Electrochemically switchable hydrogen-bonded molecular shuttles

A series of [2]rotaxanes containing succinamide and naphthalimide hydrogen-bonding stations for a benzylic amide macrocycle is described. Electrochemical reduction and oxidation of the naphthalimide group alters its ability to form hydrogen bonds to the macrocycle to such a degree that redox processes can be used to switch the relative macrocycle-binding affinities of the two stations in a rotaxane by over 8 orders of magnitude. The structure of the neutral [2]rotaxane in solution is established by (1)H NMR spectroscopy and shows that the macrocycle exhibits remarkable positional integrity for the succinamide station in a variety of solvents. Cyclic voltammetry experiments allow the simultaneous stimulation and observation of a redox-induced dynamic process in the rotaxane which is both reversible and cyclable. Model compounds in which various conformational and co-conformational changes are prohibited demonstrate unequivocally that the redox response is the result of shuttling of the macrocycle between the two stations. At room temperature in tetrahydrofuran the electrochemically induced movement of the macrocycle between the two stations takes approximately 50 micros.     

First Measurement of the Gerasimov-Drell-Hearn Sum Rule for 1H from 0.7 to 1.8 GeV at ELSA

To verify the fundamental Gerasimov-Drell-Hearn (GDH) sum rule for the first time experimentally, we measured the helicity dependent total photoabsorption cross section with circularly polarized real photons and longitudinally polarized nucleons in the photon energy range 0.68-1.82 GeV with the tagged photon facility at ELSA. The experiment was carried out with a 4pi detection system, a circularly polarized tagged photon beam, and a frozen spin polarized proton target. The contribution to the GDH sum rule in this photon energy range is [49.9+/-2.4(stat)+/-2.2(syst)] microb.     

Carrier testing program in a high-risk cystic fibrosis population from northeastern Italy. Active recruitment of relatives via probands' parents

OBJECTIVE: To assess the practicability and monitor the results of an active carrier testing program among relatives of cystic fibrosis (CF) patients. METHODS: Parents of CF patients in the Veneto and Trentino regions of northeastern Italy were asked to help recruit relatives aged between 18 and 45 years for CF mutation testing. RESULTS: Of 409 enrolled CF parents, 59.6% agreed to send to the CF Center family composition data of relatives up to the third degree, and 28.8% recruited relatives to carrier testing, providing names and addresses of those who, being contacted, expressed a willingness to be tested. The participation of parents was higher if they were young and had a child recently diagnosed with CF. Recruiting parents indicated 333 close relatives (59%) for testing. When contacted by the CF Center, 170 of these 333 (51%) attended for testing. The percentage of close relatives who spontaneously asked for the test was 5.4% before the carrier testing program started; it rose to 25.3% following the introduction of the active strategy. CONCLUSIONS: The participation of the parents of CF patients is an important factor affecting the utilization of testing by relatives. Besides this, the influence of a favorable medical and cultural context (participation of gynecologists and family doctors in testing programs, genetic education of the general population) has to be considered.     

MRI-guided neutron capture therapy by use of a dual gadolinium/boron agent targeted at tumour cells through upregulated low-density lipoprotein transporters

The upregulation of low-density lipoprotein (LDL) transporters in tumour cells has been exploited to deliver a sufficient amount of gadolinium/boron/ligand (Gd/B/L) probes for neutron capture therapy, a binary chemio-radiotherapy for cancer treatment. The Gd/B/L probe consists of a carborane unit (ten B atoms) bearing an aliphatic chain on one side (to bind LDL particles), and a Gd(III)/1,4,7,10-tetraazacyclododecane monoamide complex on the other (for detection by magnetic resonance imaging (MRI)). Up to 190 Gd/B/L probes were loaded per LDL particle. The uptake from tumour cells was initially assessed on cell cultures of human hepatoma (HepG2), murine melanoma (B16), and human glioblastoma (U87). The MRI assessment of the amount of Gd/B/L taken up by tumour cells was validated by inductively coupled plasma-mass-spectrometric measurements of the Gd and B content. Measurements were undertaken in vivo on mice bearing tumours in which B16 tumour cells were inoculated at the base of the neck. From the acquisition of magnetic resonance images, it was established that after 4-6 hours from the administration of the Gd/B/L-LDL particles (0.1 and 1 mmol kg(-1) of Gd and (10)B, respectively) the amount of boron taken up in the tumour region is above the threshold required for successful NCT treatment. After neutron irradiation, tumour growth was followed for 20 days by MRI. The group of treated mice showed markedly lower tumour growth with respect to the control group.

针对坐落于意大利帕维亚大学的TRIGA Mark II反应堆热柱结构进行优化设计,从而满足面向硼中子俘获治疗(BNCT)的单光子发射计算机断层成像(SPECT)研究要求。为提高计算效率并减小统计误差,对比分析使用SSW/SSR方法与直接使用反应堆为源项时热柱内照射位置处中子能谱,其结果基本一致,从而验证了SSW/SSR方法的可靠性。为在该反应堆开展BNCT中SPECT实验,热柱中子束需准直为笔形束。对比分析四种热柱优化方案下束流口处及探测器处热中子和光子通量：40 cm长石墨(射束口5 cm3 cm);0.5 cm厚硼包裹40 cm长石墨(射束口5 cm3 cm);30 cm长天然锂聚乙烯(射束口直径4 cm);30 cm长天然锂聚乙烯(20 cm长射束口直径5 cm,5 cm长射束口直径4 cm,5 cm长射束口直径2 cm)。结果显示,射束口处热中子通量分别为1.05108,2.52107,6.08107和5.10 107 #/(cm2s)。综合考虑中子准直效果及光子污染,方案三具有最优性能。为后续进行BNCT-SPECT理论和实验研究提供了基础,从而有效促进BNCT剂量准确评估方法的研究进程。     

High-performance liquid chromatographic-mass spectrometric assay of busulfan in serum and cerebrospinal fluid.

A liquid chromatographic-mass spectrometric method has been developed to determine busulfan concentrations in the cerebrospinal fluid and serum of some children undergoing bone marrow autotransplantation. After two liquid-liquid extraction steps with dichloromethane on a biological matrix, the separation of busulfan was carried out by isocratic reversed-phase chromatography. The mass spectrometric system was operated in electron-impact mode. Principal ions at m/z 175, 111 and 79 were observed for busulfan, but only m/z 175 was chosen for the quantification of the analyte. The retention time of busulfan was 2.5 min. The detection limit of 100 ng/ml allowed the determination of cerebrospinal fluid and serum busulfan concentrations during the four days of high-dose (1 mg/kg) treatment prior to autotransplantation in five child patients.     

The garner heavy duty quartz fiber microbalance

Summary A Garner balance is described which by means of a vertical suspension fiber is said to be capable of weighing a 5-g load with a sensitivity better than that attainable with the ordinary knife-edge balance. Its weighing chambers were especially designed to accommodate light absorption tubes for the determination of carbon and hydrogen. A practical system for eliminating vibration and furnishing a suitable mounting for sensitive quartz fiber balances in the industrial laboratory is also reported. Under a 5-g load, the Garner balance was found to give strict proportionality within±0.001 mg between torsion and weight. It reproduces within ±0.26g when weighing a 0.4-g platinum boat with the torsion fiber under continuous strain.

Zusammenfassung Eine Garner Mikrowaage mit Quarzbalken wird beschrieben, deren Empfindlichkeit die der üblichen, auf Schneiden ruhenden Feinwaagen übertrifft und die doch eine Last von 5 g tragen kann, da der Balken von einem vertikalen Quarzfaden getragen wird. Die Gehänge und Gehängekammern wurden zur Aufnahme von leichten Absorptionsapparaten speziell entworfen. Eine für das Industrielaboratorium geeignete, praktische Aufstellung der Waage, die störende Vibrationen ausschließt, wird beschrieben. Unter Maximalbelastung von 5 g wurde Proportionalität innerhalb±0,001 mg zwischen Gewicht und kompensierender Torsionskraft festgestellt. Das Gewicht eines Platinschiffchens (0,4 g) konnte mit einem mittleren Fehler von ±0,26g reproduziert werden, wenn der horizontale Torsionsfaden aus Quarz ständig unter Torsionsspannung gehalten wurde.

Résumé On décrit une balance de Garner qui, au moyen d'un fil de suspension vertical, est capable de peser une charge de 5 g avec une sensibilité meilleure que celle que l'on obtient avec une balance à couteau ordinaire. Ses compartiments ont été spécialement agencés pour loger les tubes d'absorption légers pour le dosage du carbone et de l'hydrogène. On donne aussi un système pratique pour éliminer les vibrations et pour fournir un montage convenable pour les balances à fil de quartz sensibles dans les laboratoires industriels. Sous une charge de 5 g, on a trouvé que la balance de Garner donne une proportionnalité rigoureuse à moins de±0,001 mg entre la torsion et le poids. Elle donne une valeur reproductible à moins de±0,26g pour une nacelle de platine de 0,4 g pour le fil de torsion soumis à un effort constant.
