Zusammenfassung Im Bromelin, einer Protease aus Ananas comosus Var. Cayenne, sind einige Fremdaktivitäten enthalten. Durch Gel-Chromatographie an Sephadex G-75 läßt sich die saure Phosphatase und eine Esterase, die N-Acetyl-l-tyrosinäthylester spaltet, von der Protease abtrennen. Durch Blektrofokussierung konnte die saure Phosphatase in mindestens zwei Isoenzyme vom isoelektrischen Punkt 4, 3 bzw. 6,1 aufgetrennt werden.Die Michaelis-Konstante der sauren Phosphatase gegenüber p-Nitrophenylphosphat beträgt 0,76 · 10–3 M (nach Lineweaver-Burk) bzw. 0,73 · 10–3 (nach Eadie). Das pH-Optimum in Citratpuffer liegt bei pH 5,5. Es bestehen Anzeichen dafür, daß neben der höhermolekularen Phosphatase (Mol.-Gew. > 100000) auch eine niedermolekulare saure Phosphatase in den Stengeln der Ananas comosus vorkommt.In den Strünken der Ananas lassen sich erhebliche Mengen einer pflanzlichen Ribonuclease nachweisen. Dieses Enzym läßt sich durch Ionenaustausch-Chromatographie an DEAE-Cellulose von den anderen Begleitenzymen abtrennen.Eine weitere Varietät von Ananas comosus, die in Brasilien vorkommt, enthält ebenfalls große Aktivitäten an saurer Phosphatase sowie Ribonuclease, jedoch wenig Protease und keine Aktivität gegenüber N-Acetyl-l-tyrosinäthylester.
Isolation and properties of the acid phosphatase from ananas comosus
Bromelin, a protease from pineapple (ananas comosus var. Cayenne), contains several foreign activities. Acid phosphatase and an esterase splitting N-acetyl-l-tyrosineethyl ester can be separated from the protease by gel chromatography on Sephadex G-75. The acid phosphatase can be separated into at least two isoenzymes with isoelectric points of 4.3 and 6.1, resp., by means of electrofocusing.The Michaelis constant of acid phosphatase for p-nitrophenylphosphate is 0.76×10–3 M (according to Lineweaver-Burk) and 0.73×10–3M (according to Eadie). The optimum pH value in citrate buffer is about 5.5. Besides the higher-molecular phosphatase (mol.-weight > 100000) also a lower-molecular acid phosphatase seems to occur in the stems of ananas comosus.Considerable amounts of a vegetable ribonuclease can be detected in the stalks of ananas. This enzym can be separated from other accompanying enzymes by ionexchange chromatography on DEAE-cellulose. Another variety of ananas comosus, occurring in Brazil, also contains high activities of acid phosphatase and ribonuclease, but only small amounts of protease and no activity towards N-acetyl-l-tyrosineethyl ester.

The conversion of 1,3,5-substituted benzene and mesitylene by electrophilic aromatic substitution and Sonogashira cross-coupling, respectively, furnished the C3-symmetric, hexasubstituted benzene derivatives 1 and 2 with an alternating substitution pattern. Based on the molecular scaffolds obtained, the two systems serve as model compounds for novel receptor molecules with distinct geometric features. X-ray structures have been obtained for 1 and 2, which are discussed in regard to their aptitude as receptor platforms or supramolecular building blocks. By looking at the rotational barriers for the functional groups placed around the molecular scaffolds by variable temperature 1H NMR spectroscopy, 1 and 2 turn out to exist in rapidly interconverting conformations. The alignment of these potential binding groups around the molecular scaffolds should be strongly biased by specific interactions with suitable guest molecules.  相似文献

A sequence of single photons is emitted on demand from a single three-level atom strongly coupled to a high-finesse optical cavity. The photons are generated by an adiabatically driven stimulated Raman transition between two atomic ground states, with the vacuum field of the cavity stimulating one branch of the transition, and laser pulses deterministically driving the other branch. This process is unitary and therefore intrinsically reversible, which is essential for quantum communication and networking, and the photons should be appropriate for all-optical quantum information processing.  相似文献

Here, we explore a de novo sequencing strategy in which we combine Lys-N protein digestion with differential isotopic dimethyl labeling to facilitate the (de novo) identification of multiply charged peptides in ESI-MS, both under CID and ETD conditions. For a large fraction of the Lys-N generated peptides, all primary amines are present at the N-terminal lysine, enabling specific labeling of the N-terminus. Differential derivatization of only the peptide N-terminus in combination with the simultaneous fragmentation of the corresponding isotopologues allows the straightforward distinction of N-terminal fragments from C-terminal and internal fragments. Furthermore, also singly and multiply charged N-terminal fragments can easily be distinguished due to the mass differences of the isotope labeled fragment pairs. As a proof of concept, we applied this approach to proteins isolated from an avocado fruit, and were able to partially de novo sequence and correctly align, with green plant homologues, a previously uncharacterized avocado ascorbate peroxidase.  相似文献

Physico-chemical methods to sort single-walled carbon nanotubes (SWNTs) by chiral index are presently lacking but are required for in-depth experimental analysis and also for potential future applications of specific species. Here we report the unexpected selectivity of poly(N-decyl-2,7-carbazole) to almost exclusively disperse semiconducting SWNTs with differences of their chiral indices (n - m) ≥ 2 in toluene. The observed selectivity complements perfectly the dispersing features of the fluorene analogue poly(9,9-dialkyl-2,7-fluorene), which disperses semiconducting SWNTs with (n - m) ≤ 2 in toluene. The dispersed samples are further purified by density gradient centrifugation and analyzed by photoluminescence excitation spectroscopy. All-atom molecular modeling with decamer model compounds of the polymers and (10,2) and (7,6) SWNTs suggests differences in the π-π stacking interaction as origin of the selectivity. We observe energetically favored complexes between the (10,2) SWNT and the carbazole decamer and between the (7,6) SWNT and the fluorene decamer, respectively. These findings demonstrate that subtle structural changes of polymers lead to selective solvation of different families of carbon nanotubes. Furthermore, chemical screening of closely related polymers may pave the way toward simple, low-cost, and index-specific isolation of SWNTs.  相似文献

Photocleavable polymers based on 9,9-dialkylfluorene backbone and o-nitrobenzylether were designed and synthesized to obtain stable (n,m) enriched suspensions of semiconducting SWNTs in toluene. Photoirradiation of the suspensions triggered the precipitation of the SWNTs and TEM images indicate close packing of SWNTs pointing at partial removal of the coating polymer.  相似文献

The molecular and electronic structures of a series of all-meta-substituted phenylacetylene mesitylenes peripherally substituted with donor or acceptor (D-A) groups are studied. The impact of meta- and para-substitution patterns is also analyzed by employing Raman and optical spectroscopies in conjunction with theoretical calculations. Outer phenyl rings display a partial quinoid character induced by two different motifs: (i) outer phenyls --> triple bond charge transfer for the cases where these phenyls are substituted with electron-donors; (ii) double electron withdrawing effect in the molecules with the peripheral phenyls substituted with electron acceptors. A moderate tuning of the optical gap is observed in agreement with the partial blockade of pi-electron conjugation exerted by the meta disposition. The orbital structure of the compounds partially preserves that of the mesitylene group showing extra-conjugation due to the addition of the arms, so that conjugation is not entirely obstructed but partially impeded in the ground electronic state (i.e., electron occupied orbitals). As for the excited states, the low-lying energy empty orbitals offer better conditions for full conjugation over the whole molecular scaffold. Interesting optical properties such as overlapping centers along the lowest energy optical excitations and enhanced optical transparency with importance for the application of these materials in optoelectronics have been justified on the basis of the electronic structure. A greater degree of quinoidization, and more allowed pi-electron delocalization, over the entire molecule is recognized in the case of linear phenylacetylenes substituting in para positions the central core.  相似文献

C(3)-symmetric, octopolar 1,3,5-alkynylbenzenes with terminal pyridyl substituents are presented which show an increase up to 17-fold in the second-order NLO activity upon protonation while maintaining their octopolar character, as confirmed by the depolarization ratios measured.  相似文献
