Energy dispersive X-ray fluorescence (EDXRF) determination of lanthanides after preconcentration on 1-(2-pyridylazo)-2-naphthol modified naphthalene

A method is described for the simultaneous multielement determination of yttrium and lanthanides at microgram level. This is based on the preconcentration of these lanthanides on to 1-(2-pyridylazo)-2-naphthol (PAN) modified naphthalene. The optimal conditions for quantitative preconcentration viz., pH, amount of PAN modified naphthalene, time of stirring and aqueous phase volume were systematically evaluated. The quantitation of lanthanides was carried out by energy dispersive X-ray fluorescence analyzer, employing²⁴¹Am annular source, via their characteristic K X-rays. The developed procedure gave reliable results in the analysis of xenotime samples.     

Meroterpenoids—III : Psoralea corylifolia linn.—3. Synthesis of (±)-bakuchiol methyl ether

A synthesis of (±)-bakuchiol methyl ether, involving Claisen rearrangement of geranyl vinyl ether as the key step, is described. Further thermal reorganisation of the intermediate 3-vinyl-citronellal is discussed.     

Mixed-ligand chelate extraction of neodymium(III) with thenoyltrifluoroacetone and various sulphoxides

Mixed-ligand chelate extraction of Nd(III) into benzene with mixtures of thenoyltrifluoroacetone (HTTA) and bis-2-ethylhexyl sulphoxide (B2EHSO) or di-n-octyl sulphoxide (DOSO) or diphenyl sulphoxide (DPhSO) or trioctylphosphine oxide (TOPO) from thiocyanate solutions has been studied. A very high synergistic enhancement of the order 10(3) has been observed in these systems. Thus this mixed-ligand chelate system may be useful in the extraction and separation of Nd(III) from other metal ions. The mechanism of extraction can be explained by a simple chemically based model presented in this paper.     

Flotation-spectrophotometric method for the determination of mercury using iodide and brilliant green

This paper describes a sensitive spectrophotometric method for the determination of mercury, based on the flotation of a mercury iodide-brilliant green complex with cyclohexane at pH 5 and dissolution of the complex in acetone. The absorbance of the complex is measured at 625 nm. Beer's law is obeyed from 4–500 g/l of mercury. The method is highly sensitive ( = 5.96 × 10⁵ 1 mol^–1 cm^–1) and selective, as it is free from interferences by almost all cations and anions in the presence of EDTA. Furthermore, a two-level orthogonal array design as a chemometric method has been used for optimization of the various parameters involved. The method has been successfully applied to paper industry effluents.     

Spectrophotometric determination of ziram (dithiocarbamate fungicide) by thiocyanate and rhodamine 6G method

A sensitive spectrophotometric method has been developed for the determination of ziram in water, vegetables and grains. The method is based on the dissociation of dithiocarbamate complex of zinc with thiocyanate and rhodamine 6G at pH 4 to form a pink coloured complex that is stabilized by gelatin. The method is simple and Beer's law is obeyed over the concentration range of 0.05-1 ppm of ziram. The method is free from interference of similar dithiocarbamate fungicides containing Mn(2+) and Fe(3+) ions.     

In situ measurement of conformational changes in proteins at liquid interfaces by circular dichroism spectroscopy

A new circular dichroism (CD) spectroscopy technique for studying conformational changes in proteins in situ at the air-water interface is described. By using this technique, conformations of four proteins, viz., beta-casein, bovine serum albumin (BSA), lysozyme, and fibrinogen in the adsorbed state at the air-water interface have been studied. beta-Casein, which is predominantly in a disordered state in solution, assumes a beta-sheet conformation at the air-water interface. On the other hand, lysozyme and fibrinogen, which are alpha+beta-type proteins in solution, become beta-type proteins by completely transforming their alpha-helix structure into beta-sheets. Bovine serum albumin, which is an alpha-type protein in solution, loses its alpha-helix and becomes a disordered protein at the air-water interface. The results indicated that during unfolding and film formation at the interface, structural changes in proteins, regardless of their initial native state, follow the course of increasing beta-sheet and disordered structure and decreasing alpha-helix content. Although this seems to be the general trend, the exceptional case of BSA suggests, however, that this is not universal.     

Nonspecific Effect of Double-Stranded (ds) RNA on Prophenoloxidase (proPO) Expression in Penaeus monodo n

RNA interference-mediated silencing is an effective way of controlling white spot syndrome virus (WSSV). However, the effect of RNAi on the innate immune mechanism is not well understood. Prophenoloxidase (proPO) is an important component of the shrimp innate immunity. In the present study, nonspecific effect of two double-stranded (ds)RNA-expressing constructs, one targeting vp28 gene of WSSV (pCMV-VP28-LH) and another targeting green fluorescent protein (GFP) (pCMV-GFP-LH) on proPO2 gene expression, is investigated. mRNA expression levels of proPO2 in hemocytes of DNA construct-injected shrimp were estimated using real-time PCR with elongation factor 1-α as internal control. Empty vector (pcDNA)-injected shrimp were used as experimental control. In pCMV-VP28-LH-injected shrimp, proPO2 showed significant upregulation until 48 h post-injection (p.i.). Similarly, pCMV-GFP-LH-injected animals showed high levels of expression until 72 h p.i. WSSV-challenged animals, compared to pcDNA-injected control group, showed no significant change in expression of the gene until 24 h. However, an increased expression was noticed at 48 h p.i. Our results suggest that neither the plasmids nor the long hairpin RNA expressed by the constructs has any nonspecific silencing effect on the proPO2 expression. On the contrary, the consistent upregulation of proPO2 observed in shrimp injected with dsRNA at early time-points indicates the possibility of nonspecific protection against WSSV infection.     

Regio- and stereoselective synthesis of spiro-pyrrolidine/pyrrolizidine/thiazolidine-grafted macrocycles through intramolecular 1,3-dipolar cycloaddition reaction

Regioselective synthesis of spiropyrrolidine-grafted 11-membered macrocycle was accomplished through an intramolecular [3+2] cycloaddition of azomethine ylides. The key precursor alkenyl diketone (4a–b) was obtained from simple starting materials. The dipole generated from isatin tethered to O-alkyl enone (4a–b) was reacted intramolecularly to yield the spiropyrrolidine-grafted macrocycles (6a–b). The structures of the cycloadducts were assigned by 2D NMR and confirmed by single crystal analysis.     

Synthesis of glucosylspiro-oxindole derivatives via one-pot three-component cycloaddition of azomethine ylides

Highly regio- and stereoselective synthesis of glucosylspiro-oxindole/acenaphthenone derivatives was accomplished through 1,3-dipolar cycloaddition (1,3-DC) reaction. The glucosyl α,β-unsaturated-7,3-lactone (a sugar lactone) derived from d-glucose was used as a chiral dipolarophile in 1,3-DC reaction for the first time. The azomethine ylides generated from sarcosine/l-proline/thiazolidine-4-carboxylic acid/piperidine-2-carboxylic acid with isatin and acenaphthoquinone were trapped by the sugar lactone to give a series of glucosylspiro-pyrrolidine derivatives in good yields.
