A new approach for in situ fabrication of nanoscale fibrous chitosan membrane by biospecific degradation under physiological situation was studied. The chitosan binary blend membranes were fabricated by solvent casting of chitosan solution containing highly deacetylated chitosan (HDC) and moderately deacetylated chitosan (MDC) with different ratio. The biodegradation process was performed in PBS (pH 7.4) containing lysozyme at the temperature of 37 °C. Experimental results from weight loss, reducing sugar in surrounding media, FT-IR, X-ray diffraction, gel permeation chromatography (GPC) and SEM throughout the study showed that the biospecific degradation by lysozyme had removed MDC component selectively. When the ratio of MDC in the binary blend membranes amounted to 0.5, nanoscale domains of HDC and MDC were obtained, and thus a nanoscale fibrous structure was fabricated after biospecific degradation of MDC. This nanofibrous structure and the biospecific degradation of chitosan membranes can have potential advantages and interesting implications in tissue engineering and drug delivery.  相似文献

A novel strategy for predicting bioactive components in traditional Chinese medicines using Chinese hamster ovary‐sphingomyelin synthase2 (CHO‐SMS2) cell biospecific extraction and high‐performance liquid chromatography with diode array detection and tandem mass spectrometry analysis was proposed. The hypothesis is that when cells are incubated with the extract of traditional Chinese medicines, the potential bioactive components in the traditional Chinese medicines should selectively combine with the cells, while the cell‐combining components would be detectable in the extract of denatured cells. The identities of the cell‐combining components could be determined by liquid chromatography with tandem mass spectrometry. Using the proposed approach, the potential bioactive components of Rhizoma Polygoni Cuspidati, a commonly used traditional Chinese medicine for atherosclerosis, were detected and identified. Eight compounds in the extract of Rhizoma Polygoni Cuspidati were detected as the components selectively combined with CHO‐SMS2 cells, which is a stable cell line that highly expresses sphingomyelin synthases, it was found that piceid, resveratrol, emodin‐8‐β‐d‐ glucoside, physcion‐8‐β‐d‐ glucoside, emodin, physcion, 3,5,4‘‐trihydroxystilbene‐3‐O‐(6“‐galloyl)‐glucoside, and emodin‐1‐O‐glucoside combined specifically with CHO‐SMS2 cells. The results indicate that the proposed approach may be applied to predict the bioactive candidates in traditional Chinese medicines.  相似文献

A study of specific interactions between lectins and glycoproteins has been carried out using surface plasmon resonance (SPR) in a flow-injection mode. Lectins were covalently immobilised on the surfaces of the microfluidic sensor chip via amine coupling and serum glycoproteins were injected into the flow channels. Specific lectin-glycoprotein interactions caused the shift of refractive index proportional to the mass concentration accumulated on the channel surface. Lectins showed different affinity to the tested glycoproteins and each glycoprotein displayed its own lectin-binding pattern. It is possible to distinguish and identify even glycoproteins with similar sugar structures by simple and quick screening. The working conditions of the assay were optimised. The lectin-based SPR made it possible to carry out the label-free detection of glycoproteins within a broad concentration range with a good linearity. Regeneration conditions for the surface of the sensor chip were found and optimised. Combination of 10 mM HCl and 10 mM glycine-HCl (pH 2.5) removes the bound glycoproteins from the lectin surface without damaging it. The kinetic and affinity parameters of lectin-glycoprotein binding were evaluated. The proposed method was tested on human glycosylated serum. Combination of the lectin panel with SPR is suitable both for specific screening and for sensitive assay of serum glycoproteins.  相似文献

Immobilized lectins placed in continuous flow systems were used for biospecific reversible immobilization of labile biochemical structures, e.g., enzymes such as ascorbic acid oxidase and acetylcholine esterase, and cells, such as red blood cells and lymphocytes.
