spectroscopy. The IC50 value for the β-CD inclusion compound was identical to that obtained for the nonincluded ferrocene derivative. By contrast, the inclusion compounds comprising TRIMEB and HPβCD yielded IC50 values of 25.2 and 20.0 μM, respectively. No obvious relationship could be established between the redox behavior of the compounds determined by cyclic voltammetry and the biochemical data.  相似文献

The various parts of Zizyphus nummularia has been used traditionally in several disease conditions. However, its anticancer activity and mechanism of action remain to be elucidated. Considering this, the objective of this study was to isolate, identify and screen for possible anticancer activity in vitro and in vivo of the ethanolic extract (EE) and isolated identified compound (IC) from Z. nummularia root bark. The in vitro activity against human breast cancer, leukaemia, ovarian cancer, colon adenocarcinoma and human kidney carcinoma cell lines was determined. The in vivo activity in female Swiss albino mice against Ehrlich ascites carcinoma (EAC) was determined. The isolated compound is a new triterpene derivative. EE/IC showed cytotoxicity against different cell lines. The administration of EE/IC decreased tumour parameters such as tumour volume, viable tumour cell count and increased body weight, haematological parameters and life span in comparison to the EAC control mice.  相似文献

In order to investigate a possible relationship between the intensity of lipid peroxidation (LP) in tumor cells and their proliferative activity various methods to quantify LP are desirable. In this study the decrease in the contents of fatty acids and glutathione was measured by established methods inEhrlich ascites tumor (EAT) cellsin vitro, in which LP was stimulated by the addition of ferrous iron, either as free ion or as histidinate chelate.When EAT cells were incubated for 30 min at 37 °C in the presence of 5 mM FeSO4 the following changes were observed in comparison to appropriate control cells: The content of reduced glutathione (GSH) and total glutathione (GSH+2 GSSG) decreased significantly by 24 and 30% respectively. The decrease of 4 unsaturated (C 18:1; C 18:2; C 20:4; C 22:6) and 2 saturated fatty acids (C 16:0; C 18:0) by about 15% on the average was statistically significant only for C 16:0 and C 20:4).More pronounced effects were observed with 5 mM Fe(II)-histidinate. GSH and GSH+2 GSSG decreased by 54% and 40%, resp. The decrease of fatty acids by about 40% on the average was significant for all of the 6 fatty acids tested. These results are in agreement with previous studies on LP in EAT cells showing Fe(II)-histidinate to be a more powerful promoter of LP compared with free ferrous ion. The observation, that the content not only of GSH but also of total glutathione was decreased in iron-treated tumor cells is in contradiction to the hypothesis that GSH may act as a mere redox mediator of LP under the conditions used and points to a consumption of GSH by several possible pathways. The finding of decreased levels of unsaturated as well as saturated fatty acids in the presence of Fe(II)-histidinate underlines the extraordinary potency of iron as an initiator and catalyst of LP.This work was supported by the Association for International Cancer Research, St. Andrews, U.K.  相似文献

The cytotoxic effect of protoporphyrin IX disodium salt (PPIX) on isolated Ehrlich ascetic tumor (EAT) cells induced by ultrasound exposure was investigated. Tumor cells suspended in air-saturated phosphate buffer solution (PBS, pH 7.2) were exposed to ultrasound at 2.2 MHz for up to 60 s in the presence and absence of PPIX. The viability of cells was determined by a trypan blue exclusion test. The morphological changes of cells in SDT were observed by scanning electron microscope (SEM). And the sub-cellular localization of PPIX in EAT cells was detected by confocal laser scanning microscopy (CLSM). The ultrasonically-induced cell damage increased as PPIX concentration increased, while no cell damage was observed with PPIX alone. CLSM observation revealed that the fluorescence of PPIX and rhodamine 123 (mitochondrial probe) overlapped very well in the cytoplasm. The results indicate that PPIX could enhance the ultrasonically-induced cell damage and mitochondria may play an important role during sonodynamically induced cytotoxicity.  相似文献
