The maximum fluxes (Jmax) of beta-blockers through keratinized membranes were determined in vitro and compared with their physiochemical parameters such as lipophilicity (log k'0) and melting point (mp). Rat abdominal skin and hamster cheek pouch mucosa were used as the model membranes. Propranolol, metoprolol, timolol, pindolol, nadolol and agenolol were used as beta-blockers with a variety of physicochemical characters. Linear relations of Jmax with either log k'0 or mp were observed both in intact rat skin and in intact hamster cheek pouch, suggesting that the lipophilicity and thermodynamic activity of a drug in the crystal state primarily affect the drug's permeation through these membranes. However, the slope, dJmax/d(log k'0), for cheek pouch mucosa was greater than that for rat skin, corresponding to the lack of appendigeal shunt pathways in cheek pouch. Penetration studies using the delipidized membranes and the isolated stratum corneum sheet of hamster cheek pouch mucosa clarified that the primary rate-limiting barrier function might exist in the lipid layer of the stratum corneum. Jmax values for the tape-stripped and delipidized skins correlated with both the solubilities of drugs in the vehicle and with the mp, suggesting the polar porous characteristics of both model membranes. However, a theoretical approach confirmed that the contribution of an intracellular or aqueous pore route in the intact membrane to the permeation of drugs with positive lipophilic indexes is negligible.  相似文献

Elucidation of the binding specificity of a concanavalin A-Sepharose column led to the possibility of the affinity chromatography of oligosaccharides and glycopeptides with the use of immobilized lectin columns. Subsequent addition of immobilized erythroagglutinating phytohaemagglutinin, Aleuria aurantia lectin, Datura stramonium agglutinin, Ricinus communis agglutinin and Allomyrina dichotoma agglutinin to the range of well characterized lectin columns has afforded a way to fractionate a mixture of N-linked oligosaccharides even to a single component.  相似文献

The copolymers of N-vinyl-2-pyrrolidone and 2-dimethylaminoethyl methacrylate, 2-diethylaminoethyl methacrylate, 2-dimethylaminoethyl acrylate, or 2-dimethylaminopropyl acrylamide have been prepared. Studies were made of the binding of a “binding probe,” methyl orange, by the copolymers in aqueous solution. The first binding constants accompanying the binding were evaluated. Furthermore, the intensity of fluorescence of a hydrophobic fluorescent probe, 2-p-toluidinylnaphthalene-6-sulfonate, in the presence of these polymers was investigated. The nature and phenomena of dye binding and hydrophobic fluorescent probe binding with the polymers are discussed.  相似文献

The copolymerization of aziridines and cyclic imides was studied. Aziridines copolymerized alternately with cyclic imides to give crystalline polyamides. Ethylenimine and succinimide copolymerized to nylon 2,4, melting near 300°C., without any catalyst. Similarly, the corresponding crystalline polyamides were obtained from the systems of 1,2-propylenimine–succinimide, ethylenimine–glutarimide, and ethylenimine–phthalimide. The copolymerization of aziridines and cyclic imides in the presence of BF3OEt2 gave a copolymer which was rich in aziridine units, whereas, the addition of triethylamine had no influence on the copolymer composition. A mechanism of copolymerization was proposed based on the facts that N-tetramethylenesuccinamide was obtained by the reaction of pyrrolidine and succinimide, N-acetylethylenimine reacted with acetamide to yield N,N′-diacetylethylenediamine and that the rate of this copolymerization was dependent on the electrophilicity of imide.  相似文献

The surface structure of a single-crystal ZnO wafer was studied by angle-resolved x-ray photoelectron spectroscopy (ARXPS) using synchrotron radiation. As a result, well-defined x-ray photoelectron diffraction (XPD) patterns were obtained for the (0001) and (0001¯) polar surfaces using the photoemission from the Zn 2p3/2 and O 1s core levels. The XPD patterns were indexed assuming forward scattering of photoelectrons by neighboring ions. Further, the XPD patterns for the (0001) and (0001¯) surfaces were different from each other, indicating the possibility for using the XPD technique for polarity determination.  相似文献

Olean-11,13(18)-dien-3beta,30-diol dihemiphthalate (3), which was derived from glycyrrhetinic acid (GA), has been reported to produce a potent of anti-inflammatory effect in in vivo assays. Using 3 as a lead compound, we attempted to synthesize some modified compounds which varied in the following; i) the position of a carboxyl group in the phthalate moiety, ii) the number of carboxyls attached to the benzoyl group, iii) conversion of benzene ring to another ring system, iv) the linkage form between the benzene ring and oleanene skeleton at position 3 and/or 30. These were screened for their inhibitory activity against interleukin-1 beta (IL-1 beta)-induced prostaglandin E(2) (PGE(2)) production in normal human dermal fibroblasts (NHDF). Although conversion of the ortho-carboxyl group of 3 into the meta-position or the para-position led to an increase in inhibitory activity, the elimination or increase of the carboxyl group resulted in loss of the inhibitory activity. Conversion of the ester bond to the amide bond at position 3 and/or 30 of 3 did not contribute to a significant increase in inhibitory activity. On the other hand, among the derivatives possessing an anthranilic acid moiety at position 30 of 3beta-O-acetyl-olean-11,13(18)-dien-30-oic acid (20), 3beta-hydroxy-30-nor-olean-11,13(18)-dien-20 beta-[N-(2-carboxyphenyl)]carboxamide (30) showed the most potent inhibitory activity (IC(50) 1.0 microM) in this series.  相似文献

The in vitro synthesis of chitin via a nonbiosynthetic path has been achieved for the first time by enzymatic ring-opening polyaddition of a chitobiose oxazoline monomer ( C2O ). Chitinase, a hydrolysis enzyme of chitin, recognized and polymerized the monomer regio- and stereoselectively. The structure of artificial chitin was confirmed by comparison with an authentic natural chitin sample with the use of CP/MAS 13C NMR and IR spectroscopy. X-ray diffraction as well as NMR analysis showed its crystal structure of α-chitin. Characteristic features of the present polymerization are described.  相似文献

A liquid chromatography/atmospheric pressure chemical ionization mass spectrometry (LC/APCI-MS) method based on time-of-flight MS (TOFMS) with a real-time reference mass correction technique was developed for the simultaneous determination of Fusarium mycotoxins (nivalenol, deoxynivalenol, fusarenon X, 3-acetyldeoxynivalenol, 15-acetyldeoxynivalenol, HT-2 toxin, T-2 toxin, diacetoxyscirpenol, zearalenone) and Aspergillus mycotoxins (aflatoxin B1, aflatoxin B2, aflatoxin G1, aflatoxin G2) in corn, wheat, cornflakes and biscuits. Samples were cleaned up with a MultiSep #226 column. Detection of the mycotoxins was carried out in exact mass chromatograms with a mass window of 0.03 Th. Calibration curves were linear from 2 to 200 ng x mL(-1) for trichothecenes and zearalenone, and 0.2 to 20 ng x mL(-1) for aflatoxins, by 20 microL injection. The limits of detection ranged from 0.1 to 6.1 ng x g(-1) in foodstuffs analyzed in this study. The LC/TOFMS method was found to be suitable for the screening of multiple mycotoxins in foodstuffs rapidly and with high sensitivity, and its performance was demonstrated for the confirmation for target mycotoxins.  相似文献

In 1707, approximately 0.7 km3 of magma erupted from new vents on the southeastern slope of Fuji volcano. The air-fall clasts of this explosive eruption are composed of vesicular dacite pumice, andesitic dense scoria, basaltic dense scoria, and vesiculated basalt scoria, stratified from bottom to the top of the fallout unit. Some compositional gaps are found in the variation from basalt to dacite indicating that three independent magmas, basalt, andesite and dacite, existed just prior to the eruption. Andesite and dacite magmas are mixed just prior to or during the eruption showing a linear two component mixing in the major and trace element concentration. Basalt in the later stage of the eruption shows no compositional affinity with the above two magmas. Basalt magma might have acted as a heat source to remobilize the andesite and dacite magmas, and the explosive eruption of basalt could have been caused by the abrupt pressure release due to precursory mixing and eruption of dacite and andesite magmas.  相似文献

A spatial embedding of a graph G is an embedding of G into the 3-dimensional Euclidean space . J.H. Conway and C.McA. Gordon proved that every spatial embedding of the complete graph on 7 vertices contains a nontrivial knot. A linear spatial embedding of a graph is an embedding which maps each edge to a single straight line segment. In this paper, we construct a linear spatial embedding of the complete graph on 2n−1 (or 2n) vertices which contains the torus knot T(2n−5,2) (n4). A circular spatial embedding of a graph is an embedding which maps each edge to a round arc. We define the circular number of a knot as the minimal number of round arcs in among such embeddings of the knot. We show that a knot has circular number 3 if and only if the knot is a trefoil knot, and the figure-eight knot has circular number 4.  相似文献
