A number of thiosemicarbazones of 2‐acetyl‐imidazo[4,5‐b]pyridine were prepared in order to investigate their in vitro antineoplastic activities. Three compounds: (i) 2‐acetylimidazo[4,5‐b]pyridin‐4‐ sec ‐butyl‐3‐thiosemicarbazone [(A7), NSC674098], (ii) 2‐acetylimidazo[4,5‐b]pyridin‐4‐tert‐butyl‐3‐thiosemi‐carbazone [(A9), NSC674099], (iii) 2‐acetylimidazo[4,5‐b]pyridin‐4‐cyclohexyl‐3‐thiosemicarbozone [(A11), NSC674101] showed remarkable activity against some of the cell lines tested. The Biological Evaluation Committee of N.C.I. determined that further secondary testing should be carried out (these compounds were tested against prostate cancer).  相似文献

Isoquercitrin is a flavonoid chemical compound that can be extracted from different plant species such as Mangifera indica (mango), Rheum nobile , Annona squamosal , Camellia sinensis (tea), and coriander ( Coriandrum sativum L.). It possesses various biological activities such as the prevention of thromboembolism and has anticancer, antiinflammatory, and antifatigue activities. Therefore, there is a critical need to elucidate and predict the qualitative and quantitative properties of this phytochemical compound using the high performance liquid chromatography (HPLC) technique. In this paper, three different nonlinear models including artificial neural network (ANN), adaptive neuro-fuzzy inference system (ANFIS), and support vector machine (SVM),in addition to a classical linear model [multilinear regression analysis (MLR)], were used for the prediction of the retention time (tR) and peak area (PA) for isoquercitrin using HPLC. The simulation uses concentration of the standard, composition of the mobile phases (MP-A and MP-B), and pH as the corresponding input variables. The performance efficiency of the models was evaluated using relative mean square error (RMSE), mean square error (MSE), determination coefficient (DC), and correlation coefficient (CC). The obtained results demonstrated that all four models are capable of predicting the qualitative and quantitative properties of the bioactive compound. A predictive comparison of the models showed that M3 had the highest prediction accuracy among the three models. Further evaluation of the results showed that ANFIS–M3 outperformed the other models and serves as the best model for the prediction of PA. On the other hand, ANN–M3proved its merit and emerged as the best model for tR simulation. The overall predictive accuracy of the best models showed them to be reliable tools for both qualitative and quantitative determination.  相似文献

The association between hydrophobically modified poly(sodium acrylate) (HMPA) and poly(N-isopropylacrylamide) (PNIPAM) in aqueous solution was studied using turbidimetry, viscometry and fluorescence measurements. Both the polymeric and the amphiphilic nature of the HMPA influence the association process. The tendency for association, as reflected by the increase in the cloud point and the reduced viscosity of PNIPAM, increases with the length of the alkyl group and the degree of substitution of HMPA. The fluorescence study, using pyrene as a probe, ascertains that the association is of hydrophobic nature and the association process is gradual and less cooperative than the association of PNIPAM with ionic surfactants. When high molar mass HMPA is used, the hydrophobic association between HMPA and PNIPAM leads to the formation of a reversible network with significantly enhanced thickening properties as compared to the thickening ability of the corresponding pure HMPA in aqueous solution.  相似文献

An analysis is presented of the stresses induced in a thin tape by bending over pulleys, by axial tension and by twisting of the tape when running between two out-of-plane pulleys. Analytical results are compared with some photoelastic tests on tape models made of epoxy resin. The weak retardation produced by the thin tape is multiplied by repeated light passage through the tape and is measured by a bent-beam compensator. The tests are of especial interest because the rate of rotation of the principal stresses is higher than had been encountered before. Surprisingly, the experimental results uncorrected for rotation are in good agreement with the calculated stresses.  相似文献

Structural analysis of flexible macromolecular systems such as intrinsically disordered or multidomain proteins with flexible linkers is a difficult task as high-resolution techniques are barely applicable. A new approach, ensemble optimization method (EOM), is proposed to quantitatively characterize flexible proteins in solution using small-angle X-ray scattering (SAXS). The flexibility is taken into account by allowing for the coexistence of different conformations of the protein contributing to the experimental scattering pattern. These conformers are selected using a genetic algorithm from a pool containing a large number of randomly generated models covering the protein configurational space. Quantitative criteria are developed to analyze the EOM selected models and to determine the optimum number of conformers in the ensemble. Simultaneous fitting of multiple scattering patterns from deletion mutants, if available, provides yet more detailed local information about the structure. The efficiency of EOM is demonstrated in model and practical examples on completely or partially unfolded proteins and on multidomain proteins interconnected by linkers. In the latter case, EOM is able to distinguish between rigid and flexible proteins and to directly assess the interdomain contacts.  相似文献

In this work, we study the stability and internal modes of one-dimensional gap solitons employing the modified nonlinear Schrödinger equation with a sinusoidal potential together with the presence of a weak nonlocality. Using an analytical theory, it is proved that two soliton families bifurcate out from every Bloch-band edge under self-focusing or self-defocusing nonlinearity, and one of these is always unstable. Also we study the oscillatory instabilities and internal modes of the modified nonlinear Schrödinger equation.  相似文献

The proteolytic activity of matrix metalloproteinases toward extracellular matrix components (ECM), cytokines, chemokines, and membrane receptors is crucial for several homeostatic and pathological processes. Active MMPs are a family of single-chain enzymes (23 family members in the human genome), most of which constituted by a catalytic domain and by a hemopexin-like domain connected by a linker. The X-ray structures of MMP-1 and MMP-2 suggest a conserved and well-defined spatial relationship between the two domains. Here we present structural data for MMP-12, suitably stabilized against self-hydrolysis, both in solution (NMR and SAXS) and in the solid state (X-ray), showing that the hemopexin-like and the catalytic domains experience conformational freedom with respect to each other on a time scale shorter than 10(-8) s. Hints on the probable conformations are also obtained. This experimental finding opens new perspectives for the often hypothesized active role of the hemopexin-like domain in the enzymatic activity of MMPs.  相似文献

Several methods used for the multiresidue analysis of pesticides from the environment and drinking water have been reported. However, most of these reports dealt with a small number of targeted pesticides or some special groups. A method that is simple, faster, and more cost‐effective than the environmental protection agency (EPA) method has been developed for the analysis of 82 frequently used pesticides in water samples obtained from Yeongsan and Sumjin rivers, as well as rice fields located in various locations around the two rivers. The samples were extracted by dichloromethane, and the pesticides were analyzed using a GC‐electron capture detector (ECD), followed by confirmation with GC‐MS. Recoveries were found to be between 82 and 120.1% for most of the tested pesticides, which were in agreement with the standard values dictated by the EPA. The method was potentially applied to 66 water samples for human consumption and 90 water samples from the rice fields and irrigation ditches that were collected from June to September 2007. Oxadiazon, butachlor, and alachlor were detected in some of the river water samples collected in June, iprobenfos (IBP) was detected in samples collected in August, and no pesticide was detected in September. On the other hand, chlorpyrifos‐methyl, IBP, hexaconazole, diazinon, oxadiazon, butachlor, and isoprothiolane were detected at relatively high concentrations in 48 rice paddy field water samples collected between June and September 2007. Alachlor in one sample and procymidone in some of the rice paddy field water samples were also detected in trace amounts. The results were consistent with the temporal pattern of pesticide application in Korean rice fields.  相似文献

In this paper, we propose a fuzzy logic–based mathematical model of a sequence of earthquakes using fuzzy reasoning tools. We formed a set of fuzzy implications in order to study them, and we computed their deviation, so that we could compare them and conclude about the most accurate one. The compositional rule of inference was considered, which is based on the generalized modus ponens scheme. The new fuzzy methodology was used for each implication. The data required for the implications were obtained from the aftershocks of an earthquake with significant effects in a specific area. The magnitude of the aftershocks and their time difference from the main incident provided the values for the new fuzzy algorithm application. Two samples were selected relative with the seismic activity, which occurred the following days. The one sample consisted of 30 values, and the other sample from all the values found in the data archives of the National Observatory of Athens. Results were shown for both samples. So a mathematical technique, which reproduces the incidents using basic information and based on only two parameters, is developed for the simulation of a seismic sequence, which follows a strong earthquake.  相似文献
