The ergosterol content of soil can be used as an indicator of fungal activity. A method has been developed for the extraction and determination of ergosterol in organic soils, as part of a study to assess the correlation between fungal activity and the sequestration of metal pollutants. The moisture content of the soil affected the extraction process. Four consecutive extractions with methanol removed >95% of the ergosterol that can be obtained from the fresh sample (63% moisture) by exhaustive extraction. By freeze drying the soils prior to extraction (a) up to 35% more ergosterol was extracted after a single extraction, (b) >90% of the recoverable ergosterol was collected in two extractions and (c) the repeatability of the extraction was improved. Storage of soil extracts in the absence of light prevents degradation of ergosterol. A previously reported method for determination of ergosterol by HPLC has been improved by modification of the eluant composition. With 46% methanol/46% acetonitrile/8% dichloromethane, ergosterol was eluted with good resolution approximately 8 min after injection of 20 mul of the extract. The detection limit of the HPLC method was 0.5 mug/ml ergosterol, equivalent to 0.06 mug/g in 25 g fresh soil. Changes in ergosterol contents of peaty soil treated with fungicide, and in samples of the peaty podzol and a humus iron podzol in the vicinity of fungal fruiting bodies, have been determined.  相似文献

We have observed electric field modulated Raman scattering by A1 LO phonons in CdS. The field induced scattering is observed with a geometry in which Raman scattering by A1 LO phonons is normally allowed. The interference of the field induced and allowed terms in the transition susceptibility leads to a modulated Raman scattering intensity proportional to the applied field. This is contrasted with data previously reported on field induced Raman scattering by E1 LO phonons in a configuration in which the Raman scattering is normally forbidden and in which there is no interference between linear wavevector dependent and field induced terms in the transition susceptibility. Electric field effects on Raman scattering by TO phonons and by 2 LO phonons is also discussed.  相似文献

Surfaced enhanced Raman scattering (SERS) nanotags operating with 1280 nm excitation were constructed from reporter molecules selected from a library of 14 chalcogenopyrylium dyes containing phenyl, 2-thienyl, and 2-selenophenyl substituents and a surface of hollow gold nanoshells (HGNs). These 1280 SERS nanotags are unique as they have multiple chalcogen atoms available which allow them to adsorb strongly onto the gold surface of the HGN thus producing exceptional SERS signals at this long excitation wavelength. Picomolar limits of detection (LOD) were observed and individual reporters of the library were identified by principal component analysis and classified according to their unique structure and SERS spectra.  相似文献

A number of novel, conjugated, unsymmetrically disubstituted diphenyl-diacetylenes have been synthesized and polymerized in the liquid-crystalline phase to yield liquid-crystalline polymers. The efficiency of second harmonic generation is reported for both monomers and polymers.  相似文献

We present data on forward and backward scattering in CuCl at liquid N2 temperature. Peaks at 146 and 172 cm-1 which appear in both forward and backward scattering spectra are attributed to other than first order scattering by optical phonons. The data on polarition scattering indicate that the atomic displacement contribution to the scattering by TO phonons is rather small. From the experimentally determined polarition dispersion curve we obtain a value of 174 ±2.5 cm-1 for ωT and a value of 5.3 for ?S.  相似文献

Nanocrystalline Gd0.946Fe0.054 of average grain size 68 nm was prepared by melt-spinning. The magnetic behavior in the vicinity of the paramagnetic to ferromagnetic transition was investigated via dc magnetization and ac susceptibility measurements. The transition temperature and effective critical exponents for the order parameter and zero-field susceptibility were determined using Arrott-Noakes and Kouvel-Fisher analyses. The values obtained were TC=291.71±0.07 K, βeff=0.385±0.009, and γeff=1.24±0.03, respectively. Correction to scaling analysis indicated that the asymptotic exponents were both smaller than the effective ones within the reduced-temperature range investigated, contrary to the behavior seen in monocrystalline Gd. This behavior can be explained in terms of a crossover from 3D short-range Heisenberg universality class to the 3D Ising universality class due to increased anisotropy induced by the high magnetic fields used in the measurements and also possibly due to strain.  相似文献

Quantitation of selenium in plants, soils and sludges was achieved by isotope dilution-mass spectrometry using a benchtop instrument. Samples for analysis were spiked with (76)Se isotope solution. Plant material was digested on a heating block at 150 degrees C using a mixture of nitric acid and hydrogen peroxide. Selenium in soils and sludges was released by treatment with nitric acid followed by digestion with nitric and hydrofluoric acids. Selenium in the digests was reduced to Se(IV) with hydrochloric acid and derivatised with nitro-1, 2-phenylenediamine to 5'-nitropiazselenol. Analysis by gas chromatography-mass spectrometry using selected ion monitoring was validated using certified reference materials (CRMs) and gave results within the certified range with a low standard deviation. The CRMs plant (Chinese cabbage leaves) and soil (Chinese soil) were found to contain (+/-95% confidence limits) 0.091(+/-0.007) mug g(-1) and 1.67(+/- 0.04) mug g(-1)Se respectively. The certified values were 0.083(+/-0.008) mug g(-1) and 1.56(+/-0.12) mug g(-1) respectively. The selenium content of four different freely drained acid Scottish soils under grasslands was in the range 0.5-0.8 mug g(-1) air-dried soil. Sewage sludges were found to contain measurably more selenium than the soils, and samples of three sludges taken from sites in the UK contained between 1.1 and 3.5 mug g(-1) dry matter.  相似文献

Infection of plant cells by potyviruses induces the formation of cytoplasmic inclusions ranging in size from 200 to 1000 nm. To determine if the ability to form these ordered, insoluble structures is intrinsic to the potyviral cytoplasmic inclusion protein, we have expressed the cytoplasmic inclusion protein from Potato virus Y in tobacco under the control of the chrysanthemum ribulose-1,5-bisphosphate carboxylase small subunit promoter, a highly active, green tissue promoter. No cytoplasmic inclusions were observed in the leaves of transgenic tobacco using transmission electron microscopy, despite being able to clearly visualize these inclusions in Potato virus Y infected tobacco leaves under the same conditions. However, we did observe a wide range of tissue and sub-cellular abnormalities associated with the expression of the Potato virus Y cytoplasmic inclusion protein. These changes included the disruption of normal cell morphology and organization in leaves, mitochondrial and chloroplast internal reorganization, and the formation of atypical lipid accumulations. Despite these significant structural changes, however, transgenic tobacco plants were viable and the results are discussed in the context of potyviral cytoplasmic inclusion protein function.  相似文献
