研究了不同毛细管预处理方法对组胺、5-羟色胺及儿茶酚胺类神经递质电泳分离的影响,采用优化的毛细管预处理方法及电泳分离条件基线分离了组胺、5-羟色胺、去甲肾上腺素和肾上腺素。利用多巴胺和5-羟色胺在毛细管内壁吸附效应的不同,对电泳淌度极为相近的多巴胺和5-羟色胺进行了分离和鉴定。以PC22细胞为研究对象,证实了该细胞中所含大量神经递质是多巴胺,不是5-羟色胺。  相似文献

Summary Tokaj wines (Szamorodni and Aszu wines) of Hungarian origin were investigated on the basis of free amino acids and biogenic amines. The separation and determination of these compounds was performed by an amino acid analyser equipped with an ion-exchange resin column. The total amount of free amino acids and biogenic amines was higher in Aszu wines than in Szamorodni wines. The main amino acids were proline and arginine, while the major biogenic amines were tyramine and putrescine. The free amino acid and biogenic amine content of Aszu wines depended on the vineyards the wines originated from. Summary The composition and stability of the following biogenic amine complexes have been investigated: 1,4-diaminobutane(Put), 4-azaoctane-1,8-diamine(Spd), 4,9-diazadodecan-1, 12-diamine(Spm) as well as homologues such as 1,3-diaminopropane(Put3), 4-azaheptane-1, 7-diamine(Spd3,3) and 4,8-diazaundecan-1,11-diamine(Spm3,3,3) with H+, Cu(II), Zn(II), Pb(II) and Mg(II). A potentiometric method was used. The VIS technique enabled the determination of coordination mode in copper/amine systems. It was found that Mg(II) does not form coordination compounds with any of the studied polyamines in solution. An increase in the concentration of ligand and metal was found to result in a stronger tendency towards the formation of protonated compounds accompanied by a decrease in the concentration of hydroxocomplexes. At physiologicalpH (7.4) an increase in the concentration of protonated compounds by approximately 15% was observed within the ligand concentration range from 0.001 mol dm–3 to 0.0001 mol dm–3 at a Cu(II) concentration of 0.000177 mol dm–3.
Untersuchungen zur Komplexbildung von Polyaminen mit H+, Cu(II), Zn(II), Pb(II) und Mg(II) in wäßriger Lösung
Zusammenfassung Anhand einer Analyse von potentiometrischen Daten wurden Zusammensetzung und Beständigkeit folgender biogener Aminkomplexe untersucht: 1,4-Diaminobutan(Put), 4-Azaoktan-1,8-diamin(Spd), 4,9-Diazadodekan-1,12-diamin(Spm), sowie auch deren Homologen 1,3-Diaminopropan(Put3), 4-Azaheptan-1,7-diamin(Spd3,3) und 4,8-Diazaundekan-1,11-diamin(Spm3,3,3) mit H+, Cu(II), Zn(II), Pb(II) und Mg(II). Mit Hilfe der VIS-Technik wurde die Koordinationsweise in Kupfer/Amin-Systemen bestimmt. Es wurde festgestellt, daß Mg(II) keine Koordinationsverbindungen mit den untersuchten Polyaminen bildet. Eine höhere Konzentration von Ligand und Metall führte zu stärkerer Tendenz der Bildung protonierter Verbindungen, wobei die Konzentration von Hydroxokomplexen kleiner wurde. Bei physiologischempH (7.4) wurde im Bereich der Ligand-Konzentration von 0.001 mol dm–3 bis 0.0001 mol dm–3 bei einer Cu(II)-Konzentration von 0.000177 mol dm–3 ein Anstieg der Konzentration protonierter Verbindungen um etwa 15% beobachtet.

Abstract

An analytical procedure for the sampling and quantitative determination of biogenic hydrocarbons in the atmosphere is presented. The method is based on the adsorptive preconcentration of the hydrocarbons using Tenax TA/Carbopack B filled sampling tubes followed by thermodesorption of the trapped compounds and gas chromatographic analysis. To avoid losses of the biogenic alkenes as a consequence of their reaction with ozone on the adsorbents during the sampling step, an ozone scrubber is used in front of the adsorption tubes. Diurnal variations of the monoterpene- and ozone-concentrations are determined at different heights in and above a forest stand (coniferous and deciduous trees) in the Vosges (Col du Donon, 760 m a.s.l.) in order to elucidate the importance of terpene-oxidation reactions on the formation of peroxidic products in forest air.  相似文献

The performance of three different types of ion mobility spectrometer (IMS) devices: GDA2 with a radioactive ion source (Airsense, Germany), UV-IMS with a photo-ionization source (G.A.S. Germany) and VG-Test with a corona discharge source (3QBD, Israel) was studied. The gas-phase ion chemistry in the IMS devices affected the species formed and their measured reduced mobility values. The sensitivity and limit of detection for trimethylamine (TMA), putrescine and cadaverine were compared by continuous monitoring of a stream of air with a given concentration of the analyte and by measurement of headspace vapors of TMA in a sealed vial. Preprocessing of the mobility spectra and the effectiveness of multivariate curve resolution techniques (MCR-LASSO) improved the accuracy of the measurements by correcting baseline effects and adjusting for variations in drift time as well as enhancing the signal to noise ratio and deconvolution of the complex data matrix to their pure components. The limit of detection for measurement of the biogenic amines by the three IMS devices was between 0.1 and 1.2 ppm (for TMA with the VG-Test and GDA, respectively) and between 0.2 and 0.7 ppm for putrescine and cadaverine with all three devices. Considering the uncertainty in the LOD determination there is almost no statistically significant difference between the three devices although they differ in their operating temperature, ionization method, drift tube design and dopant chemistry. This finding may have general implications on the achievable performance of classic IMS devices.  相似文献

A simple, rapid, sensitive, and environmentally friendly method, based on modified dispersive liquid–liquid microextraction coupled with high‐performance liquid chromatography was developed for the simultaneous determination of five biogenic amines in fermented food samples. Biogenic amines were derivatized with 9‐fluorenylmethyl chloroformate, extracted by vortex‐assisted surfactant‐enhanced emulsification liquid–liquid microextraction, and then analyzed by high‐performance liquid chromatography. Five biogenic amine compounds were separated within 30 min using a C18 column and gradient elution with acetonitrile and 1% acetic acid. Factors influencing the derivatization and extraction efficiency such as type and volume of extraction solvent, type, and concentration of surfactant, pH, salt addition, and vortex time were optimized. Under the optimum conditions, the method provided the enrichment factors in the range of 161–553. Good linearity was obtained from 0.002–0.5 mg/L for cadaverine and tyramine, 0.003–1 mg/L for tryptamine and histamine, and 0.005–1 mg/L for spermidine with coefficient of determination (R2) > 0.992. The limits of detection ranged from 0.0010 to 0.0026 mg/L. The proposed method was successfully applied to analysis of biogenic amines in fermented foods such as fermented fish (plaa‐som), wine and beer where good recoveries were obtained in the range of 83.2–112.5%  相似文献

Due to their physical, chemical, optical, and mechanical properties, metallic nanoparticles (MNPs) are increasingly being used, with an emphasis on silver nanoparticles (AgNPs) and gold nanoparticles (AuNPs). In recent years, green synthesis has gained prominence for exploring the use of naturally available biological sources for the obtention of metallic nanoparticles. Among these, algae and plants stand out due to the presence of polysaccharides, proteins, polyphenols, and vitamins (among others) in their composition, which can act in the reduction and stabilisation of MNPs, and these biogenic materials have been characterised mainly by spectrometric and microscopic techniques. In addition, due to the numerous advantages of nanoparticles (NPs) synthetize from biogenic source, such as their simplicity and cost benefits, they have been used in the development of sensors applied in the determination of contaminants present in environmental samples and in the catalytic reduction of organic and inorganic contaminants. Therefore, this review describes the synthesis, mechanisms, characterization, and environmental analytical applications of NPs obtained by biogenic synthesis as well as the perspectives and challenges of these NPs.  相似文献

Three sorbent materials (A18C6-MS, DA18C6-MS and AB18C6-MS) based on the crown ether ligands, 1-aza-18-crown-6, 1,4,10,13-tetraoxa-7,16-diazacyclo octadecane and 4′-aminobenzo-18-crown-6, respectively, were prepared by the chemical immobilization of the ligand onto mesoporous silica support. The sorbents were characterized by FT-IR, scanning electron microscopy-energy dispersive X-ray microanalysis, elemental analysis and nitrogen adsorption-desorption test. The applicability of the sorbents for the extraction of biogenic amines by the batch sorption method was extensively studied and evaluated as a function of pH, biogenic amines concentration, contact time and reusability. Under the optimized conditions, all the sorbents exhibited highest selectivity toward spermidine (SPD) compared to other biogenic amines (tryptamine, putrescine, histamine and tyramine). Among the sorbents, AB18C6-MS offer the highest capacity and best selectivity towards SPD in the presence of other biogenic amines. The AB18C6-MS sorbent can be repeatedly used three times as there was no significant degradation in the extraction of the biogenic amines (%E > 85). The optimized procedure was successfully applied for the separation of SPD in food samples prior to the reversed-phase high performance liquid chromatography separation.  相似文献

This paper reports a new, simple, rapid and economical method for routine determination of 24 amino acids and biogenic amines in grapes and wine. No sample clean-up is required and total run time including column re-equilibration is less than 40min. Following automated in-loop automated pre-column derivatisation with an o-phthaldialdehyde, N-acetyl-l-cysteine reagent, compounds were separated on a 3mm×25cm C(18) column using a binary mobile phase. The method was validated in the range 0.25-10mg/l; repeatability was less than 3% RSD and the intermediate precision ranged from 2 to 7% RSD. The method was shown to be linear by the 'lack of fit' test and the accuracy was between 97 and 101%. The LLOQ varied between 10μg/l for aspartic and glutamic acids, ethanolamine and GABA, and 100μg/l for tyrosine, phenylalanine, putrescine and cadaverine. The method was applied to grapes, white wine, red wine, honey and three species of physalis fruit. Grapes and physalis fruit were crushed, sieved, centrifuged and diluted 1/20 and 1/100, respectively, for analysis; wines and honeys were simply diluted 10-fold. It was shown using this method that the amino acid content of grapes was strongly correlated with berry volume, moderately correlated with sugar concentration and inversely correlated with total acidity.  相似文献

We describe simultaneous analysis of naphthalene-2,3-dicarboxaldehyde (NDA)-amino acid and NDA-biogenic amine derivatives by CE in conjunction with light-emitting diode-induced fluorescence detection using poly(ethylene oxide) (PEO) solutions containing sodium dodecyl sulfate (SDS). After sample injection, via EOF 0.1% PEO prepared in 100 mM TB solution (pH 9.0) containing 30 mM SDS entered a capillary filled with 0.5 M TB solution (pH 10.2) containing 40 mM SDS. Under this condition, 14 NDA-amino acid and NDA-amine derivatives were separated within 16 min, with high efficiency ((1.0–3.2) × 105 theoretical plates) and sensitivity (LODs at S/N = 3 ranging from 2.06 to 19.17 nM). In the presence of SDS and PEO, analytes adsorption on the capillary wall was suppressed, leading to high efficiency and reproducibility. The intraday analysis RSD values (n = 3) of the mobilities for the analytes are less than 0.52%. We have validated the practicality of this approach by quantitative determination of 9 amino acids in breast cancer cells (MCF-7) and 10 amino acids in normal epithelial cells (H184B5F5/M10). The concentrations of Tau and Gln in the MCF-7 cells were different than those in the H184B5F5/M10 cells, respectively. Our results show the potential of this approach for cancer study.  相似文献
