The O-(arylcarbamoyl)butyrhydroximoyl chlorides were synthesized, and the x-ray structural analysis (XSA) of one of them — O-(o-tolylcarbamoyl)butyrhydroximoyl chloride — was performed. The reactions of 5-methyl-3-hydroxyisoxazole with 2, 2-dimethyl, 3-substituted cyclopropanecarboxylic chlorides give O-acyl-5-methyl-3-hydroxyisoxazole and N-acyl-5-methylisoxazolin-3-one derivatives. The ratio of O-acyl to N-acyl product depends upon the acylation reagents. O-acyl derivatives can be converted to the N-acyl compounds by isomerization under acidic conditions or heating.  相似文献

Peptidosulfonamides are an emerging group of peptidomimetics with a variety of applications in medicinal chemistry. We present a novel approach to the synthesis of peptidosulfonamides, and apply it to a series of new potential inhibitors of the bacterial peptidoglycan biosynthesis enzymes MurD and MurE. The synthesis was conducted via N-phthalimido β-aminoethanesulfonyl chlorides, which are new building blocks for the synthesis of peptidosulfonamides. In the most crucial step, sulfonic acids or their sodium salts were converted into the corresponding sulfonyl chlorides using an excess of either SOCl2 or SOCl2/DMF, and then coupled to the C-protected amino acid. None of the compounds significantly inhibited MurD, however, some inhibited MurE; one had an IC50 below 200 μM, which constitutes a promising starting point for further development. Molecular modelling simulations were performed on two analogues to investigate the absence of inhibitory activity of the sulfonamide compounds on MurD.  相似文献

Transmetalation of urea derivatives of α-aminoorganostannanes with alkyllithiums provides α-aminoorganolithiums which undergo rapid N to C acyl migration to afford α-aminocarboxamides. A simple new protocol for the high yielding Suzuki-Miyaura cross-couplings of aryl chlorides with aryl boronic acids using a palladium/imidazolium salt catalytic system is presented. Reaction of perfluoro-1-ethyl-2-methyl-1-propenyliminochloromethanesulfenyl chloride with 1-allyl-2-methoxybenzene in methylene chloride and nitromethane gives as the main products β-and β-chloro sulfides plus derivative of 2,3-dihydrobenzofuran. A series of novel bidentate pyrazolone based Schiff base ligands were synthesized by interaction of 4-benzoyl-3-methyl-1-(4′-methylphenyl)-2-pyrazolin-5-one with various aromatic amines like aniline, o-,m-,p-chloroaniline and o-,m-,p-toluidine in a ethanolic medium. All of these ligands have been characterized on the basis of elemental analysis, IR and 1H NMR data. The molecular geometries of five of these ligands have been determined by single crystal X-ray study. Crystallographic study reveals that these ligands exist in the amine-one tautomeric form in the solid state. NMR study also suggests the existence of the amine-one form in solution at room temperature. Ab initio calculations for representative ligand HL1 has been carried out to know the coordination site of the ligand. Novel vanadium Schiff base complexes of these ligands with general formula [OV(L1–7)2(H2O)] have been prepared by interaction of aqueous solution of vanadyl sulfate pentahydrate with DMF solution of the appropriate ligands. The resulting complexes have been characterized on the basis of elemental analysis, vanadium determination, molar conductance and magnetic measurements, thermo gravimetric analysis, infrared and electronic spectral studies. Suitable distorted octahedral structures have been proposed for these complexes.  相似文献

The treatment of 3,4-dihydropyrimidin-2-ones with n-BuLi at −78 °C, followed by quenching with various electrophiles furnished N3-substituted derivatives, regioselectively. Further, N1,N3-diacyl derivatives were found to transfer N1-acyl groups to nucleophilic sites.  相似文献
