The activity of cellulase has traditionally been described by pH and temperature; however, the buffering medium is also an important factor, Taking plain water as a reference medium, three kinds of buffer including KH2PO4/K5HPO4, citric acid/sodium citrate, and acetic acid/sodium acetate were adopted to survey their effects on the activity of cellulase. Chromatographic assays indicated that xylose, glucose, and cellobiose were the major products and that minor products such as cellotriose and cellotetraose were present in some cases. The activities of cellulase based on glucose production showed that the phosphate buffer acted as a deactivator for cellulase and each of the two organic acid buffers acted as activators for cellulase. The concentration of activation buffer should be high to reach a high cellulase activity; however, this effect would be compensated for by the product inhibition of cellulase. The highest activity obtained was 4.16 ± 0.08 (× 10?3) IU mg?1 for the citric acid/sodium citrate buffer under pH 4.80, 40 °C and an agitation speed of 150 rpm.  相似文献

Diaminocarboxylic acid carrier ampholytes, such as L-histidine, 2,3-diaminopropionic acid, L-ornithine, and L-lysine, were reacted with glycerol-1,3-diglycidyl ether (GDGE) and poly(vinyl alcohol) (PVA) in the presence of sodium hydroxide to produce hydrolytically and mechanically stable hydrogels, supported on a PVA substrate, for use as buffering membranes in isoelectric trapping (IET) separations. The pH values of the DACAPVA membranes were determined with the help of small-molecule pI markers and proteins and were found to be in the 6 < pH < 8.5 range. The membranes were successfully used to isoelectrically trap small ampholytes, desalt ampholyte solutions in IET mode, and effect the binary separation of chicken egg white proteins.  相似文献

This paper introduces a class of unit-linked annuities that extends existing annuities by allowing portfolio shocks to be gradually absorbed into the annuity payouts. Consequently, our new class enables insurers to offer an affordable and adequate annuity with a stable payout stream. We show how to price and adequately hedge the annuity payouts in a general financial environment. In particular, our model accounts for various stylized facts of stock returns such as asymmetry and heavy-tailedness. Furthermore, the generality of our framework makes it possible to explore the impact of a parameter misspecification on the annuity price and the hedging performance.  相似文献

The landing buffer is an important problem in the research on bionic locust jumping robots, and the differ-ent modes of landing and buffering can affect the dynamic performance of the buffering process significantly. Based on an experimental observation, the different modes of landing and buffering are determined, which include the different numbers of landing legs and different motion modes of legs in the buffering process. Then a bionic locust mechanism is established, and the springs are used to replace the leg muscles to achieve a buffering effect. To reveal the dynamic performance in the buffering process of the bionic locust mechanism, a dynamic model is established with different modes of landing and buffering. In particular, to analyze the buffering process conveniently, an equivalent vibration dynamic model of the bionic locust mechanism is proposed. Given the support forces of the ground to the leg links, which can be obtained from the dynamic model, the spring forces of the legs and the impact resistance of each leg are the important parameters affecting buffering performance, and evaluation principles for buffering performance are proposed according to the aforementioned parameters. Based on the dynamic model and these evaluation principles, the buffer-ing performances are analyzed and compared in different modes of landing and buffering on a horizontal plane and an inclined plane. The results show that the mechanism with the ends of the legs sliding can obtain a better dynamic per-formance. This study offers primary theories for buffering dynamics and an evaluation of landing buffer performance, and it establishes a theoretical basis for studies and engineer-ing applications.  相似文献

This paper reports a voltammetric study of bronze in synthetic seawater (SSW). The effects of buffering and deoxygenating were particularly visible in the transpassive region. The breakdown of the anodic passive film on bronze leads to a well-defined activation peak in the transpassive region typical of a nucleation and growth of pits. The breakdown potential of the passivity was shown to vary with the experimental conditions, namely, with buffering and deoxygenating. Buffering has shown to lead to more stable passive films and deoxygenating to higher oxidation currents. Scanning electron microscopy with energy dispersive spectrometer (SEM/EDS) studies of bronze samples with 1-month exposure in non-deoxygenated buffered and non-buffered SSW under open circuit potential have shown significant differences in their morphology: a uniformly cracked surface and a surface showing large and spherical precipitates of about 50 μm uniformly distributed along the surface, respectively, for bronze coupons in buffered (pH 9) and in non-buffered SSW. The EDS technique has identified Cu, O, Cl and Na on the corrosion products of bronze in non-buffered SSW, whilst in buffered media, Sn was also identified. In non-buffered media, open circuit potentials have shown to be all the time less negative than in the buffered media. After 1-month exposure the E OCP of bronze samples in both media seem to converge to −0.131 and −0.155 V vs Ag|AgCl, respectively. This potential can be assigned to the formation of cuprite, Cu2O and nantokite, CuCl. The analysis of the SEM images after the removal of the corrosion products has shown descuprification with higher intensity on the surface from coupons in non-buffered SSW.  相似文献

The single alpha helix (SAH) is a recurring motif in biology. The consensus sequence has a di-block architecture that includes repeats of four consecutive glutamate residues followed by four consecutive lysine residues. Measurements show that the overall helicity of sequences with consensus E4K4 repeats is insensitive to a wide range of pH values. Here, we use the recently introduced q-canonical ensemble, which allows us to decouple measurements of charge state and conformation, to explain the observed insensitivity of SAH helicity to pH. We couple the outputs from separate measurements of charge and conformation with atomistic simulations to derive residue-specific quantifications of preferences for being in an alpha helix and for the ionizable residues to be charged vs. uncharged. We find a clear preference for accommodating uncharged Glu residues within internal positions of SAH-forming sequences. The stabilities of alpha helical conformations increase with the number of E4K4 repeats and so do the numbers of accessible charge states that are compatible with forming conformations of high helical content. There is conformational buffering whereby charge state heterogeneity buffers against large-scale conformational changes thus making the overall helicity insensitive to large changes in pH. Further, the results clearly argue against a single, rod-like alpha helical conformation being the only or even dominant conformation in the ensembles of so-called SAH sequences.  相似文献

Performance of electrocatalytic reactions depends on not only the composition and structure of the active sites, but also their local environment, including the surrounding electrolyte. In this work, we demonstrate that BF2(OH)2 anion is the key fluoroborate species formed in the mixed KBi/KF (KBi=potassium borate) electrolyte to enhance the rate of the oxygen evolution reaction (OER) at near-neutral pH. Through a combination of electrokinetic and in situ spectroscopic studies, we show that the mixed KBi/KF electrolyte promotes the OER via two pathways: 1) stabilizing the interfacial pH during the proton-producing reaction with its high buffering capacity; and 2) activating the interfacial water via strong hydrogen bonds with F-containing species. With the KBi/KF electrolyte, electrodeposited Co(OH)2 is able to achieve 100 mA/cm2 at 1.74 V, which is among the highest reported activities with earth-abundant electrocatalysts at near neutral conditions. These findings highlight the potential of leveraging electrolyte-engineering for improving the electrochemical performance of the OER.  相似文献
