The extraction of Al(III) and Zn(II) from an aqueous solution with two water-in-oil microemulsions, one containing di(2-ethylhexyl)phosphoric acid (DEHPA), was investigated to aid the understanding of the role of the extractant and the metal specific characteristics in the mechanism of microemulsion extraction. The extraction of Al with the DEHPA microemulsion increased by a factor of about 10 with respect to that in the conventional DEHPA system, whereas the extraction of Zn was lower than that in the single DEHPA system. Extraction with the DEHPA-free microemulsion was very low, showing that metal ion solubilization was not important in the mechanism of microemulsion extraction. It is proposed that the effect of the mixed microemulsion on the metal distribution coefficient is the result of the balance between a decrease in the complexation reaction yield due to the interaction between butanol and DEHPA, and the adsorption of the metal complex at the macro- and microinterfaces. The former leads to a decrease in Zn(II) extraction and the latter to Al(III) extraction synergism. Copyright 2000 Academic Press.  相似文献

A rapid and specific method for the quantitative determination of xylitol in human urine has been developed. The method consists of the gas-liquid chromatographic analysis of the acetate ester derivative of the alditol in deionized urine using dulcitol as an internal standard. As little as 20 ng xylitol can be detected. At concentrations ranging from 25 to 400 micrograms/ml urine, the accuracy is +/- 4.0%.  相似文献

A volume independent and a volume dependent lattice energy function involving short-range interatomic potentials were able to be fitted to the elastic constants, cohesive energy, lattice parameter and for the latter function to the vacancy formation energy and bcc-fcc lattice stability energy, as well, for fcc metals and bcc alkali metals, but not to the cohesive energy and C' elastic constant of bcc transition metals. The assumption that directional, but partial, covalent bonds exist between nearest-neighbors in the bcc transition elements provides an explanation for the latter results and in addition explains the identical dependence of C' and the bcc-fcc lattice stability upon Nd, where Nd is the average number of d electrons, for the bcc transition metals and alloys. Both the mechanical and thermodynamic stability of the bcc structure for transition metals and all transition metal alloys disappears for 5 ? Nd > 2 and <?1.  相似文献

The repulsive component of a classical inverse power pair potential is modified to take into account the crystal field induced change in shape of the electron density distribution responsible for the repulsive interaction of a chain sited atom in the A15 structure with neighboring atoms. Minimization of the lattice energy with respect to this shape parameter and also with respect to the lattice parameter results in predictions in the absence of adjustable parameters of: (1) the lattice parameters (to within an RMS deviation of the observed values of 0.4%); (2) the nearly zero value for (C11 ? C12)2 (as observed for Nb3Sn and V3Si); (3) the correct ordering of relative stability for 98 out of 101 observed comparisons between 4 different crystal structures; (4) other effects which have been or can be observed. Implicit in the predictions is the assumption that the crystal field induced shape change encounters no intraatomic resistance for elements belonging to the Ti, V and Cr columns of the periodic table—denoted “exotic” elements. Some consequences of this assumption are found to be consistent with experiment.  相似文献

A Lattice Energy Function that combines a Mie type interatomic potential and a free electron gas volume dependence has been applied to the study of grain boundary energy and structure of a Σ = 5 tilt boundary in Au, Cu and Ni and of solute segregation to the same. Interatomic potentials and volume dependencies of the solvent and solute were adjusted to fit the relative partial molar enthalpy and volume at infinite dilution order to construct a AB type potential and volume dependence. This AB interaction is then applied to calculate the binding energies of solute to various grain boundary sites and the resulting change in grain boundary energy. A relationship between the binding energy and change in grain boundary is derived. The relative values of the grain boundary energy are in agreement with experimental values of the average grain boundary energies. The relative binding energies of the tested solvent-solute systems are in agreemnet with expectations that certain systems should have larger binding energies than others. The behavior of solute binding energies and local relaxations are in agreement with other studies of grain boundary segregation which use different Lattice Energy Functions and relaxation algorithms. The change in grain boundary energy is shown to be directly proportional to the binding energy.  相似文献

Background

The emergence of multi- and extensively-drug resistant Mycobacterium tuberculosis strains has created an urgent need for new agents to treat tuberculosis (TB). The enzymes of shikimate pathway are attractive targets to the development of antitubercular agents because it is essential for M. tuberculosis and is absent from humans. Chorismate synthase (CS) is the seventh enzyme of this route and catalyzes the NADH- and FMN-dependent synthesis of chorismate, a precursor of aromatic amino acids, naphthoquinones, menaquinones, and mycobactins. Although the M. tuberculosis Rv2540c (aroF) sequence has been annotated to encode a chorismate synthase, there has been no report on its correct assignment and functional characterization of its protein product.

Results

In the present work, we describe DNA amplification of aroF-encoded CS from M. tuberculosis (Mt CS), molecular cloning, protein expression, and purification to homogeneity. N-terminal amino acid sequencing, mass spectrometry and gel filtration chromatography were employed to determine identity, subunit molecular weight and oligomeric state in solution of homogeneous recombinant Mt CS. The bifunctionality of Mt CS was determined by measurements of both chorismate synthase and NADH:FMN oxidoreductase activities. The flavin reductase activity was characterized, showing the existence of a complex between FMNox and Mt CS. FMNox and NADH equilibrium binding was measured. Primary deuterium, solvent and multiple kinetic isotope effects are described and suggest distinct steps for hydride and proton transfers, with the former being more rate-limiting.

Conclusion

This is the first report showing that a bacterial CS is bifunctional. Primary deuterium kinetic isotope effects show that C4-proS hydrogen is being transferred during the reduction of FMNox by NADH and that hydride transfer contributes significantly to the rate-limiting step of FMN reduction reaction. Solvent kinetic isotope effects and proton inventory results indicate that proton transfer from solvent partially limits the rate of FMN reduction and that a single proton transfer gives rise to the observed solvent isotope effect. Multiple isotope effects suggest a stepwise mechanism for the reduction of FMNox. The results on enzyme kinetics described here provide evidence for the mode of action of Mt CS and should thus pave the way for the rational design of antitubercular agents.
