In-beam conversion electron spectroscopy experiments have been performed on the transfermium nuclei 253, 254No using the conversion electron spectrometer SACRED in nearly collinear geometry in conjunction with the gas-filled separator RITU at the University of Jyv?skyl?. The experimental setup is discussed and the spectra are compared to Monte Carlo simulations. The implications for the ground-state configuration of 253No are discussed. Dynamic mechanical measurements on ultrahigh modulus polyoxymethylene have been undertaken over the temperature range ?150 to 20°C. Measurements of the longitudinal crystal modulus have also been made by studying changes in the (009) reflection with load, over a similar range of temperatures. The dynamic Young's modulus at 5 Hz reaches a value at low temperatures of 64.5 GPa for the most highly oriented sample. The crystal modulus at low temperatures is 105 GPa, which is almost twice the previously reported room-temperature value.  相似文献

A new reversed-phase high-performance liquid chromatography approach to the production of analytical peptide maps by pre-column derivatization using phenylisothiocyanate is described. Tryptic peptide digests were derivatized with phenyl isothiocyanate to form the phenylthiocarbamyl peptides followed by reversed-phase high-performance liquid chromatographic analysis. The phenylthiocarbamyl peptides were separated by reversed-phase high-performance liquid chromatography with the conventional gradient elution system of water-acetonitrile containing trifluoroacetic acid. The sensitivity of detection of these peptide derivatives was within the range 5-10 pmol with a constant baseline at 254-260 nm. The isolated phenylthiocarbamyl peptides can be subjected to automatic Edman degradation. The effectiveness of this method was exemplified by microsequencing of phenylthiocarbamyl peptides isolated from tryptic digests of three different proteins: alpha-lactalbumin, beta-lactoglobulin and a lambda light-chain immunoglobulin.  相似文献

Alterations in activities of one family of proteases, the matrix metalloproteinases (MMPs), have been implicated in primary and metastatic tumor growth, angiogenesis, and pathological degradation of extracellular matrix (ECM) components, such as collagen and laminin. Since hydrolysis of the collagen triple-helix is one of the committed steps in ECM turnover, we envisioned modulation of collagenolytic activity as a strategy for creating selective MMP inhibitors. In the present study, a phosphinate transition state analogue has been incorporated within a triple-helical peptide template. The template sequence was based on the alpha1(V)436-450 collagen region, which is hydrolyzed at the Gly(439)-Val(440) bond selectively by MMP-2 and MMP-9. The phosphinate acts as a tetrahedral transition state analogue, which mimics the water-bound peptide bond of a protein substrate during hydrolysis. The phosphinate replaced the amide bond between Gly-Val in the P1-P1' subsites of the triple-helical peptide. Inhibition studies revealed Ki values in the low nanomolar range for MMP-2 and MMP-9 and low to middle micromolar range for MMP-8 and MMP-13. MMP-1, MMP-3, and MT1-MMP/MMP-14 were not inhibited effectively. Melting of the triple-helix resulted in a decrease in inhibitor affinity for MMP-2. The phosphinate triple-helical transition state analogue has high affinity and selectivity for the gelatinases (MMP-2 and MMP-9) and represents a new class of protease inhibitors that maximizes potential selectivity via interactions with both prime and nonprime active site subsites as well as with secondary binding sites (exosites).  相似文献

Support vector machines (SVMs), that utilize a mixture of the L1L1-norm and the L2L2-norm penalties, are capable of performing simultaneous classification and selection of highly correlated features. These SVMs, typically set up as convex programming problems, are re-formulated here as simple convex quadratic minimization problems over non-negativity constraints, giving rise to a new formulation – the pq-SVM method. Solutions to our re-formulation are obtained efficiently by an extremely simple algorithm. Computational results on a range of publicly available datasets indicate that these methods allow greater classification accuracy in addition to selecting groups of highly correlated features. These methods were also compared on a new dataset assessing HIV-associated neurocognitive disorder in a group of 97 HIV-infected individuals.  相似文献

The spectrum of prompt conversion electrons emitted by excited 254No nuclei has been measured, revealing discrete lines arising from transitions within the ground state band. A striking feature is a broad distribution that peaks near 100 keV and comprises high multiplicity electron cascades, probably originating from M1 transitions within rotational bands built on high K states.  相似文献
