高职高专院校计算机专业建设与改革探讨

进入21世纪,高职高专院校如雨后春笋在全国各地开始冒尖,而计算机技术更是在新世纪取得了高速的发展,在各高职高专院校中大部分都开设了计算机专业,本文从计算机专业人才培养目标入手,介绍了计算机专业课程的设置,同时对计算机课程建设进行了初步的探讨。     

Influence of Length of Opposing bi-Au Cone-Tips and Different Environment on Field Enhancement in Feed Gap

Electric field enhancement distributions encountered in feed gap of opposing bi-Au cone-tips is studied using a frequency-domain three-dimensional finite element method to solve Maxwell＇s equations of electric field distributions. Both the influences of cone-tip length and surrounding medium on electric field enhancement are investigated. The maximal enhancement value is discussed in terms of a simple physical model based on a standing wave on the tip surface associated with the antenna effect and surface plasmon. Simulated results demonstrate the enhancement is sensitive to the tip length. By selecting a suitably matched scale according to the incident wavelength, a large enhancement value can be observed within a small focused spot between the opposing tips permitting a high spatial resolution. The relative position of the opposing tips is also found for the optimum enhancement. All of the results suggest that our configuration is suitable for the site-specific Raman spectroscopic analysis at nanoscale.     

A functional probe with bowtie aperture and bull's eye structure for nanolithograph

The bowtie aperture surrounded by concentric gratings(the bull’s eye structure) integrated on the near-field scanning optical microscopy(NSOM) probe(aluminum coated fiber tip) for nanolithography has been investigated using the finite-difference time domain(FDTD) method.By modifying the parameters of the bowtie aperture and the concentric gratings,a maximal field enhancement factor of 391.69 has been achieved,which is 18 times larger than that obtained from the single bowtie aperture.Additionally,the light spot depends on the gap size of the bowtie aperture and can be confined to sub-wavelength.The superiority of the combination of the bowtie aperture and the bull’s eye structure is confirmed,and the mechanism for the electric field enhancement in this derived structure is analyzed.     

Improving lithographic masks with the assistance of indentations

Indentations etched on the output surface of a metallic mask are proposed to produce fine lithographic patterns with a resolution of 500 nm using the finite-difference time domain(FDTD) method.Such a designed mask is capable of enhancing near field lithography(NFL) resolution more than three times compared with the structure without indentations.The simulation results show that the interference disturbance between the adjacent lithographic channels can be eliminated efficiently by employing the indentations.As a straightforward consequence,the channel-to-channel interspaces can be shortened significantly,maintaining a uniform field distribution and high contrast.     

光子晶体传感器对溶液折射率和浓度的测量

为了得出光子晶体缺陷透射峰位置与溶液折射率及溶液浓度的关系,从而得知溶液的折射率和溶度之间的关系。本文构造了(AB)~NC(AB)~N型的一维光子晶体模型,采用时域有限差分法对该一维光子晶体的透射光谱做了数值模拟和分析。结果表明,蔗糖溶液折射率和浓度与光子晶体透过峰位置之间为严格的线性关系,光子晶体缺陷模透射峰位置随溶液浓度百分比的改变而变化。该溶液测量方法的溶液折射率测量灵敏度高达498.087 nm/RIU。这为光子晶体溶液折射率及浓度测量传感器的设计制作提供了参考,有助于该溶液浓度检测传感器在实际应用中的推广。     

表达雪花莲外源凝集素基因的油菜转基因植株的获得

利用农杆菌素LBA4404（pCAMBIA3300RG)转化优良甘蓝型油菜恢复系W723的下胚轴节段.pCAM-BIA3300RG含有Rssl启动子引导的雪花莲外源凝集素基因（gna）和CaMV-35S启动子引导的除草剂抗性基因（bar）。经过两轮除草剂（2.5mg/L bialaphos）筛选（两周/轮）,除草剂抗性再生芽被传入生根培养基中生根,对根系旺盛生长的植株中所含gna基因进行PCR分析,PCR分析证实了这些植株确为转基因植株,利用Western印迹法对随机选择的5株含gna基因的转基因植株的分析发现,其中4株表达了gna基因。目前正对这些表达gna基因的转基因植株进行后代遗传分离分析。     

大肠杆菌RecA蛋白质的提取与纯化

作者报道了从大肠杆菌超表达菌株ＪＣ１１２７２中，制备重组酶ＲｅｃＡ蛋白的研究。利用亚精胺选择性沉淀，单链ＤＮＡ－纤维素亲和层析及阳离子换柱Ｍｏｎｏ Ｑ进行离子交换层析，从２升超表达细菌培养物中提纯了５５ｍｇ和ＲｅｃＡ蛋白。通过９％ＳＤＳ－ＰＡＧＥ电泳检测结果表明，纯化的ＲｅｃＡ蛋白纯度达９９．５％，具有催化同源ＤＮＡ链交换的能力。     

Fibrillin转基因油菜的再生及其光合特性的检测

用根癌农杆菌共培养法，以带有1～2 mm子叶柄的完整子叶为转化受体，将连有35S启动子和NPTⅡ标记基因的Fibrillin cDNA导入甘蓝型油菜主要栽培品种“2005南系”中，获得抗卡那霉素的转基因植株.对部分经卡那霉素筛选得到的再生植株进行PCR检测，有来自3个株系的植株表现为阳性，初步证实外源基因已整合到植物细胞基因组中.将转基因植株炼苗后移入大田，在3月初到4月末油菜开花结果期间，对叶片和角果进行光合指标和叶绿素荧光指标的检测以及产量性状分析，发现转基因植株和非转基因植株之间存在明显差异.     

攀枝花苏铁(Cycas Panzhihuaensis L.Zhou et S.Y.Yang)生殖生物学特性研究——大孢子发生、受精前雌配子体及颈卵器的发育

报道了Ｃ．ｐａｎｚｈｉｈｕａｅｎｓｉｓ胚珠的结构、发育和受精前的雌配子体及颈卵器的发育．幼小胚珠由珠被、珠心组织组成,珠心组织内的功能大孢子经多次分裂形成游离核的雌配子体,最终形成细胞的雌配子体．人工授粉１５ｄ后,颈卵器开始发育,颈卵器由２个颈细胞和１个中央细胞组成,通常２个颈卵器能正常发育．
