A study has been carried out on 10 patients with ovarian cancer treated by intraperitoneal mitoxantrone. The serum concentration was determined by high performance liquid chromatography with spectrophotometric detection. The extracted analyte on the cartridge was injected and eluted on-line into the analytical column by the mobile phase. This improved the signal and sample processing rate, without significant loss in analytical performance. Excellent linearity (r greater than 0.9994) was observed for the calibration curve over the range 1-2000 ng/mL, along with a precision within-day and between-day estimated as 1.5% and 5.6%, respectively. Sensitivity was an order of magnitude higher than that of the comparison method. From a clinical point of view, these preliminary results have shown that intraperitoneal administration is more beneficial than intravenous therapy. Low serum levels (1-30 ng/mL) with a maximum at the first or second hour are revealed.  相似文献

Scleroglucan is a natural polysaccharide that has been proposed for various applications. However there is no investigation on its property variations when the molecular weight of this polymer is reduced. Scleroglucan was sonicated at two different polymer concentrations for different periods of time and the effect of sonication was investigated with respect to molecular weight variations and rheological properties. Molar mass, estimated by viscometric measurements, was drastically reduced already after a sonication for a few min. Sonicated samples were used for the preparation of gels in the presence of borate ions. The effect of borax on the new samples was investigated by recording the mechanical spectra and the flow curves. A comparison with the system prepared with the dialysed polymer was also carried out. The anisotropic elongation, observed with tablets of scleroglucan and borax, was remarkably reduced when the sonicated samples were used for the preparation of the gels.  相似文献

Three polysaccharides, Rhizobium leguminosarum 8002 EPS(I), Rhizobium trifolii TA1-EPS (II), Rhizobium leguminosarum 127K87 EPS (III), produced by bacteria of Rhizobium genus have been investigated by static and dynamic light scattering combined with chirooptical measurements. All three polymers have the same backbone but differ in the length of the side chains and in the content of minor substituents. An isothermal conformational transition coil → helix was observed with I and II by adding salt (NaCl). The molecular parameters of the polysaccharides in the ordered state were determined by light-scattering data. Increasing the ionic strength a shrinking of the helix was observed accompanied by a corresponding decrease in the radius of gyration. An extraordinary chain stiffness in terms of Kuhn segment lengths was found in both cases, similar to that already observed for other microbial polysaccharides. In the case of III no disorder → order transition was induced by the salt, and the scattering behavior corresponds to that of a rather flexible polymer with a characteristic ratio C = 24. The incapability of III to form a helical structure is attributed to the effect of the very long side chain. The analysis of the time correlation functions revealed typical flexible chain behavior for all three polysaccharides. This behavior for the two ordered polymers is in agreement with a recent theory by Maggs and is due to bending modes of the rods. ©1995 John Wiley & Sons, Inc.  相似文献

Mixed crystals of composition (n-CmH2m+1NH3)2xm(n-CnH2n+1 · NH3)2?2xm ZnCl4 (m= 12; n=14, 16, 18) are formed upon annealing mechanical mixtures of layer compounds (n-CnH2n+1NH3)2ZnCl4, through solid state diffusion of the n-alkylammonium cations. From a common high temperature monophasic modification, in which the polymethylenic chains are conformationally molten, different modifications are obtained on cooling [depending on the (n ? m) value] in which chain crystallinity is developed. The structure of such modifications is speculated on the grounds of DSC and preliminary X-ray diffractometric and IR data, and compared with that of similar previously characterized systems.  相似文献

Scleroglucan is a natural polysaccharide, produced by fungi of the genus Sclerotium, that has been extensively studied for various commercial applications (secondary oil recovery, ceramic glazes, food, paints, etc.) and also shows several interesting pharmacological properties. This review focuses its attention on the use of scleroglucan, and some derivatives, in the field of pharmaceutics and in particular for the formulation of modified-release dosage forms. The reported investigations refer mainly to the following topics: natural scleroglucan suitable for the preparation of sustained release tablets and ocular formulations; oxidized and crosslinked scleroglucan used as a matrix for dosage forms sensitive to environmental conditions; co-crosslinked scleroglucan/gellan whose delivery rate can be affected by calcium ions. Furthermore, a novel hydrogel obtained with this polysaccharide and borate ions is described, and the particular structure of this hydrogel network has been interpreted in terms of conformational analysis and molecular dynamics. Profound attention is devoted to the mechanisms involved in drug release from the tested dosage forms that depend, according to the specific preparation, on swelling and/or diffusion. Experimental data are also discussed on the basis of a mathematical approach that allows a better understanding of the behavior of the tested polymeric materials.  相似文献

The bark of Ficus hispida, one component of a mixture used as a cancer-cure in Thailand, yielded a new and unusual compound when extracted with light petroleum. The structure of the new compound, 10-ketotetracosyl arachidate, was proven by degradation and synthesis. The lupeol acetate and β-sitosterol found in the light petroleum extract were identified by comparison with authentic samples.  相似文献
