Applications of chromatographic and atomic absorption methods are compared with those of electroanalytical techniques. The limitations of electroanalytical procedures and their advantages, such as speed, sensitivity and speciation are discussed. Among applications of polarography and voltammetry to a solution of basic problems, attention has been paid to the possibility to distinguish the degree of conjugation (e.g. of compounds containing the grouping CNNC), to a contribution to understanding of solution chemistry of mitomycin C and to processes involved in the reduction of cephalosporins. Among practical applications, the possibility to analyze suspensions, which can be carried out without separation offers advantages. Examples are interactions in suspensions of bile salts in the presence of metal (II) ions, adsorptions of various pesticides and other toxic materials on lignin and the possibility to follow alkaline cleavage of lignin in buffers pH 8-12 at 25 °C. Most of these studies would not be possible to carry out using other techniques.  相似文献

The electrocatalytic oxidation of cephalexin and cefazolin has been studied at a carbon paste electrode modified with cobalt salophen (CoSal) by cyclic voltammetry. The selectivity of the carbon paste modified with CoSal in detecting cephalexin and cefazolin was examined. To suggest the electrocatalytic mechanism for electro-oxidation of cefazolin, the electrochemical behavior of ceftriaxone was investigated which has a thiol group out of the beta lactam ring. The electrocatalytic oxidation of these antibiotics is shown to be irreversible at the CoSal modified electrode. Scan rate dependence of cefazolin, which is a sulfur-containing compound, has been examined. The results indicated that the electrocatalytic oxidation of the compounds is diffusion controlled. The responses of the modified electrode were compared with those of unmodified electrode and it has shown that the modified electrode has better sensitivity than unmodified electrode to the detection of cefazolin. The overall number of electrons contributed to the oxidation of cefazolin is obtained 1 by chronoamperometry; the number of electron involved in the rate-determining step was 1. The results of differential pulse voltammetry (DPV) using the modified electrode with high sensitivity were applied for the determination of cefazolin in human synthetic serum samples. The linear range was obtained from 1 × 10−5 to 1 × 10−3 M for DPV determination of cefazolin in buffered solutions (pH 3.0).  相似文献

A sensitive and selective confirmatory method for milk‐residue analysis of ten quinolones and eight cephalosporins by LC‐MS/MS has been developed herein. For the chromatographic separation of target analytes, a Perfectsil ODS‐2 (250 × 4 mm, 5 μm) analytical column was used and gradient elution was applied, using a mobile phase of 0.1% w/w TFA in water and 0.1% w/w TFA in ACN. Ultrasound‐assisted matrix solid‐phase dispersion procedure was applied for the extraction and clean‐up procedure of antimicrobials agents from milk matrix using a mixture of Bond Elut Plexa sorbent and QuEChERS. The method was validated meeting the European Legislation determining selectivity, linearity response, trueness, precision (repeatability and between‐day reproducibility), decision limit, detection capability, and ruggedness following the Youden approach. Recoveries of all antibiotics ranged from 81.7 to 117.9%, while RSD values were lower than 13.7%. Limits of quantification for all examined compounds ranged from 2.4 to 15.0 μg/kg, substantially lower than the maximum residue limits established by the European Union (30–100 μg/kg).  相似文献

A sensitive and specific liquid chromatographic–tandem mass spectrometric (LC–MS–MS) method for most of those penicillins and cephalosporins for which EU maximum residue limits (MRL) were set in Council regulation (EEC) 2377/90 was developed and validated in bovine muscle, kidney and milk. The analytes were extracted with acetonitrile/water and cleaned-up by a single reversed-phase solid-phase extraction step. Highest sensitivity for the analytes was obtained when amoxicillin, ampicillin, cephalexin, cephapirin, desacetylcephapirin, cephalonium, cefquinome and cefazolin were measured in the positive electrospray ionisation mode (ESI (+)) and cefoperazone, benzylpenicillin, phenoxymethylpenicillin, oxacillin, cloxacillin, dicloxacillin and nafcillin in the negative electrospray ionisation mode (ESI (−)). Chromatography was performed with a formic acid/methanol gradient. Collision-induced dissociation (CID) with argon was used for fragmentation of the pseudomolecular ions to achieve the required specificity. Possible adverse matrix effects on the electrospray ionisation process caused by co-eluting matrix components were investigated. The method was validated closely to the new EU guidelines and applied to positively screened samples from official food control allowing the identification and quantification of the residual β-lactams.  相似文献

This study investigates an off-line solid phase extraction (SPE) for improving the sensitivity in the capillary electrophoretic (CE) analysis of four cephalosporins. Two sorbents—LiChrolut-C18 and Oasis HLB—were used in a SPE process to detect cephalosporins in natural waters (tap, river and hospital sewage) and their performances were compared. By using Oasis HLB sorbent higher recoveries for river water were obtained (94–107% when 500 mL of sample were analyzed). The off-line SPE–CZE method was validated for river water with good detection limits (3 μg L−1) and the linearity ranged between 5 and 200 μg L−1.  相似文献

基于KMnO4在酸性介质中氧化头孢氨苄和头孢拉定产生微弱的化学发光,同时借助甲醛对发光的增强作用,采用流动注射技术,建立了以上两种头孢类抗生素药物的测定新方法。方法检出限分别为0.096和0.10μg mL;相对标准偏差分别为1.6%和1.8%(n=11),线性范围分别为0.8~25和0.8~32μg mL,进样频率为60次 h。本方法已用于药物制剂的测定。  相似文献

This paper describes our research on the degradation of ceftiofur and cephapirin at physiological temperatures in kidney extract and in alkaline and acidic solution, conditions that regularly occur during sample preparation. Degradation products were identified using LC–ToF/MS, NMR and microbiological techniques. Additionally kinetics of the degradation processes were studied. A slight instability of cephapirin and desfuroylceftiofur was observed at elevated temperatures. Ceftiofur and cephapirin degraded immediately and completely in an alkaline environment, resulting in inactive degradation products. Ceftiofur and cephapirin also degraded immediately and completely in kidney extract resulting in both formerly reported metabolites as well as not previously reported products. Our research shows that conditions often occurring during the analysis of ceftiofur or cephapirin result in rapid degradation of both compounds. From this it is concluded that underestimation of the determined amounts of ceftiofur and cephapirin is likely to occur. Therefore, a new approach is needed for the analysis of both compounds newly identified degradation products.  相似文献

Disposable amperometric magnetosensors, involving a mixture of modified-magnetic beads (MBs), for the multiplex screening of cephalosporins (CPHs), sulfonamides (SAs) and tetracyclines (TCs) antibiotic residues in milk are reported for the first time in this work. The multiplexed detection relies on the use of a mixture of target specific modified magnetic beads (MBs) and application of direct competitive assays using horseradish peroxidase (HRP)-labeled tracers. The amperometric responses measured at −0.20 V vs. the Ag pseudo-reference electrode of screen-printed carbon electrodes (SPCE) upon the addition of H2O2 in the presence of hydroquinone (HQ) as redox mediator, were used to monitor the extent of the different affinity reactions. The developed methodology, involving a simple and short pretreatment, allowed discrimination between no contaminated UHT and raw milk samples and samples containing antibiotic residues at the maximum residue limits (MRLs). The usefulness of the multiplexed magnetosensor was demonstrated by analyzing spiked milk samples in only 5 min. The results demonstrated that a clear discrimination of milk samples contaminated with antibiotics at their MRL level or their mixtures, allowing the identification of milk not complying with current legislation. These features make the developed methodology a promising alternative in the development of user-friendly devices for on-site analysis to ensure quality control for dairy products.  相似文献

We present a new system for the sensitive analysis of cephalosporins by CE using both on-line SPE and large-volume sample stacking (LVSS). Sample volumes of 250 muL were loaded onto the SPE microcolumn which was then desorbed with 426 nL of ACN. The SPE elution plug was injected into the CE system via an in-line valve interface filling approximately 60% of the volume of the separation capillary. Subsequently, LVSS was performed by applying a voltage of -5 kV, which resulted in the simultaneous removal of the elution solvent and the preconcentration of the analytes in a narrow zone. This way the amount of analyte loaded into the capillary could be considerably increased without serious loss of CE separation efficiency. LODs for cefoperazone and ceftiofur were in the ng/L range which represents an improvement of a factor of 8450 and 11 450 when compared with direct CE injection. The cephalosporin test compounds presented a good linear response (corrected peak area) between 0.5 and 10 mug/L with correlation coefficients higher than 0.995. The final method is compared with previously reported LVSS-CE and SPE-CE systems for the analysis of cephalosporins.  相似文献

In pH 2.8-3.8 BR buffer medium, the third generation cephalosporin antibiotics (TGCs) such as ceftazidime (CZD), ceftriaxone (CTRX), cefoperazone (CPZ), and cefotaxime (CFTM) react with palladium(II) (Pd(II)) to form 1:2 yellowish-brown cationic chelates, which further react with 4, 5-dibromofluorescein (DBF) to form 1:3 brown ion-association complexes. As a result, not only the spectra of absorption and fluorescence are changed, but also the resonance Rayleigh scattering (RRS) is enhanced greatly and the new RRS spectra are observed. The four TGCs products have similar spectral characteristics and their maximum RRS wavelengths are all located at 291 nm. The quantitative determination ranges and the detection limits of the four TGCs are 0.0065-1.0 μg mL−1 and 2.0 ng mL−1 for CZD, 0.0070-1.1 μg mL−1 and 2.2 ng mL−1 for CTRX, 0.0090-1.6 μg mL−1 and 2.7 ng mL−1 for CPZ, and 0.014-2.2 μg mL−1 and 4.2 ng mL−1 for CFTM, respectively. The optimum conditions of the reactions and the effects of foreign substances are investigated, and the composition of ion-association complexes is discussed also. Based on the ion-association reaction, a highly sensitive, simple and rapid method has been proposed to the determination of TGCs.  相似文献
