Fifty-eight phenolic compounds isolated from Phyllanthus emblica were screened and compared for their in vitro and in vivo antioxidant properties, as well as hyaluronidase (HAase) inhibitory activities. Among them, 20 compounds showed to be promising antioxidants due to the stronger scavenging activity in both DPPH radical and Danio rerio reactive oxygen species assays, while nine compounds were potential HAase inhibitors with 100-fold stronger activities than that of the positive control, DSCG. A caloric surplus and a sedentary lifestyle are undoubtedly known to be the leading causes of obesity. Natural products represent valuable allies to face this problematic issue. This study was planned to assess the effect of a white grape (Vitis vinifera) juice extract (WGJe) in diet-induced obese zebrafish (Danio rerio). Fish were divided into four different diet groups: (i) normally fed (NF); (ii) overfed (OF); (iii) WGJe-supplemented NF (5 mL/L in fish water); (iv) WGJe-supplemented OF. Body mass index (BMI) was extrapolated each week. After the fourth week, euthanized zebrafish were processed for both microscopic evaluations and gene expression analyses. OF zebrafish showed higher BMI values with respect to NF counterparts, an effect that was hindered by WGJe treatment. Moreover, histological analyses showed that the area of the adipose tissue, as well as the number, size, and density of adipocytes was significantly higher in OF fish. On the other hand, WGJe was able to avoid these outcomes both at the subcutaneous and visceral levels, albeit to different extents. At the gene level, WGJe restored the altered levels of ghrelin and leptin of OF fish both in gut and brain. Overall, our results support the anti-obesity property of WGJe, suggesting its potential role in weight management.  相似文献

The male gonads of Danio rerio occupy a position typical of the Teleostei species. The structure of the testes corresponds to the anastomosing tubular type with unrestricted spermatogonia and represents a cystic type of spermatogenesis. The results of this study indicate that four distinct stages of cell differentiation can be identified during spermiogenesis. These stages are characterised by chromatin condensation, the development of flagellum, nuclear rotation, the formation of nuclear fossa and the elimination of excess cytoplasm. A round head and the absence of an acrosome characterise the differentiated sperm. The midpiece is short and large, and C-shaped mitochondria form a ring surrounding the initial region of the flagellum. The axoneme shows a 9 + 2 pattern. In the D. rerio spermatozoa the flagellar axis is at an angle of 110° to the nucleus diameter running through the centriole.  相似文献

In this study, the vitellogenin (Vtg) modulation by genistein and polychlorinated biphenyl-126 (PCB-126) exposure in zebrafishes has been investigated. Both PCB-126 and genistein have been identified as aquatic pollutants and can further increase estrogenicity of waterways. Vtg is egg yolk precursor protein release by the hepatocytes during vitellogenesis. This process occurs normally in the hepatocytes in response to the activation with the estrogens such as 17-β-estradiol. Our immunohistochemical findings showed a Vtg expression that increases at 12 h and at 72 h in the liver of treated fishes with both PCB-126 and genistein, individually and in combination. Furthermore, for the first time, also hepatic stellate cells (HSC) in the liver parenchyma were strongly positive for vitellogenin.  相似文献

快速起动是鱼类重要的一种机动能力,有助于鱼类的逃逸和捕食。快速起动通常分为C形起动和S形起动两种模式。本文利用数字式高速摄像机定量地研究了斑马鱼从低速巡游状态做出捕食反应的运动过程。通过分析斑马鱼的运动图像,发现S形起动过程大体可以分解为三个阶段:第一阶段,斑马鱼的头部和尾部都迅速向相同的一个体侧方向转动,整个身体近似弯曲反写的S型。第二阶段,斑马鱼的头部转向捕食方向,尾部摆动1~2个周期,整个身体近似成S型。第三阶段整个鱼体逐渐恢复平直形态,其前端2/3的部分不再摆动,维持一种类似在水中滑翔的游动姿态。本文定量分析了各个阶段鱼体躯干曲线的形状,给出了鱼体的速度分布,并定性地解释了S形起动的力学机理。  相似文献

Trace metals such as Cadmium (Cd) and Zinc (Zn) are known to exhibit adverse effects on many aquatic organisms including early life stages of fish. In contact with contaminated sediment, fish eggs and embryos may be exposed to metals via the water phase as well as via direct contact with contaminated particles. This may result in body burdens that are difficult to predict and may vary according to individual micro scale exposure conditions. The highly sensitive total reflection X-ray fluorescence spectrometry (TXRF) may provide a tool to analyse individual embryos for internal contaminant concentrations and thus helps to develop a better understanding of dose–response relationships. To test this hypothesis, embryos of Danio rerio were exposed to Cd and Zn spiked sediment in different treatments applying an ion exchange resin for modification of bioavailable concentrations. The TXRF analysis indicated individual embryos with dramatically enhanced exposure compared to other individuals despite uniform exposure conditions on a macro scale. Ion exchanger reduced embryo Zn concentrations to values close to control value with a comparably low standard deviation. Cadmium concentrations in embryos were in the range of 4000 to 7000 µg/g with a median of 5740 µg/g. A commercial ion exchanger reduced individual body burdens by a factor 50 to 100. Individual peak body burdens of up to 3160 µg/g were accompanied by reduced weight of the fish eggs due to early death i.e. coagulation. The investigation of exposure and effects on an individual-based scale may significantly help to reduce uncertainty and inconsistencies occurring in conventional analysis of pooled fish embryo samples.  相似文献

A new alkaloid, geissospermiculatine was characterized in Geissospermum reticulatum A. H. Gentry bark (Apocynaceae). Here, following a simplified isolation protocol, the structure of the alkaloid was elucidated through GC-MS, LC-MS/MS, 1D, and 2D NMR (COSY, ROESY, HSQC, HMBC, 1H-15N HMBC). Cytotoxic properties were evaluated in vitro on malignant THP-1 cells, and the results demonstrated that the cytotoxicity of the alkaloid (30  μg/mL) was comparable with staurosporine (10  μM). Additionally, the toxicity was tested on zebrafish (Danio rerio) embryos in vivo by monitoring their development (0–72 h); toxicity was not evident at 30  μg/mL.  相似文献

近年来水污染事件频发, 给人类饮用水安全带来巨大隐患, 而生物监测法能够从生物学角度对水质状况做出综合分析. 选取斑马鱼作为受试生物, 当水质发生变化时, 鱼群在毒性物质作用下产生应激反应, 此时运用图像处理技术量化鱼类群体行为参数; 并用带特征染色体的遗传算法与支持向量机相融合的方法进行特征选择和SVM参数优化, 分析影响水质预警的关键量化参数. 结果显示, 新方法与网格搜索法、不带特征染色体的遗传算法与SVM结合的方法相比较, 有更高的准确率.  相似文献

Large doses of ionizing radiation can damage human tissues. Therefore, there is a need to investigate the radiation effects as well as identify effective and non-toxic radioprotectors. This study evaluated the radioprotective effects of Kelulut honey (KH) from stingless bee (Trigona sp.) on zebrafish (Danio rerio) embryos. Viable zebrafish embryos at 24 hpf were dechorionated and divided into four groups, namely untreated and non-irradiated, untreated and irradiated, KH pre-treatment and amifostine pre-treatment. The embryos were first treated with KH (8 mg/mL) or amifostine (4 mM) before irradiation at doses of 11 Gy to 20 Gy using gamma ray source, caesium-137 (137Cs). Lethality and abnormality analysis were performed on all of the embryos in the study. Immunohistochemistry assay was also performed using selected proteins, namely γ-H2AX and caspase-3, to investigate DNA damages and incidences of apoptosis. KH was found to reduce coagulation effects at up to 20 Gy in the lethality analysis. The embryos developed combinations of abnormality, namely microphthalmia (M), body curvature and microphthalmia (BM), body curvature with microphthalmia and microcephaly (BMC), microphthalmia and pericardial oedema (MO), pericardial oedema (O), microphthalmia with microcephaly and pericardial oedema (MCO) and all of the abnormalities (AA). There were more abnormalities developed from 24 to 72 h (h) post-irradiation in all groups. At 96 h post-irradiation, KH was identified to reduce body curvature effect in the irradiated embryos (up to 16 Gy). γ-H2AX and caspase-3 intensities in the embryos pre-treated with KH were also found to be lower than the untreated group at gamma irradiation doses of 11 Gy to 20 Gy and 11 Gy to 19 Gy, respectively. KH was proven to increase the survival rate of zebrafish embryos and exhibited protection against organ-specific abnormality. KH was also found to possess cellular protective mechanism by reducing DNA damage and apoptosis proteins expression.  相似文献

Lysine acetylation is an important post‐translational modification (PTM). Since the development of MS‐based proteomics technology, important roles of lysine acetylation beyond histones have focused on chromatin remodeling during the cell cycle and regulation of nuclear transport, metabolism, and translation. Zebrafish (Danio rerio) is a widely used vertebrate model in genetics and biologic studies. Although studies in several mammalian species have been performed, the mechanism of lysine acetylation in D. rerio embryos is incompletely understood. Here, we investigated the global acetylome in D. rerio embryos by using an MS‐based proteomics approach. We identified 351 acetylated peptides and 377 nonredundant acetylation sites on 189 lysine‐acetylated proteins in 5‐day postfertilization (hpf) embryos of D. rerio. Among lysine‐acetylated peptides, 40.2% indicated three motifs: (ac)KxxxK, (ac)KxxxxK, and Lx(ac)K. Of 190 acetylated proteins, 81 (42.6%) were mainly distributed in the cytoplasm. Gene ontology enrichment and Kyoto Encyclopedia of Genes and Genomes pathway analyses showed that lysine acetylation in D. rerio was enriched in metabolic pathways. Additionally, 17 of 30 acetylated ribosomal proteins were evolutionarily conserved between zebrafish and humans. Our results indicate that acetyllysine might have regulatory effects on ribosomal proteins involved in protein biosynthesis.  相似文献
